Abstract

Background : MFS is characterized by variable clinical manifestations mainly in cardiovascular, ocular, and skeletal systems. The major encoding gene of structural constituent of extracellular microfibrils is Fibrillin-1 (FBN1). Approximately 90% of MFS cases are caused by mutations in the FBN1 gene (15q21.1) and the other second is TGFBR2 (3p22) gene. Methods : The UMD database, Ensemble database and VUmc DNA Laboratory database of FBN1 mutations and polymorphisms were used to evaluate the DNA variations. For paternity testing, the PowerPlex system (Promega Corp. USA) was used. A 30-years old was being admitted to the hospital. CKMB and Troponin C serial. A CT angiography was performed and revealed a long type 1 aortic dissection until proximal of bifurcation, the arm span-height ratio is 1.10, high myopia, arachnodactily, positive thumb signs and wrist signs, joint laxity articulation genu, and history of spontaneous pneumothorax. Identified, his mother, two sisters and one brother are clinically MFS. Results : Genetic testing of FBN1 showed a substation at exon 15 of FBN1, c.1924G>T. Discussion: In missense mutations substituting or creating cysteine, the probability of ectopia lentis is significantly higher compared to other missense mutations. The EGF domains are interrupted by seven transforming growth factor (TGF)-binding protein domains characterized by 8 cysteine residues involved in intra-domain disulfide bonds. Conclusion : Untreated, life expectancy of patients with MFS is considerably reduced. Clinical care is complicated by variable age at onset and the wide range of severity of aortic features. Early recognition of affected status hopefully will lead to early prevention of complications that may follow.

Abstract

Latar Belakang: Skipping merupakan olahraga aerob yang telah dikenal sebagai permainan dalam masyarakat. Olahraga aerob jangka pendek dapat meningkatkan fungsi kognitif, salah satunya memori jangka pendek. Namun demikian, efek rutin atau jangka panjang dari olahraga ini terhadap memori jangka pendek belum pernah diteliti.Tujuan: Mengetahui pengaruh latihan skipping rutin terhadap memori jangka pendek kelompok dewasa muda.Metode: Penelitian quasi eksperimental dengan metode pre-test dan post-test unequivalent group. Subjek penelitian adalah mahasiswa Fakultas Kedokteran Universitas Diponegoro (n=40) berusia 18-22 tahun yang dipilih secara purposive sampling menjadi kelompok perlakuan dan kontrol. Penelitian ini menggunakan Scenery Picture Memory Test untuk pengambilan data memori jangka pendek pre-test dan post-test yang dilanjutkan analisis menggunakan uji t berpasangan, t tidak berpasangan, Wilcoxon dan Mann-Whitney.Hasil: Terdapat peningkatan bermakna pada nilai memori jangka pendek (p=0,000) setelah perlakuan  latihan skipping rutin selama 8 minggu, dengan rerata 18,00 ± 3,08 menjadi 21,60 ± 1,67. Peningkatan yang bermakna ini tidak ditemukan pada kelompok kontrol (p=0,189). Rerata selisih pre-test dan post-test nilai memori kelompok perlakuan lebih tinggi dibandingkan kelompok kontrol (p=0,008).Kesimpulan: Latihan skipping rutin dapat meningkatkan fungsi memori jangka pendek pada dewasa muda.

Abstract

Latar Belakang: Konsumsi minyak jelantah terus-menerus dapat menurunkan kadar antioksidan endogen didalam tubuh salah satunya enzim superoksida dismutase (SOD), sehingga menyebabkan stress oksidatif. Untuk mencegah terjadinya stress oksidatif diperlukan antioksidan yang dapat berasal dari alam, salah satunya adalah ekstrak kulit manggis (Garcinia Mangostana L.) yang diketahui memiliki efek antioksidan, melalui senyawa aktifnya yaitu xanthone.Tujuan : menganalisis pengaruh pemberian ekstrak kulit manggis terhadap kadar SOD tikus Wistar yang diinduksi minyak jelantah.Metode Penelitian ini merupakan penelitian eksperimental dengan desain post test only controlled group design. . Sampel adalah 24 ekor tikus wistar dibagi menjadi menjadi kelompok konrol satu (K1) diberi pakan standar, kelompok kontrol dua (K2) diberi minyak jelantah, kelompok P1 diberi ekstrak kulit manggis 400 mg/kgBB dan kelompok P2 diberi minyak jelantah dan ekstrak kulit manggis 400 mg/kgBB. Perlakuan dilakukan selama 28 hari dan dilanjutkan dengan pengukuran kadar enzim superoxide dismutase (SOD). Uji statistik menggunakan uji Anova .Hasil Uji One Way ANOVA kadar SOD menunjukkan tidak ada perbedaan yang signifikan antarkelompok dengan nilai p= 0,84 (p>0,05).Kesimpulan Tidak terdapat pengaruh yang signifikan dari pemberian ekstrak kulit manggis dalam meningkatkan kadar SOD plasma pada tikus yang diinduksi minyak jelantah.

