Abstract

The waste water of blue crab pasteurization has potential in environmental pollution. It contained TSS of 206.5mg.1-1, BOD 7,092.6mg.1-1 and COD of 51,000mg.1-1. on the other hand, it also contains an interesting flavor compound, which composed of 0.23% non protein nitrogen and 17 amino acids where the highest was glutamic acid one. In this study, pre-filtration step using filter size 0.3 µ followed by reverse osmosis has been used to reduce these pollutions load and flavor compound recovery. During pre-filtration steps, TSS was reduced to 74.8% so turbidity decrased reased until 31%. After reverse osmosis process, BOD, and COD decreased more than 99%, and there was no amino acids detected in permeate stream. Factors that affect performance of reverse osmosis were transmembrane pressure, temperature and pH. The higher transmembrane pressure, temperature and pH resulted the higher the flux permeate. The use of higher temperature make flux increasing, eventually increasing transmembrane pressure make the flux increased only at transmembrane pressure less than 716 kPa. The protein rejection was influenced unsignifanctly by transmembrane pressure, temperature and pH. During concentrating flux declined exponentially by time function. At concentration factor 2.75 resulted 79% and 12% of increasing protein and NPN, respectively. The amino acids content can be increased 2−23 times of the origin. Even arginin and sistin, the amino acids that were undetectable initially, but they can bedetected at concentration of 0.0360 and 0.0250 (w/v) respectively at the end of the process. Hidrolysis and fermentation process can increase the amino acid content 31−45 times.

The enzymes that play a role in the enzymatic process is primarily proteolytic enzymes. Cathepsin is one of the proteolytic enzymes found in animal tissue that hydrolyzes peptide bonds of proteins. This study aims to extract the cathepsin, characterize the crude extract derived from catfish. The stages of this research consist of the extraction and characterization of the cathepsin from catfish. Result of the extraction was crude extract of cathepsin with activity of 0.278 U/mL. The enzyme had optimum temperature of 50°C, pH 6 and substrate concentration of 2%. The activity of the cathepsin was inhibited by metal ions of Fe3+, Cu2+, Ca2+, but increased by metal ions of Mg2+.

Abstract

Enzim protease memiliki peranan penting dalam proses metabolisme bakteri patogen. Protease bakteri tersebut dapat dihambat aktivitasnya oleh inhibitor protease. Iinhibitor protease dapat diproduksi dari bakteri Acinetobacter baumanni yang hidup bersimbiosis dengan sponge laut (Plakortis nigra). Agar produksi inhibitor protease dapat lebih optimal maka perlu diketahui komposisi media marine broth yang baik. Perlakuan yang digunakan pada penelitian ini adalah modifikasi konsentrasi peptone (0,5% dan 1%) dan modifikasi konsentrasi yeast extract (0,1%; 0,5% dan 1%). Peningkatan konsentrasi yeast extract memperpanjang waktu propagasi bakteri A. baumanni dan memperlambat awal produksi inhibitor. Peningkatan konsentrasi pepton cenderung menghasilkan pertumbuhan yang lebih baik. Aktivitas penghambatan inhibitor protease tertinggi terjadi pada media dengan kombinasi konsentrasi pepton dan yeast extract masing-masing 0,5%.Kata kunci: inhibitor protease, Acinetobacter, simbiosis sponge, produksi

Abstract

Shrimp is a  very important export  commodity with  high market value world wide. However, it is still facing problem related to the waste and deterioration quality as main issues for the shrimp industry. In this experiment, polyphenoloxidase from the carapace of Penaeus monodon was extracted and characterized. The research was carried out to obtain the optimum extraction condition and to evaluate the properties of enzyme i.e., pH, optimum temperature for activating enzyme, kinetic enzyme, and chelating on metal ion. The best method for PPO enzyme extraction used buffer with 1:3  proportion.  The optimum activity of enzyme was at pH 7 and temperature of 35°C. The kinematic enzyme (Km) value and the maximum substrate concentration were 5.42 mM and 7.5 mM, respectively.  Na+, Ca2+, Zn2+, and EDTA with concentration 5 and 10 mM inhibited enzyme activity.  Cu2+at concentration of 10 mM and Mn2+ at concentration 5 mM also inhibited enzyme activity Keywords: carapace, characterization, polyphenoloxidase, shrimp

Abstract

In the last decade, a concern on protease as medicinal target for overcoming bacterial and viral diseases has been rapidly increased because of the obvios involvement of this enzyme in the molecular of the diseases mechanism. The porpuse of this research was to characterize proteases from fish pathogenic bacteria Aeromonas hydrophila. The bacteria were grown in media containing triptone 1%, NaCl 1% and yeast extract 0,5%. The optimum production time of A. hydrophila was 48 h, the optimum pH was 7,5, the optimum temperature was 50oC. Study on the effect of metals ion and spesific inhibitors indicated that protease from A. hydrophila was serin metaloprotease.Keywords: protease, characterization, pathogenic bacteria

