Abstract

Kappa casein (ĸ-cn) gene as one of the four casein genes commonly has two variant alleles. B allele is positively correlated with milk proteins, so potentially be used as MAS to improve milk protein content. This study aimed to identify genetic variation of the ĸ-casein gene in Holstein-Friesian (HF) from several dairy regions in West Java. Blood samples were taken through jugular vein of lactating heifers and cows, from intensive managements under the Local Government Dairy Breeding Station - Cikole (BBPT Cikole-SP) (82 hds), Embryo Transfer Station (BET) (50 hds), and Eco Farm of Animal Science Faculty of Bogor Agricultural University (EcoFarm) (20 hds); as well as from semi-intensive managements of small dairy farmers under the supervision of the North Lembang Cooperative Unit (KPSBU Lembang) in two villages of Cilumber (98 hds) and Pasar Kemis (92 hds).  Blood samples were also taken via cocsigalis vein of AI (active and non active) HF bulls at the two National AI Stations in Lembang (25 hds) and Singosari (32 hds).  Identification of genetic variation was by applying PCR-RFLP method. Three genotypes were identified, namely AA, AB, and BB, resulting two alleles of A and B. Results entirely showed that the frequencies of the BB HF female were very low, with the range of 0%-6% (vs AA genotype= 10%-54% and AB genotype= 46%-85%), despite of the relatively high frequency of the B allele over the A allele (23%-48% vs. 52%-77%). The low frequency of the observed BB females might be due to the limited AI active BB bulls used for services by the two national AI stations (0%-4%).

Abstract

Pit-1 gene has been identified as the pituitary specific transcription factor that regulates the expression of the growth hormone (GH) and prolactin (PRL) genes in the anterior pituitary. The use of polymorphic markers in breeding programmes could make selection more accurate and efficient. A total  of 320 Indonesian buffaloes from four populations and 45 FH cows from nine populations were genotyped for polymorphism of Pit1|Hinf1 gene by the polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) methods. The gene polymorphism was detected only in FH cattle. The frequency of AA, AB, and BB genotype was 2%, 44%, and 53% respectively. The frequency of A allel was 25% and B allel was 75%, but no polymorphism was detected in 320 Indonesia buffaloes.

Abstract

The research objectives were to characterize morphometric and genetic distance between populations of Indonesian local goats. The morphological discriminant and canonical analysis were carried out to estimate the phylogenic relationship and determine the discriminant variable between Benggala goats (n= 96), Marica (n= 60), Jawarandu (n= 94), (Kacang (n= 217), Muara (n= 30) and Samosir (n= 42). Discriminant analysis used to clasify body weight and body measurements. In the analysis of variance showed that body weight and body measurement (body length, height at withers, thorax width, thorax height, hert girth, skull width and height, tail length and width, ear length and width) of Muara goats was higher (P<0.05) compared to the other groups, and the lowest was in Marica goats. The smallest genetic distance was between Marica and Samosir (11.207) and the highest were between Muara and Benggala (255.110). The highest similarity between individual within population was found in Kacang (99.28%) and the lowest in Samosir (82.50%). The canonical analysis showed high correlation on canon circumference, body weight, skull width, skull height, and tail width variables so these six variables can be used as distinguishing variables among population. The result from Mahalonobis distance for phenogram tree and canonical analysis showed that six populations of Indonesian local goats were divided into six breed of goats: the first was Muara, the second was Jawarandu, the third was Kacang, the fourth was Benggala, the fifth was Samosir and the sixth was Marica goats. The diversity of body size and body weight of goats was observed quite large, so the chances of increasing productivity could be made through selection and mating programs.

Abstract

The aim of this research was to assess the use of silkworm pupae (Bombyx mori) powder extract on the blood cells and hematological variables of quails (Coturnix coturnix japonica). The treatments consisted of R0= isoenergy and isoprotein ration without pupae powder extract, R1= isoenergy and isoprotein ration + 1% of pupae powder extract, R2= isoenergy and isoprotein ration + 10% of pupae powder extract, R3= isoenergy and isoprotein ration + 1% of residue of pupae powder extract, R4= isoenergy and isoprotein ration + 10% of residue of pupae powder extract, R5= isoenergy and isoprotein ration + 1% of pupae powder, and R6= isoenergy and isoprotein ration + 10% of pupae powder. The variables measured were blood cell and hematological profiles including the number of leukocytes, erythrocytes, and leukocytes differentiation. Immune response was observed at 12, 24, and 48 h by counting the white blood cells type. The results showed that feed consumption ranged 79.19-154.70 g/quail/wk and the body weight was 71.45-149 g/quail. The addition of pupae extract at a dose of 10% in the diet significantly (P<0.05) increased the number of quail white blood cells. The addition of pupae extract, pupae powder residue, and pupae powder in the ration did not significantly affect the number of quail erythrocytes, hemoglobin, and PCV (packed cell volume). Mean number of white blood cells increased during 24 h after treatment of 10% pupae powder extract (R2) and decreased at the 48th h. It is concluded that the value of leukocytes, lymphocytes, monocytes, and heterophile increased in quails supplemented with 10% pupae powder extract (R2).

