Abstract

Mulberry (Morus alba L.) is considered as an important plant in traditional Chinese medicine, due to its various compounds, including phenols and flavanoids, which have antioxidant properties so that it can be a potential anticancer candidate. This study aimed to determine the antioxidant activity, phenolic content, and potential anticancer activity in Mulberry stem extract. The extraction was done by maceration using ethanol as the solvent, while antioxidant activity test used ABTS (2,2?-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) method, phenolic content determination used Folin-Ciocalteu reagents, and anticancer activity test used the MTT (3-(4,5- dimetiltiazol-2-il)-2,5-difenil tetrazolium bromida) method on WiDr cancer cells and Vero cells. The result of total phenolic Mulberry stem extract was 35.9%, the antioxidant activity value was 83.18 g/mL, the IC50 value for anticancer activity for WiDr cells was 71.24 g/mL and Vero cells IC50 value was 154.241 g/mL. It could be concluded that the Mulberry stem ethanol extract has strong antioxidant activity and has the potential to be an anticancer agent selectively against WiDr cancer cells.

Abstract

α-amilase is an enzyme that catalyzes the hydrolysis of starch polysaccharides into oligosaccharides to then produce glucose. Inhibitions of the α-amylase enzyme is an effective strategy in modulating blood sugar levels in diabetics. The aglycon curculigoside A has similar structure to chalcon which is able to inhibit the α-amilase enzyme for the treatment of type 2 diabetes. The aim of this study was to determine the interaction of the curculigoside A aglycone compound and its derivatives with the α-amylase enzyme by using the docking simulation method. Docking simulations were carried out by using AutoDock 4.2 and α-amilase protein (PDB ID: 1B2Y) as macro molecule. The docking results showed that the aglycone curculigoside A and its derivatives able to interacted into the active site of the α-amylase enzyme. Three best ligands according to the simulation and prediction tests were compound 10, compound 23, and compound 41 has formed hydrogen bonding to Asp197, Glu233 and Asp300 residues with free bonding energy of -7.29, -7.22, dan -7.84 kcal/mol, respectively. In conclusion, Three best ligands has the same pattern of hydrogen bonds to the native ligand AC1 (6-methyl-5-(4,5,6-trihydroxy-3-hydroxymethyl-cyclohex-2-enylamino)-tetrahydro-pyran-2,3,4-triol) via amino acids redisues of α-amylase that play a role in the substrate by hydrolysis process.Keywords: aglycon curculigoside A, α-amylase, diabetes, docking

Abstract

Karamunting (Rhodomyrtus tomentosa L.), a plant used as a traditional medicine, is widely distributed throughout Indonesia. Karamuting has the potential as an antioxidant and anticancer agents because of its phenolic and flavonoid components. This study aimed to determine total phenolic level, antioxidant activity and cytotoxic of karamunting leaves extract. The simplisia of karamunting leaves were extracted by maceration method using 96% ethanol, tested for its antioxidant activity using DPPH and its citotoxic by MTT method (3- (4,5-dimethyltiazol-2-il) -2,5-diphenyl tetrazolium bromide) on WiDr cells and Vero cells. The results of this study showed that the yield of karamunting extract were 15.635% with a total phenolic level of 0.76%, antioxidant activity (IC50 values) of 15,330 μg / mL and cytotoxicity (IC50 values) of 205.7069 μg / mL for WiDr cells and 44.87703 μg / mL for Vero cells. Ethanol extract of karamunting leaves possessed a very strong antioxidant activity while it showed a weak cytotoxic effect on WiDr cells and toxic effect on Vero cells.Keywords: Antioxidant, Anticancer, Karamunting

