Abstract

Estrogen-α (ER-α) is the main target for ER + therapy. Inhibition of ER-α is known to slow the proliferation of ER + breast cancer cells. Quinoline derivatives are known to have anticancer activity by inhibiting several types of receptors. It is not known whether quinoline can inhibit ER-α. This study aims to determine the interaction between ER-α with quinoline derivative compounds. Molecular docking of ER-α showed that quinine gave the most negative bond-free energy and the smallest inhibition constant, respectively -8.73 kcal/mol and 0.398 μM. These results provide predictions that quinine has activity as an ER-α inhibitor and has the potential to be developed in the treatment of ER + breast cancer. However, the affinity shown by quinine was lower than that of 4-hydroxytamoxifen, a potent inhibitor of ER-α.

Abstract

This study aims to determine the antioxidant activity of akar kuning (Arcangelisia flava) stem extract from Central Kalimantan using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. This research includes extraction of simplicia and test of antioxidant activity by DPPH method. Based on the analysis of antioxidant activity on ethanol extract from yellow root plant stem IC50 values of 136.81 ppm were obtained. This IC50 value indicates the antioxidant strength of the yellow root plant stems including in the moderate category. Further research on antioxidant activity in the fraction of akar kuning stems needs to be done to determine the composition of the antioxidant compounds in each solvent.

Abstract

Observation of molecular docking results was generally performed by analyzing the docking score and the interacting amino acid residues separately either in tables or graphs. Sometimes it was not easy to rank the tested ligands’ docking results, especially if there were many ligands. This study aims to introduce a new way to analyze docking results with a two‐dimensional graph between the difference in docking score and the similarity of ligand‐receptor interactions. Molecular docking was performed with one reference ligand and several test ligands. The docking score difference was obtained between the test and the reference ligands as the graph’s x‐axis. Meanwhile, the y‐axis contains the similarity of ligand‐receptor interactions, obtained from the ratio of amino acid residues and the types of interactions between the test and reference ligands. Docking result analysis was more straightforward because two critical parameters were presented in one graph. This graph could be used to support the analysis of the docking results.

Abstract

Background:Acetaminophen (PCT) is known for its pro-oxidant properties, which neutralized by the presence of internal antioxidants such as glutathione (GSH). GSH has two forms: monomers and dimers, mainly distinguished by the presence of thiol group. Purpose:This study aims to see the difference in interaction between PCT with both forms of GSH. Method:Molecular docking was performed using Autodock Vina 1.1.2 on whole GSH surfaces. The main parameter used was the free energy of binding as affinity marker, as well as the position of PCT toward GSH. Result:The docking results show that PCT has a slightly higher affinity to the dimeric form of GSH than its monomeric form with the free energy of binding -2.7 kcal/mol and -2.2 kcal/mol, respectively. The interesting thing is the acetyl group of PCT is in a position far from the thiol group in the monomeric form of GSH, in contrast to its dimeric form. Conclusion:These results show that difference in affinity of PCT to both GSH forms is influenced by the position of the acetyl group against the thiol group of cysteine in GSH. The proximity of the pro-oxidant group to the thiol group leads to an increase in the affinity of the pro-oxidant to GSH.

