Abstract

Protease (enzim proteolitik) merupakan enzim yang mampu menguraikan atau memecah molekul protein. Salah satu kelompok mikroorganisme yang mampu menghasilkan protease, yaitu aktinobakteri. Penelitian ini bertujuan untuk mengisolasi dan menguji aktitivas enzim proteolitik (protease) dari aktinobakteri isolat lokal (AKJ-09) Aceh. Penelitian ini dilaksanakan dengan melakukan peremajaan isolat, penapisan isolat yang berpotensi sebagai penghasil protease, penentuan kurva produksi protease, dan pengukuran aktivitas protease dan kadar protein. Isolat aktinobakteri AKJ-09 diseleksi untuk diisolasi dan diuji aktivitas proteolitiknya karena menunjukkan aktivitas proteolitik tertinggi dengan Indeks proteolitik (IP = 19,93) di antara kedelapan isolat aktinobakteri lainnya yang secara kualitatif menunjukkan aktivitas proteolitik pada media NAS (Nutrient Agar + Susu Skim). Berdasarkan uji aktivitas proteolitiknya, isolat AKJ-09 menunjukkan aktivitas protease tertinggi pada hari kedelapan sebesar 0,088 U/ml dengan kadar aktivitas spesifik mencapai 7,56 U/mg.

Abstract

Substrate of straw mushroom in general are straw. Spent coffee grounds are one of the wastes that can be used as an alternative substrate for straw mushrooms. The purpose of the research are to know the productivity effect of straw mushrooms on spent coffee grounds substrate, as well as to know the optimal comparison of substrate combinations for productivity of straw mushrooms. This research is experimental with a quantitative approach. The design used was a Nonfactorial Completely Randomized Design consisting of 5 treatments and 5 replications. The treatments were 100% straw (P0) as positive control, 75% straw and 25% spent coffee grounds (P1), 50% straw and 50% spent coffee grounds (P2), 25% straw and 75% spent coffee grounds (P3), and 100% spent coffee grounds (P4). Data were analyzed by Analysis of Variants (ANAVA). The results of this research indicate the substrate combination of straw and spent coffee grounds do not affect to productivity (amount) of straw mushrooms. The optimal substrate combination of straw mushrooms are P3 treatment with a combination of 25% straw and 75% spent coffee grounds.

Abstract

BKL-1 and BKU-31 bacteria isolates were collected from digestive tract of kerapu lumpur (Epinephelus tauvina) and identified as Serratia marcescens biogp 1 dan Serratia marcescens respectively. BKL-1 crude protease were obtained after 27 hours incubation period in Nutrient Broth (NB) medium added with peptone 0.5%, NaCl 0.5%, lactose 0.1%, and CaCl2 0.1%. BKU-31 crude proteases were obtained in NB medium added with 0.1% skimmed milk (NBS) for 18 hours incubation period. The highest activity of proteases of S. marcescens biogp 1 BKL-1 reached after 27 hours of incubation, namely 0.002 U/ml with protein concentration 0.064 mg/ml. The proteases of S. marcescens BKU-31, on the other hand, reached their highest activity (0.064 U/ml) at 24 hours of incubation with protein concentration 0.056 mg/ml.The optimum temperature crude BKL-1 proteases were 30°C, whereas BKU-31 proteasess had the optimum temperature of 50 ºC.

