Abstract

UJI AKTIVITAS ANALGESIK GEL FRAKSI KULIT MANGGIS (Garcinia mangostana L.) pada Mencit Jantan (Mus musculus) dengan Metode Hot Plate). Rasa nyeri dapat timbul salah satunya diakibatkan oleh adanya gangguan pada reseptor nyeri dimana reseptor nyeri tersebut merupakan ujung saraf bebas yang tersebar di kulit, otot, tulang dan sendi. Timbulnya rasa nyeri tentu akan menggangu aktivitas sehingga diperlukan obat penghilang rasa nyeri. Obat-obatan yang dikenal mampu meredakan rasa nyeri disebut sebagai analgesik. Faktanya, saat ini telah beredar berbagai macam obat analgesik di pasaran. Di samping itu, terdapat pula tumbuhan yang memiliki aktivitas sebagai analgesik yaitu manggis (Garcinia mangostana L.). Tujuan penelitian ini yaitu menilai dan membandingkan mula kerja dan aktivitas dua jenis analgesik yang diberikan secara topikal serta nilai ED50 dengan menggunakan metode hot plate. Penelitian dilakukan dengan membagi enam ekor mencit menjadi 4 kelompok Uji. Kelompok I sebagai kelompok perlakuan (Gel Manggis); kelompok II sebagai kelompok kontrol positif (Voltaren Gel); kelompok III sebagai kelompok normal (tanpa perlakuan); dan kelompok IV sebagai kelompok kontrol negatif (Basis Gel). Tiga puluh menit setelah pemberian perlakuan, tiap mencit ditempatkan di atas beaker glass pada hot plate dengan suhu 700C. Waktu yang terlewat antara penempatan hewan di hot plate dan adanya perilaku menjilati telapak kaki, gemetar, atau melompat dari permukaan dicatat sebagai respon latensi dalam hitungan detik. Hasil dalam penelitian ini yaitu gel manggis (Garcinia mangostana L.) konsentrasi 0,3%, 0,4%,dan 0,5% memiliki efek analgesik. Semakin tinggi konsentrasi gel Garcinia mangostana L. semakin tinggi daya analgesik yang didapatkan. Nilai ED50 yang diperoleh yaitu sebesar 0,037 mg/kgBB.

Abstract

PENGARUH PEMBERIAN INFUSA SELEDRI (APIUM GRAVEOLENS) TERHADAP EFEK DIURESIS MENCIT (MUS MUSCULUS L.) DENGAN METODE EKSPERIMENTAL MURNI. Pengujian pemberian infusa seledri (Apium Graveolens) yang dilakukan pada mencit untuk mengetahui efek diuresis mencit (Mus Musculus L.) dengan metode eksperimental murni. Tujuan dari penelitian ini adalah mengetahui prinsip dan etika penelitian dengan menggunakan hewan coba mencit (Mus Musculus L.), mengetahui efektivitas infusa seledri sebagai diuretik dengan tiga variasi dosis yaitu pemberian 50%, 100%, 150%, mengetahui dosis infusa seledri yang memiliki efek paling kuat sebagai diuretik dibandingkan dengan kontrol positif. Eksperimental ini menggunakan rancangan acak lengkap pola searah. Variabel kontrol dibedakan mejadi kontrol positif yaitu hewan uji yang diberikan infusa seledri dan kontrol negatif yaitu hewan uji yang tidak diberikan obat herbal (Infusa Seledri) melainkan diberikan akuades. Variabel bebas adalah dosis obat herbal (Infusa Seledri) yang diberikan. Variabel terikat adalah volume, pH , warna urin, dan waktu onset yang dihasilkan hewan uji (mencit). Hasil penelitian menunjukkan bahwa kurva hubungan antara dosis infusa seledri yang diberikan dengan volume urin yang dihasilkan mencit pada perlakuan minggu pertama hingga minggu ketiga diketahui bahwa volume urin pada minggu kedua merupakan volume urin rata-rata yang terbesar diantara perlakuan pada minggu ke I, II dan III. Konsentrasi 100% (pada minggu ke-II) mampu memberikan efek diuresis yang maksimal, konsentrasi 50% (pada minggu ke-I) memberikan efek diuresis lebih rendah dibandingkan perlakuan minggu ke-I, sedangkan dengan penambahan konsentrasi menjadi 150% (pada minggu ke-III) menunjukkan adanya efek diuresis namun lebih rendah daripada dengan perlakuan pada minggu ke-II denga pemberian dosis 100%. Berdasarkan hal di atas menunjukkan bahwa efek maksimal dari daun seledri pada penelitian ini ada pada konsentrasi 100%, dibuktikan dengan meningkatnya konsentrasi terjadi penurunan efek diuresis.

