Abstract

 ABSTRAK Protease alkaline and keratinase are a group of protease enzym which have important value in detergen industry and skin tannery. Brevibacillus agri A-03 is thermophilic bacteria isolate that comes from Ambayan Sumatera Barat hot spring and has the ability to produce protease and keratinase. The purpose of this research is to get protease alkaline and thermostable keratinase from Brevibacilus agri-A03. thermostable enzym is produced from enzym production enzym that contains kasein and keratin at various medium pH, inoculum incubation temperature and medium type. Enzym activity is measured by modified Walker methode, protein content is measured by Lowry methode. Protease alkaline is produced at exponential phase, maximum at 18th hours of incubation and keratinase is produced at stationer phase, maximum at 22nd hours. Both enzym is produced optimically at medium pH condition 9.0; incubation temperature 55°C, inoculum 5% by using modified Johnvesly and Naik medium with each protease and keratinase specific activity 1.927 and 1.047 U/mg  Keywords: Protease alkaline, Keratinase, Thermofilic, Brevibacillus agri A-03

Abstract

Endophytic bacteria live in plant tissues which utilized in plant protection against phytopathogens. This study aims to investigate the diversity of endophytic bacteria from the leaves of traditional medicinal plants that has anti-phytopathogens properties. Isolation of endophytic bacteria was done by spread plate method. The bacteria were characterised by Gram staining and the 16S rRNA gene analysis. Further screening of anti-phytopathogen activity used disc diffusion method for Ralstonia solanacearum, Xanthomonas campestris, Fusarium oxysporum, and Sclerotium rolfsii. All togethers, sixteen isolates of endophytic bacteria from the leaves of eight medicinal plants species were obtained. Fourteen isolates had an anti-phytopathogen (with eight isolates against R. solanacearum, seven isolates against X. campestris, nine isolates against F. oxysporum, and five isolates against S. rolfsii). From the 14 isolates identified, phylum Firmicutes were dominant (64.3%), followed by Proteobacteria (28.6%), and Actinobacteria (7.1%). Phylum Firmicutes consists of Bacillus indicus (BJF1, TCF1, and MCF2), Bacillus pumilus (CAF4), Bacillus sp. (CAF1), Bacillus subtilis (AAF2, MCF1, CAF3, and MCF3); phylum Proteobacteria consists of Pantoea agglomerans (CAF2), Pantoea stewartii (AAF4), Pseudomonas oryzihabitans (AAF3), and Pseudomonas psychrotolerans (AAF1); and phylum Actinobacteria consists of Kocuria kristinae (CSF1).

Abstract

Fish cultivators generally experience difficulties providing quality artificial feed (pellets) due to a lack of knowledge and skills in processing technology and making alternative feeds. Besides that, the costs incurred for feed procurement are relatively large, reaching 70 - 80% of the total production costs. However, the provision of feed is often an obstacle because, in addition to the increasingly high price, the quality of available feed does not always match the nutritional needs needed by Fish. The provision of quality feed for intensively cultivated tilapia dramatically affects the resulting growth and productivity. The higher the quality of the feed, the production will also increase. One of the technologies that can be applied to overcome this problem is the manufacture of alternative formulated diets using local raw materials. The method used in this service activity is the transfer of knowledge presented in the socialization activities in the form of 1) introduction to the types of alternative fish feed ingredients sourced from the area, (2) preparation of artificial tilapia feed formulations, (3) manufacturing technology artificial feed (pellets) and 4) monitoring and assistance. The socialization was carried out in the form of a lecture and was followed by a question and answer session. The results of farmer activities recognize the types of local feed that can be used as fish feed, know the preparation of feed formulations, and alternative artificial feed manufacturing technologies. The knowledge and skills possessed by fish cultivators are expected to overcome the difficulties of fish farmers in providing feed so that fish production and cultivation business increase.

Abstract

Thermophilic bacteria are bacteria that can produce thermostable enzymes and used in various industries. Thermostable enzymes that can be produced by thermophilic microorganisms one of them is xylanase enzyme. Xylanase produced by microbes has optimum temperature characteristics and more diverse optimum pH on various substrates, which will affect the activity of the resulting xylanase enzyme. The influence of temperature greatly determines the activity of the enzyme at the time of catalyzing a reaction. At optimum pH conditions, the enzyme has an active side conformation that is substrate-like so that it can form a complex of appropriate enzymes and produce the product to its full potential. One of the thermophilic bacterial habitats of hot springs in West Sumatera is the Aro Sapan River hot spring located in Koto Parik Gadang Subdistrict in Ateh, South Solok District. The Saw Aro River hot spring has a temperature of 75 ° C and is pH 8 or alkaline. The aim to this research was to know the profile of the growth of thermophilic bacteria. This research is a descriptive method. The data obtained were analyzed descriptively. The conclusion is the best activity of xylanase thermophilic bacteria was at the sixth time incubation.

Abstract

Endophytic bacterium are important to produce antibiotic. We have experimentally studied zodia plant (Evodia suaveolens Scheff) in attempting to isolate and characterize the bacterium. We have also examined potentiality of each isolate to produce antibiotic. We obtained three a isolate of  endophytic bacterium that produce antibiotic compounds. Among three isolates, two could form high inhibition zone. MZS-01 formed 10.4 mm inhibition zone against Eschericia coli, while MZS-03 formed 6.96 mm inhibition zone against Staphylococcus aureus. We described the isolates MZS-01 and MZS-03 based on macroscopis and microscopis characteristic of colony  culture.

