Abstract

BackgroundPapillary thyroid carcinoma is the most common endocrine gland malignancy that has the highest incidence rate of 60-80%. In papillarythyroid carcinoma BRAF V600E mutation plays an important role in carcinogenesis. BRAF V600E is a RAS-regulated-serine-threoninekinase and activator of the MAPK pathway cascade that functions to regulate cell proliferation, differentiation and survival. BRAFV600E mutations in PTC is associated with a high incidence of recurrence and reduced survival. The purpose of this study is to lookat the relationship of BRAF V600E expression with histopathological variants and lymphocyte infiltration in PTC.MethodsThis research was a retrospective observational cross sectional study with 40 cases of papillary thyroid carcinoma which werediagnosed in 4 Pathology Anatomy Laboratory in West Sumatera. Samples were obtained from paraffin blocks of tissue from surgeryand were reevaluated for histopathological variant and lymphocyte infiltration. Expression of BRAF V600E in tumor cell were analyzedusing immunohistochemistry staining. Bivariate statistical analysis of Chi-Square were used and p value <0,05 were consideredsignificance.ResultsPositive BRAF V600E expression was found in 24 case (60%). Positive BRAF V600E expression was mostly found in classic PTC(50%). Papillary thyroid carcinoma with lymphocyte infiltration mostly found with negative BRAF V600E expression (79.2%). Statisticalanalysis showed no significant correlation between BRAF V600E with histopathological variant (p=0.74) and no significancecorrelation between BRAF V600E expression with lymphocyte infiltration (p=0.12).ConclusionThe conclusion is BRAF V600E expression has no correlation with histological variants in papillary thyroid carcinoma and no significantcorrelation of BRAF V600E expression with lymphocyte infiltration.

Abstract

Latar belakangKarsinoma ovarium serosum adalah keganasan organ reproduksi wanita yang paling sering mematikankarena bersifat sangat progresif. Berdasarkan temuan terbaru, WHO membagi karsinoma ovariumserosum menjadi karsinoma serosum derajat tinggi dan rendah. Gen p53 berperan dalam patogenesismolekuler karsinoma ovarium serosum khususnya pada derajat tinggi. Mutasi p53 pada karsinoma ovariumserosum diduga berperan pada progresivitas tumor dan berhubungan dengan derajat histopatologikkarsinoma serosum. Penelitian ini bertujuan untuk menganalisis hubungan ekspresi p53 mutant danderajat histopatologik pada karsinoma ovarium serosum.MetodePenelitian ini adalah studi komparatif potong lintang analitik dengan populasi kasus karsinoma serosumovarium, yang didiagnosis di Laboratorium Patologi Anatomik di Sumatera Barat tahun 2013-2015.Berdasarkan kriteria World Health Organization (WHO) 2014, didapatkan 32 kasus yang direview derajathistopatologik. Selektif blok parafin dipisahkan dan diwarnai dengan antibodi monoklonal p53 (Clone EP9;Biocare).Ekspresi p53 dibagi menjadi ekspresi positif dan negatif. Hubungan antara ekspresi p53 danderajat histopatologik dianalisis dengan uji chi-square (p<0,05).HasilKasus karsinoma ovarium serosum memiliki rentang usia 29-67 tahun, dengan usia rata-rata 49,50 untukderajat tinggi dan 45,44 untuk derajat rendah. Ekspresi p53 menunjukkan hasil positif pada 12 kasuskarsinoma serosum derajat tinggi (75%) dan 4 kasus derajat rendah (25%). Terdapat hubungan bermaknaantara ekspresi p53 dengan derajat histopatologik karsinoma serosum ovarium (p=0,013).KesimpulanEkspresi p53 mutan berhubungan dengan derajat histopatologik karsinoma serosum ovarium

Abstract

Background Cervical squamous cell carcinoma (CSCC) represents 70-80% of all cervical cancer cases. This cancer ranks 4th most common death cause due to malignancy in women. Cervical SCC patients were found at an older age and were detected at an advanced stage. High risk human papillomavirus (HPV) infection is main cause of SCC. Protein cyclin-dependent kinase inhibitor 2A also called p16 is a well-known tumor suppressor protein involved in cell cycle regulation, senescence, apoptosis, cell invasion and angiogenesis. This p16 protein can be used as surrogate marker for HPV infection. From several previous studies, there were differences between p16 expression relationship with differentiation degree and cervical SCC tumor stage results. Relationship research between p16 expression and differentiation degree and tumor stage of cervical SCC patients has never been conducted in West Sumatra. Methods This research was retrospective observational cross-sectional study with 35 cervical SCC cases that have been diagnosed at Anatomical Pathology Laboratory, RSUP Dr. M. Djamil Padang period January 2018-December 2019. Samples were obtained by simple random sampling from paraffin blocks derived from tumor tissue. The histopathological slide was reassessed to assess differentiation degree and tumor stage data were obtained from the patient's medical record. p16 expression in tumor cells was seen by immunohistochemical examination. To determine the correlation, bivariate statistical analysis was performed using the chi-square test with significance p<0.05 level. Results p16 high expression was found in 22 cases, tended to be higher in degree of well differentiation (40.9%) and moderate differentiation (50.0%) and was higher at an advanced stage (90.9%). Statistically, there is no correlation between p16 expression and differentiation degree with p value=0.138. There was no correlation between p16 expression and tumor stage with p value=0.337. Conclusions This study concluded that p16 expression was not associated with differentiation degree and tumor stage in cervical SCC.