Abstract

UJI AKTIVITAS DIURESIS INFUSA SELEDRI (APIUM GRAVEOLENS) TERHADAP MENCIT PUTIH JANTAN (MUS MUSCULUS). Tanaman seledri (Apium graveolens L.) merupakan tanaman yang mengandung zat aktif yang banyak dimanfaatkan dalam pengobatan tradisional. Penelitian ini bertujuan untuk mengetahui keberadaan aktivitas infusa seledri (Apium graveolens L.) sebagai diuretik pada mencit jantan putih. Mencit putih jantan dibagi menjadi 4 kelompok perlakuan yaitu kelompok kontrol negatif (akuades), kelompok kontrol positif (suspensi furosemid), kelompok kontrol normal (tanpa perlakuan khusus), kelompok infusa seledri 50%, kelompok infusa seledri 100%, dan kelompok infusa seledri 150%. Pengujian terhadap efek diuretik dilakukan dengan melihat volume urin yang diukur setelah 24 jam perlakuan. Berdasarkan volume urin yang dihasilkan, hasil yang didapat menunjukkan adanya aktivitas diuretik pada infusa seledri.

Abstract

UJI AKTIVITAS ANALGESIK GEL BULUNG (Gracilaria sp.) TERHADAP MENCIT PUTIH JANTAN (Mus musculus). Penelitian ini dimaksudkan untuk mengetahui aktivitas analgesik gel bulung (Gracilaria sp.) sebagai alternatif obat analgesik. Evaluasi efek analgesik gel bulung pada hewan uji adalah dengan menghitung lama waktu bertahan mencit pada hotplate setelah diberi perlakuan uji, kontrol positif dan kontrol negatif pada hewan uji. Penelitian ini hanya meneliti satu variabel bebas saja yaitu variasi dosis obat gel fraksi bulung sangu terhadap variabel tergantungnya yaitu daya analgetik pada mencit yang berupa persen aktivitas analgetik dari gel fraksi bulung sangu. Kelompok uji (gel fraksi bulung sangu) dengan konsentrasi fraksi 1%; 1,25%; 1,5%. Dapat disimpulkan pada gel Voltaren® akan memberikan efek analgesik pada menit ke-30 setelah pemberian dan gel bulung akan memberikan efek analgesik pada menit ke-30 sampai 90 setelah pemberian.

Abstract

Mangosteen rind is a valuable waste, rich in saponins, xanton, and tannin. Liquid soap was made by saponification reaction of ethanol extract of mangosteen rind and fats are reacted with KOH. This study aims to perform the optimization of liquid soap formula and determine its physical and chemical properties. Extract of mangosteen (Garcinia mangostana L.) obtained by maceration for 3 days with 70% ethanol, and vaccuum evaporated. Liquid soap was prepared by heating method for ± 3 hours. The evaluation consists of organoleptic test, pH, and specific gravity determination. The results show that the liquid soap from 2% ethanol extract of mangosteen rind is dark brown, homogenous liquid, has a characteristic odor and specific gravity between 1.022 to 1.036 g / ml, and pH between 12.16 to 12, 92. From the test results, liquid soap formulas which meet commercial criteria are formula V and VI.

Abstract

Mangosteen peel has been studied to contain many chemical constituents. However, differences in the geographical location of a plant can result in variations in the content of a plant metabolites. This study aimed to determine differences in the characteristics of the extract as well as content owned chemical peel ethanol extract of mangosteen (Garcinia mangostana L.) were obtained from Tabanan and Karangasem regency, Bali province. This study is experimental designed.  Extraction process used the method of maceration with 70% ethanol. Organoleptic characterization of the extract was conducted. Water content, content of water-soluble extract and ethanol soluble extract were also determined. Phytochemical screening of the extract covered the content of alkaloids, glycosides, flavonoids, saponins, phenolic, and steroids / terpenoids. The results showed that the ethanol extract of mangosteen peel taken from Tabanan regency contain saponins, alkaloids, flavonoids, phenolics, terpenoids, and glycosides, while the ethanol extract of mangosteen rind taken from Karangasem regency contain saponins, alkaloids, flavonoids, phenolic, terpenoids. The ethanol extract of mangosteen peel taken from Tabanan regency has a different chemical content with ethanol extract of mangosteen peel taken from Karangasem regency.

