Abstract

The acquisition of complete DNA is usually the first and most critical step in many basic molecular biology applications. Isolation of complete total DNA from tissue samples is related to different physical and biochemical properties of tissue. Long fragments of DNA are easily degraded and fragmented, which poses a challenge for extracting complete long fragments of DNA. By comparing several different extraction methods and verifying the concentration and length of extracted DNA, it is proved that the kit we used has high efficacy. We provide tissue kit DNA extraction for long fragments and obtain long fragments of high purity DNA from almost any type of tissue, especially muscle and blood tissues.

Abstract

The aims of this study is to apply the Auto Sales Forecasting software to predict sales transaction data. The Auto Sales Forecasting software consists of two main features namely descriptive analysis and forcasting features along with its visualization. Forecasting methods contained in the Auto Sales Forecasting application are forecasting methods of Simple Moving Average, Robust Exponantial Smoothing, Auto ARIMA, Artificial Neural Network, Holt-Winters, and Hybrid Forecast. The Auto Sales Forecasting software can intelligently choose the best forecasting method based on RMSE values. The results showed that the Auto Sales Forecasting software successfully analyzed the sales transaction data. From the analysis it was found that there were 43 types of products produced and sold by the Futry Bakery & Cake Store. Three of them are the types of products that are most in demand by consumers, namely Sweet Bread, Maros Bread, and Traditional Cakes 3500. The best selling product type, Sweet Bread, is used to build forecasting models. The best forecasting method is the Robust Exponential Smoothing method with the smallest RMSE value of 0.83 on the variable number of sold out products. Forecasting results using the Robust Exponantial Smoothing method show that the average number of products to sell for the next seven days ranges from 116 products with a certain confidence interval value.

Abstract

Building construction consists of two parts, namely the upper building and lower building. The upper building transmits forces to the lower building, which then from the lower building is channeled to the supporting soil. Pile foundation is a part of the lower structure that is used to receive and distribute the load from the upper structure to the supporting soil which is located at a certain depth determined by the results of the soil investigation. Piles are used as building foundations if the soil under the building base does not have enough bearing capacity to carry the weight of the building working on it. The stake used in this comparison is the Concrete Spun Pile and Bored Pile foundation. The results of the study show that the Bored pile method is slightly more complicated than the Concrete spun pile method. In terms of time required for the Concrete spun work is 6 hours, while for Bored piles per point is 9 hours assuming the work is 3 months In terms of the cost of foundation work of 1000mm diameter and 30m depth using the Concrete spun pile method is Rp. 13,704,241,634.10 for 156 points. whereas for the Bored pile method with a diameter of 1200mm and a depth of 30m is Rp. 14,242,797,932.85 for 78 points. So the foundation work with the Concrete spun pile method is more effective and efficient than the Bored pile method based on the method of implementation, time and cost.

Abstract

In the world of construction there are various methods and types of materials used to support the passage of a construction work. One of them is composite girder plate. Composite girder plate is one of the many construction methods that combine two construction materials that are physically different in nature, namely concrete with steel. This type of composite girder plate construction is commonly used for bridge construction work with a fairly large span and width. In its use, of course, it must be preceded by stages of careful planning on a standard and valid basis as well. In the following research will discuss and look for similarities and differences regarding the two types of rules in the planning of composite girder plates, namely the rules of planning composite girder plates using AASHTO LRFD BRIDGE DESIGN SPECIFICATIONS 2017 with SNI 1729: 2015. After doing the initial stages of modeling using CSI Bridge software using the profile cross section constraints of the AASHTO provisions, the internal force obtained is Moment Force (Mu) of 3469.13 kNm and Shear Force (Vu) of 225.98 kNm. Then proceed with the analysis of calculations with the help of Microsoft Excel software namely calculating using the AASHTO LRFD BRIDGE DESIGN SPECIFICATIONS 2017 regulations for stability requirements of strong boundary conditions on the bending requirements. Then a Nominal Moment (ØMn) value of 6420.19 kNm is obtained. Then proceed to calculate the same planning constraints, but this time using SNI 1729: 2015 regulations. Obtained Nominal Moment Value (ØMn) of 6579.88 kNm. Then it can be concluded that the two regulations produce a safe and strong planning, of course in accordance with applicable regulations namely: Moment (Mu <ØMn).

Abstract

The earthquake that often hit Indonesia caused thousands of lives and caused damage to buildings. These earthquakes often occur because Indonesia is in two regions, namely the Pacific earthquake path (Circum Pacific Earthquake Belt) and the Asian earthquake lane (Trans Asiatic Earthquake Belt). Earthquake disasters cause damage to building structures. When an earthquake occurs, it is expected that the building can accept earthquake force at a certain level without significant damage to its structure. In general, earthquake analysis is divided into two major parts, namely static earthquake analysis and dynamic earthquake analysis. In buildings that are very high, irregular, multilevel, and buildings that require enormous accuracy are used dynamic analysis planning, which consists of a variety of spectral response analysis and dynamic time response dynamic analysis. This study aims to determine the building's security in terms of displacement, drift, and base shear. The method used is a dynamic analysis of the response spectrum using the ETABS program. The maximum total drift in the X direction is 0.0200475 m and in the Y direction is 0.020405 m, so the building is safe against ultimate boundary performance (0.02h) and service boundary performance {(0.03 / R) x h}. So that the displacement in the building does not exceed the maximum displacement, the building is safe from earthquake plans.

Abstract

Construction of the Kedunglarangan river flood control system designed to prevent flooding every rainy season in the Bangil sub-district. Kedunglarangan River is a river that flows in two regencies Sidoarjo and Pasuruan which has an area of 282.67 km2 watershed with a river length of 23.7 km. Kedunglarangan river has 4 (four) watershed sub-systems. The scope of this flood prevention work-study is the normalization of the Kedunglarangan River starting from the meeting with the Wrati River downstream up to 7 km. Normalization work is carried out with excavation and river widening to meet flood discharge in accordance with the conditions of the study area. If the river excavation work is done in accordance with the design master will form a basin that causes the creation of a dike. In this condition, it will be a temporary water reservoir where the water velocity is very low. So the work carried out the impact is only temporary. From the results of analysts, it is more efficient to do river widening and embankment raising rather than increasing river depth. River excavation work like that is very risky to create very fast sedimentation. Normalization method with river widening is one way to maintain the river flow downstream and flood water levels
