Abstract

In addition to decorate the yard, plant namnam (Cynometra cauliflora) turns out to have merit as an herbal remedy because it has biological activities such as antimicrobial, antioxidant, cytotoxic, antiviral, anti-inflammatory, antitumor and antidiabetic. It is study will add to scientific information about plants Namnam the potential to be developed as a functional food and natural herbal remedies in particular as antidiabetic agents and antidiarrheal. Ability as an antidiabetic agent tested by the ability of plants Namnam inhibit α-amylase. Meanwhile, antidiarrheal activity will be tested against the isolated rabbit intestine recorded on kymograph. The analysis showed that the leaf extract of plant Namnam (C. cauliflora) at a concentration of 100 mg/mL, 250 mg/mL and 300 mg/mL each is able to inhibit α-amylase activity by 20.68%; 70.24% and 72.59% and IC50 of 200.67 ± 0.53 mg/mL. The study also showed that the sample tested has the ability to decrease the activity of isolated rabbit intestine in the pattern described in kymograph. Thus the plant leaves Namnam (C. cauliflora) potential to be developed as antidibetes agent and antidiarrheal.  This may partially explain the use in traditional medicine for the treatment of diarrhea. Abstract

 This experiment was designed to study the effect of pegagan (Centella asiatica) and turmeric (Curcuma longa) extracts on the activities of glutathione peroxidase (GSH- Px) in liver of white Spraque Dawley rat aged 2 months old with average weight of 200 grams. Pegagan was extracted using infuse method, and turmeric was extracted using dekokta method. The activity of GSH-Px was measured using Flohe and Gunzler’s methods. Extract pegagan and turmeric were assumed to have both preventive and curative treatments. For the preventive treat- ment, extract pegagan and turmeric at concentrations of 22.05 mg/ml : 184.1 mg/ml could increased the activity of GSH-PX enzyme, i.e. 232.60 ± 21.40 mU / mgprotein. This was approximately 79% than the levels of GSH- Px enzyme in normal liver without paracetamol which was 190.78 ± 9.28 mU/mgprotein. For the curative treatment, extract pegagan and tumeric at concentration of 22.05 mg/ml : 184.1 mg/ml could improved GSH- Px enzyme activity (i.e. 239.01 ± 47.40 mU/ mgprotein). This figure was about 92% higher than the levels of GSH-Px enzyme in the liver normal without paracetamol which was 190.78 ± 9.28 mU/mg protein. Pegagan and turmeric extracts with a high concentration could increased the activity of GSH-Px enzyme, thus potential as curative treatment.

Abstract

Kurkumin yang terdapat dalam rimpang temulawak, selain dapat menginduksi terjadinya proliferasi sel progenitor pada otak tikus dewasa juga dapat menghambat kerja enzim tirosinkinaseyang berperan penting dalam mengatur pertumbuhan dan diferensiasi sel. Meskipun demikianrespon sel saraf terhadap ekstrak temulawak pada masa pertumbuhan perlu kajian lebih lanjut. Efekekstrak temulawak terhadap proliferasi dan diferensiasi sel otak besar atau serebrum pada masapertumbuhan anak diteliti pada sel otak anak tikus Spague Dawley berumur tiga hari yangditumbuhkan dalam media DMEM (Dulbecco’s Modified Eagle’s Medium). Perlakuan dikelompokkan dalam kelompok kontrol positif (mDMEM+30 µg/mL asiaticoside (AC), kontrolnegatif (mDMEM), kelompok yang memperoleh ekstrak temulawak (CZ) 100 ppm (mDMEM+100ppm CZ), CZ 200 ppm (mDMEM+200 ppm CZ), dan CZ 400 ppm (mDMEM+400 ppm CZ). Kultur diinkubasi pada suhu 37 o C dalam inkubator CO 5 % selama 6 hari. Parameter yang diamatiadalah population doubling time, komposisi sel saraf dan sel glia, panjang akson dan dendrite yangdiukur masing masing menggunakan hemositometer, pewarnaan Hematoxyilin Eosin (HE) danmikrometer. Hasil penelitian menunjukkan ekstrak temulawak pada konsentrasi 100 ppmmemperlambat prolfperasi, pada konsentrasi 400 ppm meningkatkan diferensiasi sel yangditunjukkan dengan meningkatnya ratio sel glia terhadap sel saraf dan mempengaruhi panjangakson dan dendrite.Kata kunci : Curcuma xanthorrhiza Roxb., neuron, sel glia, dendrite