Abstract

ABSTRACTMushroom cultivation has bright prospects because the natural and environmental conditions in Indonesia are very supportive. Straw is a planting medium that is commonly used for the growth of edible mushrooms. Cardboard, klaras and husks are one of the household wastes that can be used as an alternative planting medium for mushroom as a substitute for straw. This study seeks to find out the effect of a combination of cardboard. Klaras and rice husks as a growing medium for mushroom and to find out the comparison of the composition of cardboard waste and klaras growing media that can have the best effect on the growth of edible mushrooms. This research was conducted in Rukoh Village, Banda Aceh from October to November 2019 for ± months. This study used a quantitative method in the form of an experiment with a completely randomized  design (CRD) consisting of 7 treatments and 4 replications. Data were analyzed using further test of Least Significant Difference (LSD). The results showed that cardboard waste, klaras and husks had the best effect on the growth of edible mushrooms on the wet weight of mushroom and the number of fruit bodies was in treatment 6 with the composition (cardboard 25% + Straw 25% + and husks 50%) with average weight. Wet 25.97 grams and the average number of fruit bodies 4.53. Whereas the effect of cardboard waste, klaras and husks has an effect on the growth of mushroom with the best growth in the size of mushroom fruit body diameter in treatmen 4 with a combination of 50% cardboard waste, 25% klaras and 25% rice husks with an average body diameter. Mushroom fruit is 6.57 cm.Keywords: Mushrooms, Waste, Cardboard, Klaras, Rice Husk, Planting Media ABSTRAKBudidaya jamur memiliki prospek yang cerah karena kondisi alam dan lingkungan di Indonesia sangat mendukung. Jerami merupakan media tanam yang umum digunakan untuk pertumbuhan jamur merang. Kardus, kelaras dan sekam merupakan salah satu limbah rumah tangga yang dapat dijadikan sebagai alternatif media tanam jamur merang sebagai pengganti jerami. Penelitian ini berupaya mencari tahu pengaruh kombinasi kardus, kelaras dan sekam padi sebagai media tanam jamur merang serta mengetahui perbandingan kombinasi komposisi media tanam limbah kardus dan kelaras yang dapat memberikan pengaruh terbaik pada pertumbuhan jamur merang.  Penelitian ini dilaksanakan di Desa Rukoh, Banda Aceh pada Bulan Oktober sampai November 2019 selama ± 2 bulan. Penelitian ini menggunakan metode kuantitatif dalam bentuk eksperimen dengan Rancangan Acak Lengkap (RAL) terdiri dari 7 perlakuan dan 4 ulangan. Data dianalisis menggunakan uji lanjut Beda Nyata Terkecil (BNT). Hasil penelitian menunjukkan limbah kardus, kelaras dan sekam berpengaruh  terhadap pertumbuhan jamur merang yang paling baik terhadap berat basah jamur merang serta jumlah tubuh buah adalah pada perlakuan 6 dengan komposisi (kardus 25% + jerami 25% dan sekam 50%) dengan rata-rata berat basah 25,97 gram dan rata-rata jumlah tubuh buah 4,53. Sedangkan pengaruh limbah kardus, kelaras dan sekam berpengaruh  terhadap pertumbuhan jamur merang yang paling baik pertumbuhan ukuran diameter tubuh buah jamur merang yang paling baik pada perlakuan 4dengan kombinasi 50% limbah kardus, 25% kelaras dan 25% sekam padi dengan rata-rata diameter tubuh buah jamur merang adalah 6,57 cm.Kata Kunci: Jamur Merang, Limbah, Kardus, Kelaras, Sekam Padi, Media Tanam

Abstract

The low critical thinking skills of learners are triggered by less supportive learning. The learning process has not emphasized the development of critical thinking skills, it is because in learning learners tend to memorize concepts so that critical thinking skills are not trained, making learners less active in the learning process and not achieving minimum completion criteria, there needs to be efforts to overcome them. The formulation of the problem in this study is: Is there a significant difference in the improvement of students' critical thinking skills with the implementation of STEM-based Experiential Learning learning in class VII MTsN Aceh Besar? This research aims to find out the significance of differences in improving learners' critical thinking skills with the implementation of STEM-based Experiential Learning learning. The method used in this study is an experimental method with the design of One Group Pretest-Postets design. The population of this study is all students of class VII MTsN Aceh Besar which amounted to 120 learners. The instrument used to measure critical thinking skills using multiple choice questions is 10 questions. Data analysis using the One Simple t-Test test at a significant level of 0.05. The results of the One Simple t-Test test of the students' critical thinking skills were obtained by Sig. (0.00) < (0.05), showed that learners' critical thinking skills were better and there was a significant improvement with the implementation of STEM-based Experiental Learning.

Abstract

The learning process has not emphasized the development of skills; this is because in learning students tend to memorize concepts so that learning outcomes and self-efficacy are not trained optimally. The formulation of the problem in this study is: is there a difference in learning outcomes and student self-efficacy between the use of the Problem Base Learning model combined with the E-STEM-based Student Worksheet with the Problem Base Learning model on environmental pollution material at MAN Banda Aceh? This research is quantitative with an experimental research approach. The research design used was pretest-posttest control group design. The population of this study was all students of class X MAN 3 Banda Aceh, totaling 84. The sampling technique used a total sampling of 84 students. The instrument used to measure learning outcomes is multiple choice questions using 30 questions and self-efficacy is measured using a questionnaire. Data analysis of learning outcomes and self-efficacy of students using independent sample t-test at a significant level of 0.05. The results showed that there were differences in student learning outcomes and self-efficacy after applying the Problem Base Learning model combined with the E-STEM-based Student Worksheet on environmental pollution materials at MAN Banda Aceh