Abstract

Tanaman daun asam jawa (Tamarindus indica L.) dan rimpang Kencur (Kaempferia galanga L.) merupakan salah satu tanaman obat yang berpotensi sebagai antikanker. Penelitian ini bertujuan untuk mengetahui aktivitas sitotoksik ekstrak kombinasi daun asam jawa dan rimpang kencur terhadap sel kanker WiDr dan MCF-7 berdasarkan nilai IC50. Simplisia daun asam jawa dan rimpang kencur diekstraksi dengan metode maserasi menggunakan etanol 96%. Ekstrak etanol selanjutnya dilakukan uji sitotoksik dengan metode MTT(3-(4,5- dimetiltiazol-2-il)-2,5-difenil tetrazolium bromida) terhadap sel WiDr dan sel MCF-7. Perolehan nilai IC50 ekstrak daun asam jawa, ekstrak rimpang kencur, ekstrak kombinasi daun asam jawa dan rimpang kencur dengan perbandingan (3:7) dan (7:3) sebesar 31,31 ?g/mL, 88,79 ?g/mL, 126,13 ?g/mL dan 271,80 ?g/mL terhadap sel WiDr dan 128,63 ?g/mL, 270,43 ?g/mL, 265,83 ?g/mL dan 369,79 ?g/mL pada sel MCF-7. Berdasarkan hasil penelitian ini ekstrak kombinasi daun asam jawa dan kencur tidak tidak menunjukkan potensi yang lebih baik sebagai antikanker pada sel WiDr dan MCF-7 dibanding ekstrak tunggalnya.

Abstract

Bunga kacapiring (Gardenia augusta Merr) dan lemon (Citrus limon L.) merupakan tanaman yang mengandung beberapa senyawa aktif diantaranya yaitu flavonoid, saponin, alkaloid, dan tanin yang berkhasiat sebagai antibakteri, pada penelitian ini kedua bahan tersebut dibuat menjadi sediaan paper soap. Pembuatan sediaan paper soap ini adalah karena paper soap merupakan sebuah inovasi produk sediaan sabun yang unik yang lebih praktis, ringan dibawa kemana-mana dan dapat berkhasiat sebagai antibakteri. Tujuan penelitian ini adalah untuk membuktikan bahwa ekstrak bunga kacapiring (Gardenia augusta Merr) dan kulit lemon (Citrus limon L.) dapat diformulasikan menjadi sediaan paper soap dan untuk membuktikan bahwa formulasi sediaan paper soap ini memiliki aktivitas antibakteri terhadap Staphylococcus aureus. Metode penelitian ini merupakan penelitian eksperimental dengan evaluasi sediaan secara fisika dan kimia serta pengujian aktivitas antibakteri dengan metode difusi teknik paper disc. Hasil penelitian yang didapatkan adalah sediaan paper soap kombinasi ekstrak etanol bunga kacapiring (Gardenia augusta Merr) dan kulit lemon (Citrus limon L.) memiliki efek sebagai antibakteri terhadap Streptococcus aureus.

Abstract

Daun Bidens biternata (Lour) Merr. & Sherff digunakan oleh masyarakat secara empiris di Nusa Tenggara Timur untuk pengobatan penyakit liver. Kandungan flavanoid dalam tanaman ini sekitar 60 jenis yang mempunyai berbagai macam aktifitas seperti antioksidan dan anti-inflamasi. Penelitian ini bertujuan untuk mengetahui efek hepatoregeneratif ekstrak etanol daun B. biternata pada tikus yang mengalami kerusakan hati akibat induksi parasetamol dengan parameter ALT, AST dan bilirubin. Sebanyak 12 ekor tikus yang dibagi dalam 4 kelompok perlakuan: kontrol negatif (1% Na.CMC); dan 3 kelompok perlakuan (dosis 50; 100; dan 200 mg/kgBB). Berdasarkan analisis statistik, tidak ada perbedaan yang signifikan antara 3 kelompok perlakuan ekstrak (p>0.05), akan tetapi didapatkan perbedaan yang bermakna antara kelompok perlakuan dengan kontrol negatif. Kesimpulan penelitian ini adalah ekstrak etanol B. biternata pada semua dosis perlakuan dapat menurunkan kadar ALT, AST dan bilirubin yang mengindikasikan bahwa ekstrak memiliki efek hepatoregeneratif pada tikus yang diinduksi parasetamol.

Abstract

Daun Pare (Momordica charantia L.) mempunyai kandungan flavanoid yang memiliki potensi sebagai obat penurun panas. Penelitian ini bertujuan untuk memformulasi sediaan patch ekstrak daun Pare (Momordicacharantia L.) dan menguji efektivitasnya sebagai antipiretik. Pembuatan sediaan patch menggunakan basis HPMC dan PVP dengan metode penguapan pelarut (solvent evaporation casting). Metode pengujian efek antipiretik yang dilakukan dalam penelitian ini adalah induksi pepton 5% sebanyak 1 ml secara subkutan. Karakteristik sediaan didapat dari evaluasi meliputi uji organoleptik, uji keseragaman bobot, uji ketahanan lipatan, uji ketebalan dan uji susut pengeringan. Hasil uji efektivitas diperoleh pada konsentrasi 1% dengan penurunan suhu yang optimal pada menit 90 dan 120.

