Abstract

Abstract: The skin of the pineapple (Ananas comosus L) in the form of extracts already researched can inhibit the growth of bacteria cause dental caries i.e. Streptococcus mutans. Pineapple-skin contains an enzyme bromelin that has easily degraded and lipofil, therefore required a system of delivering medicines that can increase the stability of the enzyme bromelin in pineapple-skin extract, so the activities antibacterial can be improved. The system transfers that can be developed is using nanoparticles with a form of material nanoemulsion. Nanoemulsion in nanomouthwash shape created by using spontaneous nanoemulsification method. Nanomouthwash tested physical characteristics include: determination of the particle size, polydispersity index, organoleptic, transmitan test, viscosity, and weight of type. Furthermore, nanomouthwash of pineapple-skin extract tested activity against Streptococcus mutans bacteria and computed the diameter zone of inhibition (DDH). The results the DDH were statistical analysis with Kruskall Wallis's method to see a meaningful difference. The results of the test of the physical characteristics of nanomouthwash of pineapple-skin extract has a particle size 86,16 nm, polydispersity index 0,29, tanned yellow liquid, the smell of oil typical sweets and have a sweet taste with mint sensation. Test results of viscosity 13,07 cp; weights types 1,11; and has a value of pH is 5. The antibacterial activity test of nanomouthwash of pineapple-skin extract shows the value of the diameter zone of inhibition to Streptococcus mutans of 8.6 mm smaller than pineapple-skin extract of 13.8 mm.Keywords : Pineapple-skin, nanoemulsion, mouthwash, physical characterictics, Streptococcus mutans Abstrak: Kulit nanas (Ananas comosus L) dalam bentuk ekstrak dengan konsentrasi 25% telah diteliti dapat menghambat pertumbuhan bakteri penyebab karies gigi yaitu Streptococcus mutans. Kulit nanas mengandung enzim bromelin yang memiliki sifat mudah terdegradasi dan bersifat lipofil, oleh karena itu diperlukan suatu sistem penghantaran obat yang dapat meningkatkan stabilitas enzim bromelin, sehingga aktivitas antibakteri dapat ditingkatkan. Sistem penghantaran yang dapat dikembangkan adalah nanopartikel dengan bentuk sediaan nanoemulsi. Sediaan nanoemulsi dalam bentuk nanomouthwash dibuat dengan menggunakan metode nanoemuslifikasi spontan. Nanomouthwash ekstrak kulit nanas diuji karakteristik fisiknya meliputi: ukuran partikel, indeks polidispersitas, organoleptis, pH, viskositas, dan bobot jenis. Selanjutnya, nanomouthwash ekstrak kulit nanas diuji aktivitasnya terhadap bakteri  Streptococcus mutans dan dihitung diameter daya hambatnya (DDH). Hasil DDH dianalisa statistika dengan metode Kruskall Wallis untuk melihat adanya perbedaan yang bermakna. Hasil pengujian karakteristik fisik sediaan nanomouthwash ekstrak kulit nanas memiliki ukuran partikel 86,16 nm, indeks polidispersitas 0,29, berbentuk cairan jernih berwarna jingga kecoklatan, berbau khas mint, dan memiliki rasa manis diikuti sensasi mint. Hasil uji viksositas sebesar 13,07 cp; bobot jenis 1,11; dan memiliki nilai pH 5. Pengujian aktivitas antibakteri nanomouthwash ekstrak kulit nanas menunjukkan nilai diameter daya hambat terhadap bakteri Streptococcus mutans sebesar 8,6 mm lebih kecil dibandingkan dengan ekstrak kulit nanas yaitu sebesar 13,8 mm.Kata kunci: Ekstrak kulit nanas, nanoemulsi, mouthwash, karakteristik fisik, Streptococcus mutans