Abstract

Kacang goat is one of Indonesian local goat which has not been optimized in exploration. Kacang goat has potency as a dairy goat. Milk and colostrum from this type of goat is one of lactoferrin sources which has various benefit, such as antimicrobial activity. The milk as a lactoferrin source is expected to be a solution for bacterial 9astrointestinal infection cases which is a major problem in Indonesia. This research described the identification of lactoferrin from milk and colostrum of kacang goat by single radial immunodiffusion (SRID) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). SRID is based on the diffusion of whey protein from a circular well into a homogeneous gel containing anti-lactoferrin. SDS-PAGE was performed in 7.5%  polyacrylamide gel. Both methods were able to identify lactoferrin in colostrum and milk from the sample, but SRID showed low sensitivity toward low concentration of lactoferrin in both colostrums and milk. The estimation of lactoferrin molecular weight by relative mobility of protein from the bands that perform of colostrum and milk of kacang goat is approximately 74,100 Dalton. Based on the ring diameter of the precipitin, the lactoferin level in colostrum and milk increased until 48 hours after postpartum and subsequently decreased. Abstract

The objectives of this study were to identify polymorphisms in ovine K-casein gene exon 4 and its association with milk yield, percentage of protein and milk fat in sheep under extcnsif management at JASTRU (Jonggol Animal Studies Teaching and Research Unit) farm, Fact. of Anim Sci, Bogor Agric. University. The 83 blood sample from ewes which have record of milk production were used to determined polymorphism by using PCR-SSCP (Polymerase Chain Reaction-Single Strand Conformation Polymorphism) methode. The research activities were carried out through blood collection, DNA isolation, and fragment DNA amplification by PCR and separation in electrophoresis with silver staining. Electrophoretic pattern (PCR-SSCP) revealed th rce types of K-casein gene were dcsignitcd A, Band C type. The highest proportion was A type 69.57% and followed hy B 28.99% and the lowest C 1.45%. Abstract

Madura cattle is one of the Indonesian local cattle breeds derived from crossing between Zebu cattle (Bos indicus) and banteng (Bos javanicus). Branched-chain α-ketoacid dehydrogenase (BCKDH) is one of the main enzyme complexes in the inner mitochondrial membrane that metabolizes branched chain amino acid (BCAA), ie valine, leucine, and isoleucine. The diversity of the nucleotide sequences of the genes largely determine the efficiency of enzyme encoded. This paper aimed to determine the nucleotide variation contained in section  intron 7, exon 8, and intron 8 genes BCKDHA on Madura cattle. This study was conducted on three Madura cattle that used as bull race (karapan), beauty contest (sonok), and beef cattle. The analysis showed that the variation in intron higher than occurred in the exon. Simultaneous indel found at base position 34 and 68 in sonok cattle. In addition, the C266T variant found in beef cattle. These variants do not cause significant changes in amino acids. There was no specific mutation in intron 7, exon 8, and intron 8 were found in Madura cattle designation. This indicated the absence of differentiation Madura cattle designation of selection pressure of BCKDHA gene.

Abstract

Giving a half-dose of an anthelmintic is a simple method for detecting resistance in parasites infesting small ruminants. When a single anthelmintic fails in native sheep from Indonesia, a combination of anthelmintics from different chemical classes with different mechanism of action is an alternative parasite-control strategy. This study compared the anthelmintic effectiveness of ivermectin (IVM) and albendazole (ABZ) have given either separately as a full-dose or half-dose or co-administered to sheep naturally infected with Ostertagia. Twelve sheep from Bogor, Indonesia were divided into six groups based on the anthelmintic treatment: half-dose IVM, full-dose IVM, half-dose ABZ, full-dose ABZ, combined (half-dose IVM + half-dose ABZ), and control groups. The treatment effectiveness was determined using the Faecal egg count reduction (FECR) on before and after 7, 14, 21, 28, 35, and 42 days of treatment. FECR of half-dose IVM, full-dose IVM, half-dose ABZ, full-dose ABZ, and both combined ranged from 78-100%, 96-100%, -71-89%, -521-64%, and 54-100%, respectively. The results showed that full-dose IVM was the most effective to against Ostertagia until 42 days and followed the combined that effective until 28 days. Half-dose and full-dose ABZ weren’t effective to against Ostertagia.