Abstract

ABSTRAKSalah satu penyebab peningkatan kadar gula darah pada penderita penyakit diabetes mellitus adalah tingginya aktivitas pemecahan karbohidrat menjadi glukosa oleh enzim α-glukosidase. Beberapa penelitian menunjukan bahwa daun asam kandis diketahui memiliki kandungan senyawa xanthone yang mampu menghambat aktivitas enzim α-glukosidase.     Penelitian ini bertujuan untuk mengetahui aktivitas antidiabetes ekstrak heksan, etil asetat dan etanol daun asam kandis (Garcinia parvifolia (Miq.) Miq.) dengan mengukur daya hambat ekstrak daun asam kandis terhadap aktivitas enzim α-glukosidase secara in vitro. Pengujian daya hambat terhadap aktivitas α-glukosidase dihitung berdasarkan nilai IC50 dengan  senyawa acarbose sebagai pembanding (kontrol positif). Hasil uji daya hambat ekstrak daun asam kandis terhadap aktivitas enzim α-glukosidase menunjukan bahwa aktivitas inhibisi (IC50) dicapai pada pemberian  ekstrak heksan, etil asetat dan etanol daun asam kandis berturut-turut sebesar 226,4 ± 24,18 ppm, 46,96 ± 10,48 ppm, dan  7,32 ± 2,14 ppm. Dari hasil penelitian  dapat disimpulkan bahwa ekstrak etanol berpotensi paling tinggi sebagai inhibitor enzim α-Glukosidase.  Kata Kunci:   Antidiabetes, asam kandis, inhibitor α-glukosidase

Abstract

ABSTRAK Hepatoproktektor adalah senyawa yang dapat mencegah dan memperbaiki sel-sel hepar yang rusak. Parameter yang diamati adalah perubahan kadar SGPT pada hari ke 0,2, 5, 9 dan 13. Hasil penelitian dari 9 kombinasi dosis selama 13 hari menunjukkan semua kombinasi menurunkan kadar SGPT dengan aktivitas hepatoprotektor terbaik adalah terdapat pada tumbuhan ektrak air pegagan: ekstrak etanol kunyit dengan perbandingan 2:3.Kata kunci: Pegagan (Centella asiatica L.Urban), Kunyit (Curcuma longa Linn) dan hepatoprotektor ABSTRACT Hepatoprotector is a compound that can prevent and repair liver cell damage. This research aimed to study the activity and effectivity of combination Centella asiatica water extract and Curcuma longa ethanol extract as hepatoprotector by SGPT (Serum Glutamat Piruvat Transaminase) in Sprague Dawley male white rats induced paracetamol. Parameters measured were changes in ALT levels at day 0, 2, 5, 9 and 13. Combinations of doses for 13 days showed all combinations of those extract potencial hepatoprotector. Abstract

Hepatoprotektor merupakan senyawa yang dapat mencegah dan memperbaiki selhati yang rusak akibat metabolism senyawa toksik. Penelitian ini bertujuan untuk mengetahui aktivitas hepatoprotektor dan dosis efektif dari ekstrak air herba pegagan daun kecil (Centella asiatica L. Urb) terhadap tikus putih jantan Sprague Dawley yang diinduksi dengan parasetamol. Tikus putih jantan galur Sprague Dawley L. yang digunakan berumur 3-4 bulan dengan bobot 180 g – 220 g. Tikus diinduksi parasetamol dengan dosis 180 mg/200 g BB untuk menaikan kadar SGPT (Serum Glutamat Piruvat Transaminase). Selanjutnya tikus diberi perlakuan ekstrak herba pegagan dan diukur penurunan kadar SGPT selama 14 hari.  Hasil penelitian menunjukkan adanya aktivitas hepatoprotektor pada ekstrak air herba pegagan dengan persentase penurunan kadar SGPT pada dosis 50 mg/200 g BB, dosis 25 mg/200 g BB, dosis 12,5 mg/200 g BB dan dosis 6,25 mg/200 g BB masing-masing sebesar 77,81%, 38,46%, 79,95% dan 55,20%. Berdasarkan penurunan kadar SGPT dan hasil histopatologi jaringan hati, ekstrak pegagan dosis 12,25 mg/200 g BB merupakan dosis paling efektif untuk menurunkan kadar SGPT pada tikus uji.Kata  kunci:  Herba  pegagan  daun  kecil,  hepatoprotektor,  Serum  Glutamat  PiruvatTransaminase (SGPT)

