Abstract

The technology of fish germ cell transplantation had been established to create broodstock systems by which a target offspring can be produced from a surrogate parent. Donor cell for transplantation is derived from transgenic fish carrying green fluorescent protein gene functions as a marker to distinguish the donor from recipient cell. In this study, an alternative technique was developed for identifying gouramy-derived donor cell and Nile tilapia as recipient by PCR amplification method using growth hormone (GH) gene as a molecular marker.  Specific primer for GH gouramy was designed by using Genetyx version 7 software.  ß-actin gene was used as an internal control of DNA loading. The result showed that a specific PCR amplification product of 340 in length was obtained when the DNA template was from gouramy, while no PCR product from Nile tilapia. The minimum concentration of genomic DNA of gouramy mixed with a 700 ng/µl of Nile tilapia that could be detected by PCR was 1 ng/µl. Thus, PCR method with specific GH primer may be useful to detect the incorporation of donor cell in recipient gonad towards development of surrogate broodstock technology.

Abstract

Molecular marker is useful in the development of testicular cells transplantation for detecting donor-derived germ cells in the recipient gonad. In this study, a giant gourami (Osphronemus goramy) vasa-like gene (GgVLG) was cloned and characterized for use as a molecular marker for germ cells in this species. Nucleotide sequence analysis revealed that GgVLG comprises 2,340 bps with an open reading frame of 1,962 bps encoding 653 amino acids. The deduced amino acid sequence contained 17 arginine-glycine or arginine-glycine-glycine motifs and eight conserved motifs belonging to the DEAD-box protein family. The GgVLG sequence showed high similarity to Drosophila vasa, common carp vasa homolog and tilapia vasa homolog for 66.2, 85.9, and 90.7%, respectively. In adult tissues, the GgVLG transcripts were specifically detected in ovary and testis. In situ hybridization analysis showed that GgVLG mRNA was detected in oocytes of the ovary and spermatogonia of the testis. There was no signal detected in the spermatocytes, spermatids and other gonadal somatic cells. Thus, consensus sequences, specific localization of GgVLG mRNA in the germ cells, amino acid sequence similarity and phylogenic analysis all suggest that GgVLG is the giant gourami vasa-like gene. Further, GgVLG can be used as a molecular marker for giant gourami germ cells.

Abstract

Testicular cell transplantation technology can be used in fish seed production engineering. In this study, optimization of transplantation using donor cells from young gouramy and triploid tilapia (3N) as recipient. Triploid tilapia is produced using heat shock method. The testes of male gouramy (body weight of 400- 850 g) was dissociated using 0.5% trypsin. Dissociated testicular cells was injected into the peritoneal cavity of tilapia larvae. Analysis of donor cell colonization was carried out using PCR method with DNA template that had been extracted from the gonad of 2-month-old tilapia. PCR was performed using specific primers for the growth hormone gene and (3-actin as an internal control of DNA loading. The results of nucleoli preparation showed that the success of triploidyzation was 88.5%. The gonad size of diploid (2N) and 3N recipient were relatively similar, while in not transplanted 3N tilapia was rudimentary. PCR results showed that the transplanted 3N tilapia has a DNA band of the same size with gouramy, while in control was not. This indicated that donor cells have been colonized in the gonads of recipient. The donor cell colonization in recipient 3N (78%) was higher than that of 2N (50%). Further research is required to determine the ability of donor cells differentiate into sperm and eggs in recipient gonad.