Abstract

Latar Belakang : Hiperurisemia menyebabkan kerusakan membran sel seperti sel hepar dan ginjal akibat reaksi berantai peroksidase lipid. Kerusakan tersebut menimbulkan peningkatan kadar stress oksidatif. Tomat mengandung likopen dan flavonoid yang berperan sebagai antioksidan, sehingga dapat mengoptimalkan kinerja glutation (GSH) dalam tubuh. Tujuan : Mengetahui pengaruh ekstrak tomat (Lycopersicon esculelntum Mill.) terhadap kadar GSH darah tikus wistar hiperurisemia. Metode : Penelitian ini menggunakan jenis penelitian True Experimental dengan rancangan Post Test Only Control Group Design. Sampel sebanyak 24 ekor tikus wistar jantan yang memenuhi kriteria inklusi dan eksklusi, diadaptasi selama 7 hari, diberi pakan dan minum standar. Kelompok K1 tidak diberi perlakuan, Kelompok K2 diberi pakan dan minum standar, hati ayam, dan potasium oksonat. Kelompok P1 diberi pakan dan minum standar, hati ayam, potasium oksonat, dan ekstrak tomat 1 mg/200grBB/hari. Kelompok P2 diberi pakan dan minum standar, hati ayam, potasium oksonat, dan ekstrak tomat 1,5 mg/200grBB/hari. Ekstrak tomat diberikan mulai hari ke-21 sampai ke-35. Pada hari ke-36, sampel darah  diambil di retroorbita kemudian diterminasi. Plasma darah kemudian dianalisis untuk mengetahui kadar GSH. Hasil : Kadar GSH pada kelompok K1 = 154 ± 21,9 µg/ml; K2 = 228 ± 30,3 µg/ml; P1= 193 ± 21,5 µg/ml; dan P2= 176 ± 23,9 µg/ml. Simpulan : Pemberian ekstrak tomat (Lycopersicon esculentum Mill.) menurunkan kadar GSH darah tikus wistar hiperurisemia dan secara statistik didapatkan perbedaan yang tidak bermakna.Kata Kunci : ekstrak tomat (Lycopersicon esculentum Mill.), kadar GSH, hiperurisemia

Abstract

The particulate matter in vehicle emission has been shown to cause an increase in free radical levels which can increase serum MDA levels and leukocyte count. Spirulina platensis is an algae that has antioxidant effects that can scavenge free radicals. In this research, twenty-four of wistar mice are divided into 4 groups. Groups X1 and X2 received vehicle emission smoke exposure treatment and were given oral Spirulina platensis extract at a dose of 500 and 750 mg / kgBW / day for 14 days, the negative control group (C2) was given 0.9% NaCl orally and the normal control group did not get vehicle emission smoke exposure and Spirulina platensis extract. Serum MDA levels were checked by ELISA method and leukocyte count by giemsa staining. Data analysis was performed by using the one-way ANOVA test followed by Post Hoc Bonferroni. The results showed the mean serum MDA levels there was a significant difference between group C2 and group X2. In the mean absolute value of neutrophils and lymphocytes, there were significant differences between the C2 group with the administration of X1 and X2. In the mean absolute value of eosinophils and monocytes, there were significant differences between groups C1 and C2 and group X2 and between groups X1 and C1. Furthermore, the given doses of Spirulina platensis extract at a dose of 750mg / kgBW had an effect on serum MDA levels as well as the number of neutrophils and lymphocytes and the administration of doses of 500 and 750mg / kgBW affected the number of eosinophils and monocytes.
