Abstract

White-tailed forest mice (Maxomys hellwandii) are endemic to the Sulawesi. This study aims to determine the genetic diversity of white-tailed forest mice (Maxomys hellwandii) based on RAPD. The analysis of the variability of white-tailed forest rats was carried out in several stages, namely the extraction and purification of DNA, the amplification of the target gene using the PCR method, and finally visualization of the amplification results with 0.8% agarose gel electrophoresis. Data analysis using the SPSS IBM Ver. 20 program. Results obtained using OPA-3 primers from four different localization samples show samples of Likupang 1 and South Minahasa show similarity of 1%, and samples of Likupang 2 and Southeast Minahasa show similarity of 25%. Whereas the primary OPA-2 samples from the villages of Likupang 1, Likupang 2 and South Minahasa show a similarity of 8% and the sample of Southina Minahasa has a similarity of 25%. On the basis of the dendogram grouping of four different samples, he showed genetic diversity.

Abstract

. Nutmeg fruit is widely used in the medicine formula and also various kinds of processed foods and beverages. Nutmeg meat can be processed into syrup, fruit juice, jam, dodol pala and wine. The fruit is believed to be one of the natural habitats of bacteria, including lactic acid bacteria (BAL). The study aims to determine the type of BALES isolated in nutmeg grapes (Myristica fragrans Houtt) which has the ability to produce exopolysaccharides. Based on the results of the study obtained 15 BAL isolates namely AP1, AP2, AP3, AP4, AP5, AP6, AP7, AP8, AP9, AP10, AP11, AP12, AP13, AP14, and AP15. The exopolysaccharides produced by the isolates are 400 mg/L, 530 mg/L, 1200 mg/L, 730 mg/L, 60 mg/L, 460 mg/L, 600 mg/L, 200 mg/L, 930 mg/L, 1130 mg/L, 200 mg/L, 130 mg/L, 1000 mg/L, 800 mg/L. The next 15 isolates are identified using the profile matching method. The results of the identification show that 15 BAL isolates were included in the genus Lactobacillus and are a species of Lactobacillus bulgaricus.    Keyword : Isolation, Identification, nugmeg grapes (myristica fragrans houtt), lactic acid bacteria, exopolysaccharides

Abstract

White-tailed forest rats (Maxomys hellwandii) are other meat-producing animals that are consumed by the Minahasa community as exotic food. This study aims to determine the nutritional content of white-tailed forest rat meat. Nutrient content analysis was performed at the UNIMA chemistry laboratory and the BARISTAND laboratory in Manado. The procedure for this study is the analysis of nutrient content using the procedure of the Association of Official Analytical Chemists (AOAC 2005). The results of research on fresh forest rat meat showed the content of water, ash, fat, crude fiber, protein and carbohydrates 71.17%, 0.97%, 3.17%, 0.03%, 19, 16%, 5.53% and roasted forest rat meat, i.e. 69.14% water, 1.29% fat and 4.5% protein  Â