Abstract

ABSTRACTExperiment was performed to assess the effect of 17a-methyltestosterone (MT) treatment on Congo tetra fish larvae.  To evaluate the optimal pattern of MT treatment, three different treatments were administrated.  Three months old larvae were submerged in three different doses of MT; 1, 2 and 4 mg/l.  These studies showed that the highest percentage of male fish was obtained by 4 mg/l MT treatment, 87,17%.  The 2 mg/l and 1 mg/l MT treatments obtained 77,53% and 69,86% male respectively, two times higher than control, 38,96%.  On the other hand, the 4 mg/l MT treatment also resulted the highest percentage of hermaphrodite fishes, 17,58%.  The highest survival rate was shown by 1 mg/l MT treatment, 62,77% and the lowest was shown by the 4 mg/l MT treatment, 47,20%.  The highest rate of fish length and weight was shown by the 4 mg/l MT treatment, 4,4 cm and 1,65 gram respectively.  These findings suggest that MT treatment offers an advantage in growth of  tetra Congo larvae. Key word :  Sex reversal, methyltestosterone, Congo tetra fish, Micraleptus intterruptus. ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh perendaman larva di dalam larutan hormon 17a-metiltestosteron pada dosis 1, 2 dan 4 mg/l larutan.  Persentase tertinggi ikan jantan dihasilkan  oleh perlakuan 4 mg/l, yaitu 87,17%.  Perlakuan 2 mg/l dan 1 mg/l masing-masing menghasilkan 77,53% dan 69,86% sedangkan kontrol menghasilkan 38,96% jantan.  Efek lain dari perlakuan MT ini adalah hermafroditisme.  Perlakuan 4 mg/l menghasilkan persentase hermafrodit tertinggi yaitu 17,58%, sedangkan pada kontrol kelangsungan hidup tertinggi diperoleh pada perlakuan 1 mg/l (62,77%) dan terendah pada perlakuan 4 mg/l (47,20%).  Hasil tersebut menunjukkan adanya pengaruh dosis hormon terhadap kelangsungan hidup ikan.  Pengukuran bobot dan panjang ikan pada setiap perlakuan menunjukkan nilai tertinggi dihasilkan oleh perlakuan 4 mg/l  yaitu 1,65 gram dan 4,40 cm.  Hal ini diduga bahwa hormon MT merangsang pula pertumbuhan ikan.Kata kunci :  Pergantian kelamin, metiltestosteron, ikan tetra Kongo, Micraleptus intterruptus.

Abstract

ABSTRACTProblems faced in supplying seed out of Java especially Bogor to Sumatera, for instance, are it only contains few seed (100 seeds/l) and it has a high mortality rate because of stress during transport.  This experiment was conducted to know the effect of MS 222 in keeping fish metabolism activity down while taking note of its survival rate and change of water quality. The experimental design used here was factorial design 3x5 with 3 repetitions.  Dosages of MS–222 were 0, 25 and 50 ppm and the fish densities were 100, 300, 400, 500 and 600 per liter.  As container was plastic bag (volume 10 l).  After putting the fish into the plastic bag, it was filled up with oxygen, which the volume was 3 times than water volume, than the plastic bag was closed by tying it.  After 18 hours of treatment the survival rate and water quality were checked.  Based on evaluation of survival rate, it was concluded that the combination between 25 ppm of MS 222 and fish density, which was 500 per liter, gave an optimum result.Keywords: Catfish, Pangasius sutchi, live transport, stress, MS–222, survival rate ABSTRAKKendala yang dihadapi dalam pemasokan benih ikan patin dari pulau Jawa terutama Bogor, keluar pulau Jawa terutama Sumatera adalah jumlah ikan yang terangkut masih sedikit (± 100 ekor/l) dan tingkat kematian yang cukup tinggi, karena stres selama perjalanan. Penelitian ini dilakukan untuk mengetahui pengaruh penggunaan MS–222 dalam menekan aktivitas metabolisme ikan dengan memperhatikan tingkat kelangsungan ikan dan perubahan kualitas air. Penelitian ini menggunakan rancangan faktorial 3x5 dengan 3 ulangan. Dosis MS–222 yang digunakan: 0,25 dan 50 ppm dan kepadatan ikan: 100, 300, 400, 500 dan 600 ekor/l. Wadah pengangkutan berupa kantong plastik (volume 10 l). Ikan yang diangkut dimasukkan ke dalam kantong plastik, kemudian dipompakan oksigen sebanyak 3 kali volume air, selanjutnya dilakukan pengikatan. Setelah 18 jam perlakuan dilakukan perhitungan terhadap tingkat kelangsungan hidup ikan dan perubahan kualitas air media. Hasil penelitian ini menunjukkan kombinasi penggunaan MS–222 25 ppm dan kepadatan ikan 500 ekor/l memberikan tingkat kelangsungan hidup yang optimal.Kata kunci: Ikan patin, Pangasius sutchi, pengangkutan hidup, stres, MS–222, tingkat kelangsungan hidup.