Abstract

Beberapa bakteri patogen memproduksi enzim hidrolitik seperti protease dan hialuronidase yang berfungsi untuk mendegradasi komponen matrik ekstraseluler sehingga dapat merusak struktur jaringan inang. Tujuan penelitian ini adalah untuk melakukan karakterisasi protease dari bakteri Staphylococcus epidermidis yang diisolasi dari koleksi Rumah Sakit Pertamina Jakarta. Bakteri ditumbuhkan pada media Luria broth (LB) yang mengandung tryptone 1 %, NaCl 1 % dan yeast extract 0,5 %. Hasil karakterisasi menunjukkan bahwa protease S. epidermidis ini memiliki pH dan suhu optimum 8 dan 50 oC. Ion logam Mn2+ (5 mM) dan Ba2+ (5 mM) merupakan aktivator kuat protease S. epidermidis yang dapat meningkatkan aktivitas protease masing-masing 3 dan 2 kali lipat dari protease kontrol, sedangkan Na+ (1 mM), K+ (1 mM), Fe3+ (1 dan 5 mM), Zn2+ (5 mM), dan Ca2+ (1 mM) merupakan inhibitor ion logam yang kuat. Protease S. epidermidis digolongkan ke dalam serin metaloprotease karena dapat dihambat secara sempurna oleh PMSF dan EDTA. Protease tersebut mempunyai berat molekul sekitar 35 kD.Kata Kunci: bakteri patogen, karakterisasi, protease, Staphylococcus epidermidis.

Abstract

Abstrak Proses perkembangan jiwa tidak harus dipahami sebagai perkembangan fungsi-fungsi yang tidak saling mempengaruhi satu sama lain, melainkan sebagai perwujudan dari suatu rencana, di mana kekuatan jasmani dan rohani dalam struktur yang berurutan memperoleh pelajaran atau latihan yang penting untuk pembentukan yang tepat. Pendidikan berarti mewujudkan atau melaksanakan rencana kodrat alam tersebut.Namun, realitas yang berkembang di masyarakat, proses pendidikan berbenturan dengan fungsi-fungsi perilaku perkembangan dalam berinteraksi dengan sesamanya baik di sekolah maupun dalam masyarakat. Untuk itu para remaja atau siswa idealnya memperoleh bantuan psikologis sepenuhnya sehingga interaksi sosial dan perkembangannya optimal.Dengan demikian, layak bila sekolah memberikan pelayanan bimbingan psikologis dalam menghadapi semua tantangan, kesulitan,dan masalah aktual yang timbul dewasa ini demi perkembangan setiap peserta didik yang semaksimal mungkin. Pelayanan ini tidak terbatas pada bidang belajar di sekolah, tetapi mencakup seluruh aspek kehidupan siswa. Namun, semua aspek selalu dipandang dari sudut perkembangan individual dan integrasi kepribadian bagi masing-masing peserta didik.Kata Kunci: psikologi, perkembangan dan interaksi sosial.

Abstract

Fish skin is one of many parts from fish that can be utillized beside the muscle. Proteolytic enzymes like cathepsin and collagenase are known as one of the factors that influence in fish skin quality deterotiation after death. The research purposes were to determine post mortem phase, quality deterioration pattern, cathepsin and collagenase activity, and its correlation to the parameters of milkfish freshness. The observations were done on fasted and fed fish before harvested at room and chilling temperature storage. The highest chatepsin and collagenase activity i.e. 7800-1429 U/ml and 0,0667-0,0792 U/ml respectively, were found in fish during the post-rigor phase. Simple linear correlation analysis showed that chatepsin and collagenase activity have a very close linear relationship (r 0.7) with fish freshness quality parameters during the process of quality deterioration from pre rigor until post rigor phase.

Abstract

African catfish (Clarias gariepinus) is important freshwater fish commodity in Indonesia. African catfish also has high protein content, which is potential to be used as protein hydrolysate. Fish protein hydrolysate offered many advantages in food, feed, agriculture, microbiology and medicinal’s field. This research aimed to determine the optimal condition for hydrolysis of African catfish protein and to characterize protein hydrolysate from African catfish. The optimal concentration of papain used for hydrolysis of African catfish protein was 5% (w/v) for 6 hours hydrolysis time and degree of hydrolysis was 35.37%. Protein hydrolysate from African catfish revealed 21.16% of yield and water content of 5.46%; ash content of 5.71%; protein content of 53.29%; and fat content of 1.94%. Protein hydrolysate from African catfish contained 15 types of amino acids, with 9 types of essential amino acids and 6 types of non essential amino acids. The highest essential amino acid composition was lysin of 5.23%, while the highest non essential amino acid composition was glutamic acid of 7.77%. Protein hydrolysate from African catfish showed protein digestibility by in vitro enzymatic hydrolysis of 98.57%.Key words: African catfish, amino acid, fish protein hydrolysate, papain

Abstract

The purpose of this study is to determine the micro minerals composition i.e. iron, zinc, copper, and iodine) and heavy metals i.e. lead, cadmium, and mercury) on three groups of weight of milkfish (Chanos chanos). Micro minerals and heavy metals were analyzed using atomic absorption spectrofotometric (AAS). The results showed that the highest levels of micro minerals were found at weight ±150 g (6.95±0.16 mg of zinc/kg of dry basis, copper 0.55±0.01 mg/kg of dry basis , and iron 12.14±0.06 mg/kg of dry basis. The highest iodine content contain was found at weight 102 g ± is 76.33±0.01 mg/100g of dry basis. The concentrations obtained in lead was 0.143±0.0057 mg/kg, cadmium was 0.0433±0.011 mg/kg, and mercury was 0.143±0.0057 mg/kg of dry basis,. The heavy metals level from Tanjung Pasir Tangerang District are safe to be comsumed since it is still below the limit for food savety.