Abstract

The aim of this study is to compare the potential of ethanolic and water extracted of teki grasstuber on   white  male  rat strain Sprague  Dawley  at  two  months of age and ± 200 g of body weight. The rats were divided among four groups. Each group consists of six rats. The  groupswere ethanolic and water extracted  of  teki  grass tuber, positive control group (Paracetamol) and a negative control group (distilled water). Analgesic potency was tested by inducing pain stimuli previously to the animals, in the form of  heat stimuli  by putting it ona hot plate with  the temperature of 55°C. The responses of rats observed were licking and moving or lifting of their legs. Observations  were made for 1 minute, before  and  after  administration  of  pain stimulus at 15, 30, 45  and 60 minute. The results showed that ethanol and  water  extract  at an effective dose of 49 mg / 200 g of BW, gave relatively the same effect as an analgesic or   positive control group. Keywords : analgesic, ethanol and water extract of teki grass tuber, pain

Abstract

This study aimed to determine the effect of a combination of extracts of Noni leaves (Morinda citrifolia L) and fruit kandis (Garcinia cowa Roxb) on body weight of male rats induced by high- fat calories food Test animals were used 25 rats male Sprague Dawley strain age of 2 months with body weight ± 200 g. White mice were divided into 5 groups of 4 rats each. The treatment committed agains an obesity rats. The treatment group consisted of rats doses of 1 (8,75mg/kgBB noni leaves ekstract and sour fruit kandis 7mg/kgBB), dose II (4,75 mg/kgBB noni leaves ekstract and sour fruit kandis 3,5mg/kgBB), dose III (17,5 mg/kgBB noni leaves ekstract and sour fruit kandis 14mg/kgBB), the positive control group were given Orlistat and negative group were only given food to eat and drink. The observed parameters include weight loss, abdominal width, body length, skin fold, body mass indeks. The results showed that noni leaf extract and fruit kandis can be used as a weight loss with the most effective dose is 17,5 mg /kg BB noni leaf + 14 mg/kg BB fruit kandis with the most effective treatment is 14 days. Key words : Noni Leaf and fruit kandis, Feeding a High-Fat Calories,  Obesity Parameters, Rats male Sprague Dawley.

Abstract

Background: Nonsteroidal anti-inflammatory drugs (NSAIDs) gastropathy is a common complication, which has characteristic symptoms of dyspepsia syndrome. Mostly, it includes epigastric discomfort with bloating and nausea. The aim of this study was to provide evidences that clinical symptoms of dyspepsia are related to macroscopic changes of gastric form in rats, which are expected to be applied in human. Method: The study was conducted in 20 white rats (Rattus norvegicus, Sprague-Dawley strain) at the Department of Pathology and Clinical Reproduction, Bogor Agricultural University between January and December 2008. The rats were divided to treatment group and control group and each group consisted of 10 rats. Acetyl salicylic acid (ASA/aspirin) was administered at 400 mg dose, diluted in distilled water and was given to the treatment group using gastric cannula, once daily for three days period; while the control group had received aquabidest only. Subsequently, necropsies were conducted for both groups, followed by macroscopic observation and measurement of sagittal and transversal diameter. Gastric incisions along the minor curvature were performed in both groups to recognize any macroscopic changes of gastric mucosa. ANOVA test was utilized for data analysis, which was followed by Duncan test when the results were significant. Results: Gastric diameters in treatment group with positive lesion were significantly different from the control group and the treatment group with negative lesion on anthrum/pylorus region, with p < 0.05. Conclusion: Prominent gastric dilatation at anthrum/pylorus region found in the treatment group may become the initial cause and signs of dyspepsia in human. Abstract

Background: Mucus is pre-epithelial gastric layer that may prevent damages due to direct contact between aspirin and gastric epithelial cells. The integrity of gastric mucosa and mucous cellular reaction may serve as primary and secondary prevention of extended aspirin-induced gastric mucosal damage. The aim of this study was to prove the function of mucus as defensive factor in rats. Method: The study was conducted in twenty white rats of the Sprague-Dawley strain at Department of Pathology and Clinical Reproduction, Bogor Agricultural University, between January and December 2008. The rat in the treatment group were given 400 mg aspirin diluted in aqua bidest through intra- gatric canules; while the control group received aqua bidest only once daily for 3 days. Necropsies, macroscopic and microscopic observation were performed by counting the number of Alcian blue- periodic acid Schiff-stained mucous cells at fundus/corpus and antrum/pylorus regions. Data analysis was performed using ANOVA and Duncan test. Results: The number of mucous cells with positive lesions in the treatment group was significantly different from the control group at both regions. There was no significant difference of negative lesions between treatment and control group at both regions. At antrum/pylorus region, there was no difference of negative lesions between treatment and control groups; however, both groups demonstrated significant difference of positive lesions in treatment group. Conclusion: In primary prevention for gastric mucosal lesions, there is no increasing number of mucous cells in normal mucosa. Increasing number of mucous cells is a secondary prevention against extended aspirin-induced gastric mucosal damage. Keywords: NSAIDs/ASA, mucus cells, gastric mucosal lesion, rat