Abstract

Growth of giant gourami is relatively slow. Administration of recombinant growth hormone (rGH) by severaltimes immersion in water containing 0.9% NaCl for one hour can significantly increase the growth. In thisstudy, giant gourami juvenile were immersed in freshwater containing 120 mg/L rGH to determine the longimmersion time that provides higher growth and survival. Juvenile at seventh-day-old after egg yolk absorbedwas immersed for 0.5, 1.0, 2.0, and 3.0 hours in rGH solution. As controls, fish was immersed in freshwaterwithout rGH (control 1: C1), and water containing 0.9% NaCl, and 120 mg/L rGH (control 2: C2). Fish waskept under controlled conditions in 20 L aquarium for three weeks, then in 50 L aquarium until at end ofexperiment, and fed at satiation. The results showed that highest growth in body weight, survival, and biomass(p<0.05) were obtained in rGH immersion treatment for 0.5 hour. Biomass of fish immersed by rGH for 0.5hour was about 32% higher compared to control C1, and 13% compared to C2. Survival of the 0.5 hour rGHimmersedfish (100%) was similar to control C1 and C2. Thus, immersion of giant gourami juvenile infreshwater containing rGH for 0.5 hour allowed highest growth rate, and this method is relatively easy to beadopted by fish hatchery.Keywords: immersion, hormone, growth, freshwater, giant gourami

Abstract

ABSTRACT This research was conducted to examine the different dose of vitamin E in the diet on female broodstock reproduction performance of the fantail goldfish Carassius auratus auratus. This research consisted of four treatments with three replications. The use of vitamin E doses was 0, 125, 250, and 375 mg/kg. The vitamin E was dissolved in vegetable oil and mixed with albumen as a binder in feed. The vitamin E was sprayed at feed and was air dried. Female broodstock of the fantail goldfishes were reared for 40 days. The result showed that 375 mg/kg treatment performed the highest quality of reproduction. Egg diameter, gonadosomatic index, fecundity, and germinal vesicle breakdown of fish which are treated by 375 mg/kg vitamin E were respectively 0.92±0.05 mm, 8.86±4.62%, 56.00±29.18%, and 67.35±17.67% higher than control. Thus, 375 mg/kg of dietary vitamin E suplementating was a best dose to improve female broodstock productivity of the fantail goldfish Keywords: female broodstock, fantail goldfish, vitamin E, reproduction quality  ABSTRAK Penelitian dilakukan untuk menguji suplementasi vitamin E dengan dosis berbeda dicampur ke dalam pakan komersial terhadap produktivitas induk betina ikan komet Carassius auratus auratus. Penelitian ini menggunakan empat perlakuan dengan tiga ulangan. Dosis vitamin E yang diberikan, yaitu 0, 125, 250, dan 375 mg/kg pakan. Vitamin E dilarutkan dalam minyak nabati dan dicampur dengan putih telur sebagai perekat pada pakan. Vitamin E disemprotkan ke pakan dan dikeringanginkan. Induk betina ikan komet pascasalin dengan bobot 72,78±19,47 g diberi perlakuan selama 40 hari. Hasil penelitian menunjukkan bahwa induk betina ikan komet yang diberi suplementasi vitamin E sebanyak 375 mg/kg dalam pakan memiliki diameter telur 0,92±0,05 mm, gonadosomatic index 8,86±4,62%, fekunditas 56,00±29,18 butir/g ikan, dan germinal vesicle breakdown 67,35±17,67% yang lebih tinggi dibandingkan kontrol. Dengan demikian, suplementasi vitamin E sebesar 375 mg/kg pada pakan adalah dosis terbaik dalam meningkatkan produktivitas induk betina ikan komet. Kata kunci: induk betina, ikan komet, vitamin E, kualitas reproduksi

