Abstract

Keladi tikus (Typhonium flagelliforme (Lodd) BL) is used traditionally as anti cancer and anti virus. Dewandaru (Eugenia uniflora Linn) have known to have antibacterial activity. This experiment was aim to examine whether the extract of Eugenia uniflora Linn and Typhonium flagelliforme (lodd) BL leaves contain protein which had activity to cut double helix supercoiled DNA. The experiment was conduct by incubating DNA plasmid (pUC18) with protein isolated from Eugenia uniflora Linn and Typhonium flagelliforme (lodd) BL leaves extract at room temperature for 1 hour. Crude extracts were made by extraction in 0.14 M NaCl-5 mM sodium phosphate buffer pH 7,2. Extract were considered to have RIPs activity when it was able to cut double helix supercoiled DNA to linear nick circular form. Electrophoretogram was observed under ultraviolet light. Result showed that Typhonium flagelliforme (Lodd) BL) leaves extract contain protein which had activity to cut double helix supercoiled DNA to linear nick circular form. While Eugenia uniflora Linn leaves extract gave negative result.   ABSTRAK Tanaman keladi tikus (Typhonium flagelliforme (Lodd) BL) digunakan sebagai antikanker dan antivirus. Tanaman dewandaru (Eugenia uniflora Linn) telah diketahui memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui apakah ekstak gubal daun dewandaru (Eugenia uniflora Linn) dan daun keladi tikus (Typhonium flagelliforme (lodd) BL) mengandung protein yang mampu memotong DNA superkoil untai ganda. Penelitian ini dilakukan dengan cara menginkubasikan DNA plasmid (pUC18) dengan sejumlah protein dari ekstrak gubal daun Eugenia uniflora Linn dan daun Typhonium flagelliforme (Lodd) BL) pada suhu kamar selama 1 jam. Ekstrak gubal daun dibuat dengan ektraksi daun dalam buffer natrium fosfat 5Mm yang mengandung NaCl 0,14 M Ph 7,2. Ekstrak akan memiliki aktivitas RIPs jika mampu memotong DNA superkoil untai ganda menjadi bentuk nick circular dan linier. Hasil elektroforesis dilihat di bawah sinar UV. Hasil penelitian menunjukkan bahwa ekstrak gubal daun keladi tikus (Typhonium flagelliforme (Lodd) BL) mengandung protein yang mampu memotong DNA superkoil untai ganda menjadi bentuk nick circular dan linier seperti pada RIPs. Sedangkan ekstrak gubal daun dewandaru (Eugenia uniflora Linn) tidak menunjukkan kemampuan memotong DNA superkoil untai ganda menjadi bentuk nick circular dan linier.