Abstract

ABSTRAK Akar kuning (Arcangelisia flava Merr.) merupakan salah satu tanaman yang tumbuh di Kalimantan Tengah dan secara turun temurun digunakan sebagai pilihan pengobatan beberapa penyakit kulit dan terbukti mempunyai aktivitas antibakteri. Pembuatan sediaan topikal antibakteri berbahan fraksi aktif A. flava sangat potensial dilakukan untuk mengoptimalkan penggunaan dalam pengobatan sekaligus meningkatkan nilai ekonomis dari A. flava. Tujuan penelitian ini untuk mendapatkan formula gel fraksi aktif A. flava sebagai antibakteri yang paling baik secara fisik.Formula gel dibuat dengan variasi jenis dan konsentrasi gelling agent.Gel fraksi aktif A. flava diuji secara fisika, kimia, dan biologi seperti organoleptis, homogenitas, daya sebar, pH, daya lekat, dan daya hambat terhadap Staphylococcus aureus. Hasil formulasi sediaan gel fraksi aktif A. flava dengan basis NaCMC memenuhi syarat sifat fisik dan zona hambatan 11,6 mm pada Formula 1 dan 13,4 mm pada Formula 2, sedangkan sediaan gel dengan basis Carbophol 943 pada Formula 1 dan 2 memenuhi syarat sifat fisik dan zona hambat 10,9 mm pada Formula 2.Formula gel yang memenuhi kriteria secara fisik adalah Formula 2 dengan Carbophol 1,5% sebagai gelling agent. Kata Kunci: Akar kuning, antibakteri, gel, Staphylococcus aureus  ABSTRACT Akar kuning (Arcangelisia flava Merr.) is a plant that grows in Central Kalimantan and has been used as a treatment option for several skin diseases and has been shown to have antibacterial activity. The making of topical antibacterial preparations made from the active fraction of A. flava is very potential to be done to optimize the use in treatment while increasing the economic value of A. flava. The purpose of this study was to obtain the active A. flava fraction gel formula as the best natural antibacterial. The gel formula was made with variations in the type and concentration of the gelling agent. Arcangelisia flava Merr. active fraction gel was tested physical, chemical, and biological, such as organoleptic, homogeneity, spreadability, pH, adhesion, and bacterial inhibitionagainst Staphylococcus aureus. The results of the formulation of the active gel of A. flava based on NaCMC base fulfills physical condition requirements and shows an inhibition zone of 11.6 mmin Formula 1 and 13.4 mm in Formula 2, while gel preparation based on Carbophol 943 Formula 1 and 2 fulfill the physical characteristic requirements and show the inhibition zone of 10.9 mm in Formula 2.Gel formula that meets the physical criteria is Formula 2, with 1.5% Carbophol as a gelling agent. Keywords: Akar kuning, antibacterial, gel, Staphylococcus aureus

Abstract

ABSTRAK             Rambusa (Passiflora foetida L) adalah tumbuhan obat yang banyak terdapat di berbagai daerah di Indonesia, termasuk di Kalimantan Tengah. Beberapa bagian tumbuhan dari rambusa diketahui memiliki aktivitas antioksidan termasuk di bagian daunnya. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dari daun rambusa yang berasal dari Kalimantan Tengah. Metode yang digunakan adalah metode 2,2-diphenyl-1-picrylhydrazyl (DPPH) yang diukur serapannya dengan Spektrofotometer UV Vis. Hasil yang diperoleh menunjukkan nilai IC50 dari ekstrak etanol daun rambusa senilai 93,269 μg/mL. Meskipun nilai IC50 yang diperoleh lebih rendah dibandingkan senyawa antioksidan standar seperti kuersetin, nilai IC50 dari ekstrak etanol daun rambusa asal Kalimantan Tengah masih lebih tinggi dibandingkan daun rambusa yang diperoleh dari daerah lain Kata Kunci—antioksidan, DPPH, Kalimantan Tengah, Passiflora foetida, rambusa  ABSTRACT Rambusa (Passiflora foetida L) is a medicinal plant that is widely found in various regions in Indonesia, including in Central Kalimantan. Some parts of the plant of rambusa are known to have antioxidant activities including in the leaves. This study aims to determine the antioxidant activity of rambusa leaves from Central Kalimantan. The method used was 2,2-diphenyl-1-picrylhydrazyl (DPPH) method which was measured by UV Vis spectrophotometer. The results obtained showed IC50 values of ethanol extract of rambusa leaves worth 93.269 μg/mL. Although the IC50 values obtained were lower than the standard antioxidant compounds such as quercetin, the IC50 value of the ethanol extract of the leaves of rambusa from Central Kalimantan was still higher than that of the rambusa leaves obtained from other regions or some other medicinal plant extracts. Keywords— antioxidants, DPPH, Central Kalimantan, Passiflora foetida, rambusa