Abstract

ABSTRACT This study aimed to examine the effect of extract of purwoceng Pimpinella alpina for masculinization of Betta splendens. This research used completely randomized design with three treatments that were distinguished by doses of purwoceng extract, which were 10, 20, and 30 µL/L, and 0 µL/L as control. The treatments were given by immersion to 35 embryos at eye spots phase or about 28-hours after fertilization, for eight hours. The immersion process was done for 8 hours. The result showed that 20 µL/L dose of purwoceng extract produced 62.66% male, and hatching rate was 85.71%. This male percentage was higher compared to control (45.91%), but the hatching rate was lower than that of control (98.57%). At the higher dose (30 µL/L), male fish population and hatching rate reduced respectively 39.72% and 68.57%, respectively. Keywords: masculinization, embryo immersion, purwoceng extract, Betta splendens  ABSTRAK Penelitian ini bertujuan untuk mengevaluasi pengaruh dosis ekstrak purwoceng Pimpinella alpina extract dalam maskulinisasi ikan cupang hias Betta splendens. Penelitian ini menggunakan metode rancangan acak lengkap dengan tiga perlakuan dan tiga ulangan dosis ekstrak purwoceng. yaitu 10, 20, dan 30 µL/L, serta perlakuan 0 µL/L sebagai kontrol. Perlakuan diberikan melalui perendaman selama delapan jam terhadap 35 embrio saat memasuki fase bintik mata atau sekitar jam ke-28 setelah pembuahan. Hasil penelitian menunjukkan bahwa dosis ekstrak purwoceng 20 µL/L menghasilkan persentase ikan jantan sebesar 62,66%, dan daya tetas telur sebesar 85,71%. Persentase jantan tersebut lebih tinggi daripada kontrol (45,1%), tetapi derajat penetasannya lebih rendah daripada kontrol (98,57%). Pada dosis yang lebih tinggi (30 µL/L), populasi ikan jantan (39,72%), dan derajat penetasan (68,57%) adalah menurun. Kata kunci: maskulinisasi, perendaman embrio, ekstrak purwoceng, Betta splendens

Abstract

ABSTRACT The aim of this study was to evaluate the effect of electric field exposure duration at the voltage of 10 volt on goldfish Carassius auratus auratus gonadal development maintained in 3 ppt salinity media. The experiment consisted of four treatments in triplicates i.e. control, two, four, and six minutes of electrical-field exposure. The experiment design used was completely randomized design. Fish used was female goldfish at the density of 4 fish/aquarium with an average total length of 12.27±0.05 cm and average body weight of 22.29±0.54 g. Result of study showed that the electrical-field exposure at 10 volt for all duration treatments in 3 ppt of media salinity did not give significant effect on gonadosomatic index (GSI) and gonadal development of goldfish. Keywords: electrical field, Carassius auratus auratus, gonad, salinity  ABSTRAK Tujuan penelitian ini adalah untuk mengevaluasi pengaruh lama waktu pemaparan medan listrik dengan voltase 10 volt terhadap perkembangan gonad ikan komet Carassius auratus auratus yang dipelihara pada media bersalinitas 3 ppt. Perlakuan pada penelitian ini terdiri atas empat perlakuan, yaitu: perlakuan kontrol, dua, empat, dan enam menit. Rancangan percobaan yang digunakan adalah rancangan acak lengkap dengan empat perlakuan dan tiga ulangan. Ikan uji yang digunakan adalah ikan komet betina. Jumlah ikan yang digunakan adalah 4 ekor/akuarium dengan panjang total rata-rata 12,27±0,05 cm dan bobot tubuh rata-rata 22,29±0,54 g. Hasil penelitian menunjukkan bahwa pemberian medan listrik sebesar 10 volt dengan lama waktu pemaparan medan listrik pada semua perlakuan durasi di media pemeliharaan bersalinitas 3 ppt tidak memberikan pengaruh yang nyata terhadap parameter indeks gonadosomatik (GSI) dan perkembangan gonad ikan komet. Kata kunci: medan listrik, Carassius auratus auratus, gonad, salinitas
