Abstract

This study aimed to prove that giving hCG injection at the time of artificial insemination and five days afterartificial insemination in dairy cows with repeat breeder can cause pregnancy. This study employed 30 dairycows with repeat breeder. All samples of dairy cows were divided into two treatments, P1 using hCG at adose of 100 IU given intramuscularly at the time of artificial insemination and P2 using hCG at a dose of100 IU intramuscularly given five days after artificial insemination. The results were analyzed using the Chi-Square test. Analysis using Chi-Square showed that all treatments caused pregnancy (100%). In summary,hCG injection at the time of artificial insemination and five days after artificial insemination in dairy cowsexperiencing repeat breeding can cause pregnancy.

Abstract

This research was aimed to determine the addition of L-arginine on motility and sperm plasma membrane of Sapudi sheep after two hour equilibration time in egg yolk tris diluent. This research used fresh samples of sheep Sapudi semen collected by artificial vagina, then were devided into 4 treatments and 6 replications. Analysis of the data using Analysis of Variance (ANOVA) then proceed to the Duncan Test to determine significant differences between treatments. The first treatment P0 was no L – Arginine added as control. P1 was treated with 0,004 M L – Arginine. P2 was treated with 0,005 M L – Arginine, P3 was treated with 0,006 M L – Arginine. Result of two hour equilibration time for the sperm motility showed: (P0) 46,67a   5,16, (P1) 49,16ab   5,84, (P2) 55,00bc   7,74, (P3) 58,33c   6,83. Result of two hour equilibration time for the sperm plasm membrane were: (P0) 39,33a   4,92, (P1) 43,33ab   4,22, (P2) 46,50 bc   3,44, (P3) 50,83 c   3,48. The addition of L-arginine with a concentration as much as 0.006 M, shows the highest result in sperm motility and sperm plasm membrane intact of Sapudi semen.

Abstract

Reproduction is one of essential factor directly affecting profitability in a dairy production system. Some of the reproductive factors that influenced to the developing of dairy cattle of small holder’s farmer are Conception Rate (CR) Service per Conception (S/C). The study was conducted to know the CR and S/C of Friesian Holstein in KUD Argopuro, Probolinggo. The data is taken primary and secondary data. Primary data retrieval is done by direct observation, which includes several variables, where the variables include : the identity of the breeder, cages, and feeding and drinking. As for the secondary data obtained by recording the card Artificial Insemination (AI) owned by Inseminator in KUD Argopuro. The data which by collected from this sample are CR and S/C. This research was a case study. The materials used cattle as the acceptors were 10% from 728 lactation cattle. Data from the results of this research on CR (30%) and S/C (2,49). The conclusion of this research for CR the result were lower than normal and S/C the result were higher than normal.

Abstract

The aim of this study was to find out protein tyrosine kinase profile in Merino Sheep seminal plasma. This study consist of collecting semen of Merino Sheep containing plasma and spermatozoa, the separation between seminal plasma and spermatozoa, then tyrosine kinase analysis using Sodium Dodecyl Sulphate Polyacrilamide Gel Electrophoresis (SDS-PAGE). Semen was collected by artificial vagina and then centrifugated for 40 minutes at 4000 rpm to separated seminal plasma and spermatozoa. Protein was analyzed using SDS-PAGE, to separate each protein based on their molecular weight. The result showed that there were 13 protein bands in 3 seminal plasma samples with an average 149,63 kDa, 139,7 kDa, 114,97 kDa, 109,3 kDa, 97,33 kDa, 93,83 kDa, 86,23 kDa, 77,6 kDa, 64,6 kDa, 52,3 kDa, 41,93 kDa, 38,13 kDa, and 34,5 kDa. In conclusion, it is believed that tyrosine kinase located on the sixth band with molecular weight 93,83 kDa.

Abstract

Aims of this research was to determine the effect of the duration of straw count and the position of straw from liquid nitrogen surface to the motility and viability of post thawing frozen Madura cattle. This study used a sample derived from Madura cattle (Pasean OO 1220) from BBIB singosari as much as 54 straw divided into 9 treatments. P1 (1cm 10 sec), P2 (1cm 20 sec), P3 (1cm 40 sec), P4 (2cm 10 sec), P5 (2cm 20 sec), P6 (2cm 40 sec), P7 (3cm 10 sec), P8 (3cm 20 sec), P9 (3cm 40 sec) and repeated as much as 6 times then observed for motility and viability of post thawing spermatozoa. The data obtained were analyzed using SPSS 16.0. The results showed that the duration of straw count and the position of straw from liquid nitrogen significant (P<0,05) on post thawing motility. The position of straw from liquid nitrogen signifant (P<0,05) on post thawing viability. The conclusion of this research showed that 1 cm 40 sec give the higher post thawing motility.