Abstract

A research on bacteriological condition of ground well water at Air Dingin, Padang was conducted from January to March 2013 at the Health Laboratory Agency (UPTD Balai Laboratorium Kesehatan) of West Sumatra. The water samples were collected by using purposive method at three sampling points in distances of 250 m, 350 m and 450 m from garbage disposal locations. Bacteriological test was done using Most Probable Number method (MPN) with combination of 3:3:3. Escherichia coli was described using indole, methyl red, voges proskauer, citrate (IMViC test). The result showed that MPN of Escherichia coli of ground well water for each distance of 250 m, 350 m and 450 m ranges from 0 cell/100 ml to 2400 cell/100 ml. Based on bacteriological parameter test, quality of the ground well water around the landfill area was not appropriate to be consumed by people, except for ground well water was located at 450 m from landfill.Key words: ground well water, Escherichia coli, landfill, qualilty.

Abstract

Antibacterial activity of crude extracts of pisang kepok kuning plants (Musa paradisiaca Linn.) were tested against Staphylococcus aureus and Escherichia coli. Roots, corms, pseudoterms, flowers, and ripe fruits of pisang kepok kuning cultivar were macerated with ethanol 96% and evaporated in a rotary evaporator at 50C. All of crude extracts were screened for its antibacterial activity by using Agar Well Diffusion Method with concentration based on 200 grams of fresh weight. This research used Completed Randomized Design (CRD) with nested pattern, four replications, and consist of two factors. First factor was bacteria and the second factor was crude extracts of pisang kepok kuning such as roots, corms, pseudoterms, flowers, and ripe fruits. The results showed that all of crude extracts have significant antibacterial activity against S. aureus and E. coli. Crude extract of the corm has highest antibacterial activity against S. aureus with diameter of inhibition area was 20,391 mm with inhibition zone type isirradical and diameter inhibition area 18,962 mm for E. coli that has radical zone.Keywords : Musa paradisiaca Linn., S. aureus, E. coli, antibacterial

Abstract

A study on the characterization of thermophilic bacteria in producing neutral protease enzyme has been conducted in the Laboratory of Microbiology, Biology Department, Andalas University from October 2013 to March 2014. The bacteria were isolated from a hot spring at Sungai Medang, Kerinci. Water samples  were collected purposively based on gradient temperature at the hot spring which ranged from 450C to 880C. Colony of the bacteria were inoculated using nutrient agar (NA). We obtained 10 isolates of neutral protease enzyme from the thermophilic bacteria with proteolytic index ranged from 0.27 to 3.89. The MIV-14 isolate has the highest proteolytic index which collected from 830C of the hot spring. Characterization of macroscopic, microscopic and biochemical tests were reported. Generally, the colonies have round shape, wavy edge, white, producing endospores and motile. Keywords: characterization, neutral protease, thermophilic bacteria.

Abstract

Research about partial optimization of thermophilic bacteria isolate M5-24 on protease production was conducted from May 2013 to December 2013 in Laboratory of Microbiology, Biology Department, Andalas University, Padang. Purpose of the research was to find out optimum condition of protease production by isolate M5-24. This study consisted of three steps. Step 1: Investigating the effect of temperature and pH used completely randomized design in factorial with two replications.The first factor was temperature (450C, 500C, 550C, 600C), the second factor was pH (7, 8, 9, 10). Step 2: Investigating the effect of inoculum doses and inducers used completely randomized design in factorial with three replications. The first factor was inoculum doses (1%, 5%, 10%) and the second factor was inducer (yeast extract 0,5%; pepton 0,1%;  kasein 1%; meat extract 0,5%). While Step 3: Investigating the effect of agitation (125 rpm, 150 rpm, 175 rpm, 200 rpm) used completely randomized design with six replications. The result showed that the best duration of growing was at 18th hours after incubation, the highest enzyme activity was at temperature 500 C;  medium’s pH at 8; the best of inoculum dose was at 5%; the best  inducer was casein 1% and optimum agitation was at 175 rpm. Extrinsic optimization can increase protease production of  thermophilic isolate M5-24 up to 1.7 times.Keywords : Optimization, Protease,  thermophilic, bacteria

Abstract

A study about the effect of fresh extract of Curcuma spp on the growth of Candida albicans, Staphylococcus aureus and Escherichia coli was conducted from March to July 2012 at Microbiology Laboratory, Faculty of Sciences Andalas University. This experiment used nested completely randommized design. The result showed that fresh extracts of Curcuma spp. have different growth inhibition effects on C. albicans, S. aureus dan E. Coli. Temulawak (Curcuma xhantorrhiza) has the highest inhibition effect to the growth of C. albicans (13.07 mm), S. aureus (15.75 mm) and E. coli (31.56 mm). The lowest bactericidal consentration of fresh extract rhizome of temulawak for E. coli were 12.5% and 25%, but there is no inhibition effect to the C. albicans and S. aureus.Keywords: Curcuma, antimicrobial, extract, pathogen microbial