Abstract

This study aims to evaluate the stability of herbal extract creams with a combination of neem leaves, gotu kola, and soursop extract to cure dermatitis complex in dogs., This herb extract has been able to inhibit in vitro the growth of bacteria that cause dermatitis. This research was conducted in the Laboratory of Non-Sterile Formulation and Technology at the Department of Pharmacy, Faculty of Mathematics and Natural Sciences, Udayana University. Cream preparations were made in three formulations; each formulation was evaluated for stability at room temperature. Evaluation of the stability of cream includes organoleptic test, homogeneity test, pH test, adhesion test, spreadability test, and viscosity test. The results showed the cream was stable at room temperature for one month and the stable formula was formula 1. It can be concluded that the herb of formula 1 extract cream was stable at room temperature for one month.

Abstract

Red calliandra (Calliandra calothyrsus Meissn.) leaf have the potential as phytoestrogens. In this study, adult female ovariectomized rats (as a model of menopause women) were given calliandra leaf extract containing estrogenic flavonoids. This study was an experimental design with a Completely Randomized Design (CRD). Forty-five ovariectomized female rats were divided into five groups i.e. untreated control (K), and the treatment group given 80% of C. calothyrsus ethanolic leaves extract doses 25 mg/ kg bw (P1), 50 mg/ kg bw (P2), 75 mg/ kg bw (P3) and 100 mg/ kg bw (P4). The test suspension was given once a day at 24-hour intervals by gavage as much as 0.5 ml/rat/day for two months. At the end of the treatments, blood was collected for blood biochemical tests including hemoglobin levels, hematocrit values, blood glucose and proteins levels. The results showed that C. calothyrsus leaf extract did not negatively affect the health status of the animals through blood biochemical parameters, i.e. hemoglobin levels, hematocrit values, and glucose and protein levels of ovariectomized female rats.

Abstract

Background: Maceration is a non-heating extraction method the result of which is affected by the type of solvent and the maceration time. Objective: To get the higher alpha-mangostin concentration (Garcinia mangostana L.) rind that was macerated using ethyl acetate solvent and to determine the optimum maceration time needed to produce high concentration of alpha-mangostin. Methods: the mangosteen rind was macerated using ethyl acetate with the variation of the maceration time of 6, 12, 24 and 48 hours. The extract was then dried using the freeze drying method. The optimum maceration time was determined by looking at the highest concentration of alpha-mangostin from each extraction time using TLC-densitometry method with a silica gel stationary phase of GF254 and a mobile phase of chloroform—methanol (10:0.1, v/v). Results: The results showed that the difference in the maceration time can affect the concentration of alpha-mangostin compound contained in the extract. Conclusions: It was found out that the 24-hour maceration time produced the highest concentration of alpha-mangostin that is 3031.34 ng.

Abstract

Abstract Alpha-mangostin is a xanthonoid compound contained in mangosteen rind (Garcinia mangostana L.) and has an antibacterial effect against Staphylococcus aureus, the bacterium that causes acne. In this study, mangosteen rind extract is formulated in a gel dosage form. This study aimed to determine the optimum formula, the physical and chemical properties as well as the optimum formula release profile of the gel dosage form of mangosteen rind extract. The mangosteen rind extract gel formula consists of viscolam, propylene glycol, glycerin, microcare®, ethyl acetate extract of mangosteen rind, and distilled water. The formula was optimized by varying the concentrations of viscolam (2% and 5%), propylene glycol (5% and 20%) and glycerin (2% and 15%) using a factorial experimental design program called Design Expert 7.0.0. The 8 formulas were developed into gel dosage form and the physical and chemical properties were then evaluated. The evaluation tests include viscosity, dispersive power, and pH tests. The evaluation results were processed using the Design Expert 7.0.0 program to determine the optimum formula. The results of the analysis showed the optimum formula of gel dosage form of mangosteen rind extract with viscolam percentage of 4.97%, propylene glycol of 9.91%, glycerin of 12.23%, microcare® of 0.3%, TEA (q.s), ethyl acetate extract of mangosteen rind of 1%, and distilled water of 45%. The evaluation results of the physical and chemical properties of the gel optimum formula revealed the viscosity of 2,345 cps, dispersive power of 6.59 cm, and pH of 6.74. Alpha-mangostin release test on the optimum formula revealed a flux value of 41.327 ?g/cm2/t1/2. From these results, it is concluded that the optimum formula has met the physical and chemical characteristics of a good gel.