Abstract

TPACK competence of teachers can improve the quality of learning in accordance with the demands and changes. Information on TPACK competencies is needed regarding the ability of teachers to know the professional development of teachers. This study aims to determine the competence criteria of biology teachers in Aceh Province in the components of CK, TK, PK, PCK, TCK, TPK, and TPACK, to find out the significant differences in TPACK competence of biology teachers based on teacher certification and teaching experience. This type of research is descriptive with a quantitative approach. The population in this study were all Biology teachers at state high schools in Aceh Province, namely civil servants, and non-civil servants, totaling 692 teachers. Sampling using a simple random sampling technique, obtained 232 respondents which is a total of 33.53% of the number of teachers in each district. The research instrument in the form of a questionnaire was adapted from Schmidt which consists of 38 items and has gone through face validity. Data analysis for the TPACK component is described according to the predicate criteria. The TPACK competence of biology teachers based on certification was analyzed by independent sample t-test, while the TPACK competencies of biology teachers based on teaching experience were analyzed using ANOVA. The results showed that the highest competency criteria for the TPACK component were PK with a score of 83.35% (very good), while the lowest was TK with a score of 76.84% (good). The results also showed that there was no significant difference (p> 0.05) in the TPACK competence of biology teachers based on certification and teaching experience

Abstract

Critical thinking skills are competencies that are expected to be possessed by every student in 21st century learning. One of the efforts that can be made to improve students' critical thinking skills is to apply the flipped classroom learning model. This study aims to determine the effect of students' critical thinking skills through the application of the blended learning model of the flipped classroom. The research approach used is quantitative with a quasi-experimental type of research, the research design used is pretest-posttest non-equivalent control group design. Sampling used a total sampling technique from the entire population of 102 students of class XI IPA at SMAN 1 Peulimbang. The instrument used to measure critical thinking skills is in the form of essay questions accompanied by an assessment rubric. Data on critical thinking skills were analyzed by parametric statistics using the independent sample t test. The results showed that the value of < 0.05, then there was an effect of students' critical thinking skills after the application of the blended learning model of the flipped classroom on the respiratory system material

Abstract

Potensi adalah kemapuan atau dasar pengetahuan yang dimiliki oleh seorang individu yang bisa dikembangkan menjadi lebih baik lagi. Kreativitas ialah kemampuan individu dalam menyelesaikan maslah dengan menemukan ide ataupun gagasan yang baru dari unsur yang sudah ada sebelumnya. Kajian ini bertujuan untuk memberikan informasi tentang potensi individu yang sudah ada sejak lahir agar bisa dikembangkan untuk kepentingan semua orang dengan ide ide kreatifnya. Metode yang digunakan ialah kajian pustaka dari sumber yang relevan yang berkaitan dengan potensi bakat. Abstract

AbstrakKajian ini bertujuan untuk mengurai tentang intelegensi dan bakat pada prestasi siswa di pembelajaran di sekolah. Metode yang dilakukan pada kajian ini melalui studi pustaka dan juga literature research dengan hal-hal yang berkaitan dengan persoalan tersebut. Pembahasan pada kajian ini berdasarkan pada pendapat para ahli dan juga hasil penelitian terdahulu yang berkaitan dengan persoalan intelegensi dan bakat pada prestasi siswa. Belajar merupakan sesuatu yang mutlak harus dilakukan dalam pendidikan. Pendidikan merupakan upaya yang dilakukan untuk meningkatkan sumber daya manusia siswa dengan cara membimbing dan juga memberi fasilitas untuk proses belajar tersebut. Selama proses belajar terdapat hasil belajar yang didapatkan siswa dari proses belajar yang dilakukan dan dinilai oleh guru. Dari hasil belajar di sekolah didapatkan pencapaian prestasi siswa yang disebut juga dengan prestasi akademik. Ada banyak faktor yang mempengaruhi prestasi akademik dari seorang siswa, diantaranya terdapat faktor internal yaitu tingkat intelegensi dan juga bakat yang dimiliki oleh setiap siswa tersebut. Kata kunci: Intelegensi, Bakat, Prestasi.