Abstract

Soursop leaves are reported to have several active compounds such as flavonoids, alkaloids and fatty acids and soursop leaves are reported to have many pharmacological effects, one of which is for the treatment of blood cholesterol levels. This study aims to compare the effectiveness of the active fraction of soursop (Annona muricata L.) ethanol extract on the reduction in total cholesterol levels of male mice (Mus musculus). The method used is to use 5 groups of male mice that were given 5 different treatments. Then proceed with a statistical test using LSD analysis. The results obtained showed that the effect of reducing total cholesterol of soursop leaf extract fraction at a dose of 100 mg/kg BB has no significant effect with positive control with a value of p = 0.501 > 0,05 and soursop leaf extract fraction dose of 150 mg/kgBB body weight with a value of p = 0.580 > 0.05. So, both are thought to be able to reduce total cholesterol levels which are almost the same as positive control.

Abstract

This experiment aims to determine the correlation of total phenolic and flavonoid content of jati putih leaves fraction (Gmelina arborea Roxb.) towards Antioxidant activity . Sample was extracted by maceration method using ethanol 70% to obtain the ethanol extract (EE), followed by liquid-liquid extraction method to obtain fraction of ethyl acetate (EA) and n-Hexane (EH). The phytochemical screening and determination of total phenolic and flavonoid content were done by colorimetric method. Antioxidant activity were done by DPPH, FRAP and ABTS methods. Phytochemical screening showed positive results for flavonoids, phenolic and saponins. The largest total phenolic content was found on EA (11,59 µg/ml ± 0,3 %b/b EAG) and the largest total flavonoid content was on EA (3,88 µg/ml ± 0,02 %b/b EK). The total phenolic and flavonoid content of Jati putih leaves has a correlation with antioxidant activity. The coefficient correlation of activity on reducingDPPH radical was 56,7% (total of phenolic content) and 57,8% (total of flavonoid content) and on iron reduction power in FRAP method was 99,9% (total of phenolics and flavonoids content). The relationship with the activity in reducing radical ABTS obtained coefficient correlation of 57,0% and 58,1% for total phenolic and flavonoids contents, respectively.

Abstract

Pre-eklampsia merupakan hipertensi yang ditandai dengan meningkatnya tekanan darah yakni ≥140/90 mmHg, terjadi setelah kehamilan berusia 20 minggu dan kebanyakan kasus ditegakkan dengan adanya proteinuria. Disertai dengan sakit kepala, kelainan pada penglihatan, nyeri dada serta dyspnea. Penelitian memiliki tujuan untuk mengetahui jenis antihipertensi yang dipergunakan untuk pasien pre-eklampsia di Rumah Sakit Umum Daerah Latemmamala Soppeng periode April 2020-Maret 2021, karena didasari oleh fakta tingginya kematian akibat hipertensi kehamilan serta bervariasinya terapi yang diberikan. Data dikumpulkan secara retrospektif yaitu pengambilan data rekam medis pre-eklampsia, berupa data karakteristik pasien (usia pasien, usia kehamilan), tekanan darah, diagnosa, hasil laboratorium berupa kadar proteinurea dan data pengobatan antihipertensi (jenis obat, dosis obat, frekuensi penggunaan obat). Dari hasil penelitian, didapatkan terapi antihipertensi yang digunakan pada pasien pre-eklampsia yaitu CCB dengan persentase 46,15%, diuretik sebanyak 3,84%, kombinasi antihipertensi sebanyak 34,58%. Berdasarkan hasil penelitian, ditarik kesimpulan profil antihipertensi yang digunakan untuk pasien pre-eklampsia di Rumah Sakit Umum Daerah Latemmamala Soppeng yaitu menggunakan obat golongan CCB sebagai antihipertensi terbanyak. Yang dimana berdasarkan literatur, nifedipin termasuk terapi lini yang pertama untuk digunakan pada penanganan pre-eklampsia.
