Abstract

Based on a study conducted by the WHO Global Burden of Disease in 2010 stated, OA ranks 11th, the cause of disability in the world, and ranks 6th in East Asia and the Asia-Pacific countries, ranked 10th in North America, ranked 7th in Eastern Europe , and ranked 13th in Western Europe. Pathogenesis of osteoarthritis (OA) as degenerative joint disease that was once thought caused by a mechanical process. The prevalence, etiology, pathogenesis and progression of OA are still not understood until now, mainly because of the many influencing factors. This type of research is a analytic study. It was conducted in Rheumatology Polyclinic Dr. Mohammad Hoesin Palembang from February to December 2019. Affordable research population is all genu osteoarthritis sufferers with effusion who go to Dr. RSUP Mohammad Hoesin Palembang as 80 samples. Comparison of NFATC1 mRNA levels in genu osteoarthritis patients with effusion shows that there is no difference in gender and history of diabetes, whereas hypertension history and degree of osteoarthritis based on Lawrence Kalgren shows significantly different expression of NFATC1 mRNA. Age with NFATC1 mRNA expression showed no significant correlation, in contrast to BMI, Stroke Test and joint effusion volume showed a weak significant correlation, even though the direction of the correlation was negative on Stroke Test and joint effusion volume.

Abstract

Gout is a dominant disease in adult man and found in woman before menopause period. Prevalence of gout n the United States 3.9%, Britain 2.5%, New Zealand 6.1% and in Indonesia 7%. There are 2 pillars of gout treatment, pharmacology and non pharmacology. Non-pharmacological therapy includes patient education, diet and joint rest. Lack of knowledge about lifestyle and things that can cause gout is one of the factors that can increase the incidence of gout arthritis. The aim of this study was to identify patient knowledge about gout arthritis in Rheumatology Clinic Central General Hospital Dr. Mohamma Hoesin Palembang. A total of 80 patients who fit the inclusion criteria were included in the study. The data were obtained from interview of patients at Rheumatology Clinic Central General Hospital Dr. Mohammad Hoesin Palembang on September-November 2019. The result of this study showed that from 80 patients, 21 patients suffered from gout arthritis and 59 patients suffered from non gout arthritis. Knowledge level of gout patients were enough (52,4%) and good (47,6%) meanwhile knowledge level of non gout patient are bad (52,5%), enough (45,8%) and good (1,7 %). Knowledge level of gout patients are better than non-gout patient's in Rheumatology Clinic Central General Hospital Dr. Mohammad Hoesin Palembang.

Abstract

Background: Centella asiatica (L.) Urb is a native herb from Asian countries such as India, China, and Indonesia. This herb has been widely used as a cure for various diseases. However, studies investigating the aqueous extract of Centella asiatica as a nootropic in healthy individuals are still very limited. Objective: This study aims to investigate the potential of aqueous extract of Centella asiatica in enhancing cognitive function of healthy male Wistar rats. Methods: Rats were randomly allocated to four treatment groups, i.e. without treatment and aqueous Centella asiatica extract at doses of 200, 400 and 800 mg/kg. To determine enhancement of cognitive function, novel object recognition (NOR) test was conducted after the course of treatment. Acetylcholine content was assessed by enzyme-linked immunosorbent assay. Results: There was a significantly high preference index towards the novel object in the NOR test in groups treated with 200 mg/kg and 800 mg/kg of the aqueous extract compared to control. This was further confirmed by a significant increase of brain acetylcholine content in rats treated with 200 mg/kg of the extract. Conclusion: Therefore, this study confirms that the aqueous extract is effective in enhancing cognitive performance of healthy Wistar rats.

Abstract

Background: Low bone density has been known as one of the factors that increased fracture risk. Aging, estrogen deficiency, low BMI, and inadequate calcium intake are known factors that contribute to decreased bone density. Other than the known factors, some research show that sleep duration can also lower bone density. Sleep affects bone density in a way that it causes increased level of cortisol, proinflammatory cytokine, and decreased in physical activity. Method: This study is an observational analytic study with cross-sectional design. It is conducted on menopausal patients that has been examined with bone mineral densitometry at Poliklinik Reumatologi Penyakit Dalam RSUP Dr. Mohammad Hoesin Palembang in November 2017 to Oktober 2018. Samples are collected using total sampling technique. Results: From 93 subjects that participated in this study, 20,4% of them had osteopenia, and 51,6% had osteoporosis. Patients with low bone density, mainly slept for 6 to 8 hours at night per day, 30 minutes of nap per day, and has a total sleep duration for less than 8 hours each day. This study shows no significant association between night-time sleep, daytime napping, and total sleep duration with bone density (p value = 0,168, p value = 0,831, p value = 0,984). Analysis on other risk factors show significant association between body mass index and low bone density (p value = 0,002). Conclusion: There are no significant association between sleep duration and bone density in menopausal patients at Poliklinik Reumatologi RSUP Dr. Mohammad Hoesin Palembang.

Abstract

Introduction. Along with the COVID-19 pandemic, the need for a thermal cyclerdevice for examining COVID-19 is getting bigger. Many laboratories have overusedthe use of thermal cyclers due to the limited availability of this tool. This study aimsto test the effectiveness of the Sriwijaya thermal cycler based on a smart controllerin conducting PCR DNA sequences. Methods. The research design was anexperimental study with a posttest control group approach, in order to see anoverview of the results of the polymerase chain reaction (PCR) in the form ofintermediate DNA bands using Sriwijaya Thermal Cycler compared to groups usingfactory-made Thermal cyclers. Results. The PCR results showed precise results fromthe image and band separation, and there was a clear separation of the bands onthe marker. The PCR results from the ACE I / D gene showed quite good and optimalresults in the separation of the PCR results band. Conclusion. Sriwijaya thermalcycler is effective in DNA polymerase chain reaction (PCR) sequences comparable tothat of the manufacturer's Thermal Cycler.

Abstract

Rheumatoid Arthritis or RA disease is a chronic inflammatory and systemic disease associated with broad synovitis resulting in erosion of the articular cartilage and marginal bone causing joint damage. RA is an autoimmune disease with the discovery of an autoantibody, namely rheumatoid factor. The relevant antibody is anti-citrullinated protein (ACPA) antibody. Citullination on the introduction of several proteins such as fibrin, vimentin, fibronectin, collagen type II, which is expressed in the synovial membrane during inflammation by ACPA. In a recent study increased HDAC activity in the synovial tissue of RA patients has increased. Histone deacetylase or abbreviated as HDAC is an enzyme that is important in regulating gene transcription by altering the acetylation of histone proteins which results in an important regulation of repression in the implementation of inflammation. HDAC enzyme inhibitor (HDACi) is an HDAC enzyme inhibitor that can provide benefits for the treatment of various diseases including malignancy and inflammation. In inflammatory disease HDACi overcomes inflammatory cytokines such as TNF-α, IL-6 and IL-1β. One of the HDAC classes is HDAC1 which is highly expressed in the synovial fibroblasts of RA patients. HDACi can burden swollen joints, reduce mononuclear cell infiltration, request pannus orders, inhibit bone and cartilage damage.