Abstract

Proto-oncogene tyrosine-protein kinase Src is also known as simply Src is a tyrosine kinase protein which is one of the targets in various cancer therapies such as leukemia. Meanwhile, akar kuning (Arcangelisia flava) has gained significant attention as a medicinal plant that has a cytotoxic effect on various types of cancer cells. This study aims to determine the potential of secondary metabolites of akar kuning as Src inhibitors. Molecular docking was carried out using Autodock Vina 1.1.2 with 2HCK receptors, that quercetin and dasatinib were used as reference ligands. The docking results showed that the highest affinity was shown by berberine with a ΔG value of -9.0 kcal/mol, exceeded quercetin and dasatinib. However, the highest amino acid similarity to quercetin and dasatinib was produced by jatrorrhizine, with 93.33% and 73.91%, respectively. Interestingly, berberine is the ligand with the third-highest similarity after jatrorrhizine and palmatine, while jatrorrhizine has the second-highest affinity after berberine. The results concluded that the combination of berberine and jatrorrhizine is predicted to be optimally used as an Src inhibitor in cancer therapy.

Abstract

Previous research has shown that berberine, an alkaloid found in several plants such as akar kuning (Arcangelisia flava) shows the potential for inhibition of Human epidermal receptor-2 (HER2). Pharmacophore modification on the berberine was predicted could increase the affinity of berberine derivatives against HER2. The present study aims to determine the main pharmacophore of berberine with the highest influence towards berberine affinity against HER2. Molecular docking was performed on several modified pharmacophore of berberine against HER2. The docking results show that O atom at position number 23 has the most important influence on the berberine affinity towards HER2, where modification of O atom resulting in a decrease of berberine affinity. The highest affinity showed by derivate berberine-6 with the free energy of binding score and dissociation constant -10.80 kcal/mol and 12.17 nM, respectively. In contrast with other berberine derivatives, the berberine-6 derivate does not interact with the amino acid threonine at position 862. It provides a prediction that interaction of that amino acid potentially decrease the berberine activity towards active site of HER2. Further, derivate berberine-6 could be developed into HER2 inhibitor and should be potential to be developed as HER2-positive breast cancer therapy.

Abstract

Food safety is a complex issue that involves stakeholders in its handling. The point of food safety is growing widely and is becoming a concern in the community because it is related to the quality of consumers' good health and well-being. The community service activities based on quality education focused on Good Processed Food Manufacturing Practices (CPPOB), safe use of Food Additives (BTP), and consideration of packaging selection that ensures food safety. Education is carried out to increase the understanding of SMEs actors regarding regulations of CPPOB, the use of BTP, and provisions on Food Packaging. The education process is carried out online using the Zoom Meeting platform. Learning activity is carried out in one direction through lectures and questions and answers sessions. The increase in participants' understanding was measured by comparing the distribution of average scores, pretest, and post-test statistical tests. The effectiveness and quality of the educational process were measured using an evaluation questionnaire with predetermined indicators. The education carried out related to CPPOB, the use of BTP, and the selection of food packaging performed by the community service team of the Department of Pharmaceutical Sciences, Faculty of Pharmacy, Universitas Airlangga to SMEs in Balikpapan has proven to increase the understanding of business actors. It can fulfill their needs and target activity evaluation indicators that have been determined.

Abstract

This study discusses the development of small and medium micro enterprises (MSMEs) in Central Kalimantan with rattan woven craft industries being used as samples to represent other MSME sectors. The purpose of this study is to describe the improvement in the performance and strategy of the MSME group covering various aspects such as job training, capital support, equipment/technology assistance, and access to information and communication. The findings of this study indicate that the development of MSMEs carried out by regional governments in Central Kalimantan has gone in the right direction, with several factors still making improvements possible. Support in the form of training should be conducted regularly and scheduled with the topic of continuous training and productive participants. Aid in the form of equipment requires periodic evaluations, such as a reward-punishment system for business groups that have received various support to ensure the support provided is on target. Another strategy is to form the centre of the MSME industry in the Capital of the Province, to facilitate the access of tourists who just want to visit looking for typical Central Kalimantan souvenirs. The easiest way to do this now is to implement a movement of love for standard Central Kalimantan handicraft products to be displayed in various offices, offices, and government agencies as well as different infrastructure and public facilities.