Abstract

The purpose of this study was to prove that the addition of Vitamin B12 in egg yolk skim milk extender can maintain the motility and viability of post thawing Sapudi sheep spermatozoa. This study used sample of Sapudi sheep fresh semen that was collected with artificial vagina, and divided into 3 different treatments. The control treatment or P0, Sapudi sheep semen was diluted in the egg yolk skim milk extender + vitamin C 0,02 mg / ml, treatment group (P1 and P2), Sapudi sheep semen was diluted in the egg yolk skim milk extender+ vitamin B12 5 μg / ml and grouped in P1, Sapudi sheep semen was diluted in the egg yolk skim milk extender + Vitamin C 0.02 mg / ml + Vitamin B12 5μg / ml and grouped in P2. The data analysis was use One Way Analysis of Variant (ANOVA), then to identify the real difference among the treatments continued with Duncan distance test. The result of data analysis showed that P2 treatment was significantly different (p <0.05) with P1 treatment and P0 treatment or control. While P1 treatment was not significantly different (p> 0,05) with P0 treatment or control. The result showed that the addition of vitamin C 0.02 mg / ml with vitamin B12 5 μg / ml combination in egg yolk skim milk extender was the best addition to maintain the motility and viability of the post thawing Sapudi sheep spermatozoa.

Abstract

The purpose of this research is to determine the effect of mucos acidity of vaginal mucous when estrus towards conception rate at KUD Tani Wilis Tulungagung District and KSU Tunas Setia Baru Pasuruan District. Measured samples were taken from 30 dairy cattles in each KUD. Mucos acidity of vaginal mucous were obtained by using a digital acidity meter before insemination was done on dairy cattle. After measuring the acidity from mucosal vagina mucous, artificial insemination has done on estrus dairy cattle. On the 60th day after insemination, rectal palpation was done to check the conception rate. Research result from 30 dairy cattle on each KUD showed that mucos acidity of vaginal mucous has signifficant effect towards conception rate. Mucos acidity of vaginal mucous range 6.5-8.5 with the highest range of conception rate in KSU Tunas Setia Baru Pasuruan District 7.5-7.7 and in KUD Tani Wilis Tulungagung District with the highest range of conception range 7.2-7.4 and 7.5-7.7.

Abstract

The study was conducted to know Conception Rate and Service per Conception of  Bali cattle acceptors in west Sumbaw district. Data were collected fot the periode of January to September 2017 from inseminator in west Sumbawa district. The data were taken by primary and secondary data. Primary data retrieval is done by direct observation, which includes several variables, where the variable include the identitiy of the breeder, the cages, feeding, drinking and knowladge of breeders. The secondary data were obtained by recording of the officer insemination. All data was analyzed to find a percentage of Conception Rate and find number of Service per Conception (S/C) by analyzed descriptive of SPSS. Risults of this research showed that Conception Rate and Service per Conception’s average of Bali Cattle is 60 % and 1,7. The conculusion of this research showed that Conception Rate is lower than normal value and Service per Conception of Bali cattle in West Sumbawa district is better. This research was expected to be a reference for the next artificial insemination programmed by the government.

Abstract

This study aimed to determine the relationship between heat detector reading results and the feature of vaginal swab preparations as a parameter for determining estrus in Peranakan Etawa goats. Five healthy non-pregnant goats weighing 20-30 kg, were synchronized using PGF2α (7.5 mg Dinoprost, Enzaprost) intramuscularly twice with 11 days interval. Vaginal cytology examination began the day after the second PGF2α injection. The heat detector (Draminski®, Dramiński SA, Poland) is an estrus detecting device based on the amount of electric current flowing through the vaginal mucus. Spearman correlation analysis showed a relationship (p <0.05) between heat detector reading and estrus detection based on vaginal cytology. The heat detector readings were very significantly correlated positively (p <0.01) with the number of epithelial cells (coefficient correlation, r= 0.564), but negatively correlated with the number of cornified cells (r = –0.854), and also significantly correlated positively (p <0.05) to the number of leukocytes (r = 0.487). The estrus stages were significantly correlated positively (p <0.05) only to the number of leukocytes (r = 0.918). Likewise, the number of epithelial cells was highly correlated negatively (p <0.01) only to cornified cells (r = –0,643), and the number of cornified cells was significantly correlated negatively (p <0.05) only to the number of leukocytes (r = –0.472). It could be concluded that the features of the proestrus, estrus and metestrus stages based on the goat vaginal cytology was correlated with the reading results of the heat detector.

Abstract

This research was conducted to identify non-specific bacteria found in the cervical mucus of dairy cattle in the follicular and luteal phase. Twenty cervical mucus samples were collected from ten cows during their follicular phase and ten cows during their luteal phase. Sampling was carried out with the help of an artificial insemination gun and a plastic sheath, which were protected by a sheath cover. After insertion into cervix, the tip of the plastic sheath was cut and put into a tube prefilled with phosphate buffered saline. All samples were cultured in Blood Agar media and isolated in Eosin Methylene Blue Agar, Mac Conkey Agar, and Manitol Salt Agar. Gram staining was applied to distinguish gram-positive and gram-negative bacteria; and followed with the catalase, spore, motility, and biochemical tests to further identify bacteria. The bacteria identified were of the genus Enterobacter, Staphylococcus, Corynebacterium, Streptococcus, Escherichia, and Pasteurella. It could be concluded that non-specific bacteria of the same genus were identified in cervical mucus of dairy cows in their follicular and luteal phase, in a various of percentages.
