Abstract

Highly  pathogenic avian influenza (HPAI) H5N1 virus is a known pathogen in birds. Recently, the virus has been reported to cause sporadic fatal disease in tigers, leopards, and other exotic felids as well as domestic cats  in Thailand. The present study was carried out to investigate the presence of AI H5N1 virus infection in stray cats roaming around residential, traditional and chicken farms in Bogor, West Java. Ninety serum samples were tested using HI test to screened for the presence of antibody to AI H5N1. Virus isolation was done in SPF embrionated chicken eggs and identify using HI, AGP and RT-PCR. The results showed that 18,9% of stray cats developed antibodies against H5 with geometric mean titre 23,1 . Stray cats lived in traditional markets 18–40% developed antibodies in the titre ranging from 22,8  to 24,5. Only two out of nine stray cats which lived in chicken farm developed low antibody titres again H5 (21). None of the stray cats lived in residencial area have developed antibodies against H5. This study revealed that stray cats have been contact with AI H5. Avian influenza H5 viruses were isolated in eight out of 33 pooled of rectal swab samples. The viral cleavage site sequences are CCTCAAAGAGAGAGC AGAAGAAAGAAGAGAGGT which represent amino acid sequences of PQRESRRKKRG. Based on the cleavage site sequence, the isolates are similar with the AI H5 virus subtype isolated from human in Indonesia during 2005–2007.

Abstract

Identification of pathotype of Avian Influenza Virus (AIV) subtype H5N1 isolates is very important. Thisresearch aimed to identify the pathotype of AIV subtype H5N1 isolated from household waterfowls in West Javabased on molecular markers of amino acid sequences of the Hemagglutinin (HA) cleavage site. Fragments of HAgenes of 21 isolates were amplified using RT-PCR with a primer pair that flanking the cleavage site region, andsequenced with dideoxy-termination method with ABI automatic sequencer (Applied Biosystems). Multiple alignmentof nucleotide and their deduced amino acid sequence were analyzed using ClustalW from MEGA 3.1 program.The result shows that all H5N1 isolates (21 isolates) possess polybasic cleavage sites with 2 patterns ofamino acid sequence, i.e QRERRRKKR (20 isolates) and QRESRRKKR (1 isolate). This finding indicates that all ofthe viruses isolated in this research were of highly pathogenic avian influenza (HPAI) strains.Keywords: cleavage site, waterfowls, HPAI

Abstract

A study was conducted to disclose the habit, food type palatability, and the nest type preference of gelatik jawa (Padda oryzivora). As many as 54 gelatik jawas from Surabaya, Malang, Ngawi, Bekasi, Karawang, Cirebon, Bogor, Semarang, Magelang, Yogyakarta and Prambanan were captured using soft nylon net and then caged for two weeks for adaptation. Each bird was given a ring to make it easier to identify. After the adaptation period, the birds were moved to a wider cage. Every day the habit and mating behaviour as well as the preference of food and nest were observed. The highest  frequency of daily feeding (997.75 ± 72.23) and moving (795.50 ± 138.27); and the lowest  frequency of daily self care (4.126 ± 138.27) and communicate (93.75 ±  7.59) were recorded at 08.00 – 10.00. Food palatability and preference over nest, habit, and biological clock of gelatik jawa could be the basis of ex-situ conservation. The monogamy character of gelatik jawa could also serve as the basis of ex-situ conservation that male and female in one cage must be in equal number.

Abstract

Keong mas (Pomacea canaliculata) hama padi yang banyak ditemukan di Mijen, Kota Semarang, diolah menjadi pupuk organik cair asam amino (pocamino) karena memiliki kandungan protein tinggi dan lemak yang rendah. Hal tersebut merupakan kajian potensi dan masalah lokal, dapat dimanfaatkan sebagai sumber penyusunan bahan ajar kontekstual untuk memberikan pengalaman belajar nyata bagi siswa. Bahan ajar yang dikembangkan berbentuk booklet, berukuran kecil dan tipis, berisi penjelasan ringkas, sistematis, dilengkapi gambar/ilustrasi. Penelitian ini bertujuan untuk memaparkan pengembangan booklet berdasarkan kajian potensi dan masalah lokal sebagai suplemen bahan ajar materi pemupukan di SMK Pertanian dan menganalisis kelayakan booklet. Merupakan penelitian dan pengembangan yang diawali riset di lapangan mengenai pengolahan hama keong mas menjadi pocamino. Data yang diambil yaitu analisis kebutuhan lingkungan dan sekolah, data kandungan dan hasil aplikasi pocamino, data kelayakan booklet oleh penilaian ahli, pengguna dan hasil belajar siswa. Hasil penelitian menunjukkan, pocamino memiliki kandungan unsur hara N, P, K dan asam amino dengan kadar yang beragam. Disimpulkan bahwa booklet yang dikembangkan berdasarkan kajian potensi dan masalah lokal dinilai layak sebagai suplemen bahan ajar materi pemupukan di SMK Pertanian karena memperoleh skor rata-rata 4,7 dari ahli, skor 95,7% dari tanggapan guru, 88,87% dari siswa dan mendapatkan hasil belajar dengan nilai rata-rata 89,2.
