Abstract

Penelitian ini bertujuan untuk mengetahui rendemen ekstrak, senyawa fitokimia dan aktivitas antibakteri tumbuhan perairan kiambang (Salvinia molesta) dengan proses ekstraksi menggunakan pelarut n-heksan, etil asetat dan etanol. Penelitian ini menggunakan metode eksperimental laboratorium dan analisis data dilakukan secara deskriptif. Parameter yang diamati yaitu rendemen ekstrak, kandungan fitokimia dan aktivitas antibakteri. Rendemen ekstraksi menggunakan etanol memiliki rendemen yang paling tinggi (14,334%) dibandingkan dengan pelarut n-heksan (0,229%) dan pelarut etil asetat (1,767%).  Pada ekstrak dengan pelarut etanol mengandung senyawa fitokimia  yaitu tanin, saponin, steroid dan triterpenoid, estrak menggunakan etil-asetat didapatkan senyawa tanin, steroid dan triterpenoid, sedangkan ektrak menggunakan pelarut n-heksan didapatkan senyawa fitokimia steroid dan triterpenoid. Diameter penghamabatan tertinggi pada ekstrak kiambang dengan pelarut etanol yang memiliki diameter penghambatan sebesar 9,5 pada konsentrasi 2000 ppm.  Diamater penghambatan cenderung meningkat dengan semakin tingginya konsentrasi ekstrak kiambang. Hasil penelitian ini dapat dimanfaatkan sebagai sumber pengawet alami yang nantinya dapat diaplikasikan pada fillet ikan, surimi dan edible film sebagai pengemas pempek.

Abstract

Sensoris Characteristic of Kamaboko Adding Purun Tikus (Eleocharis dulcis) Extract. pp. 172-175.This study aimed to determine sensory characteristics of kamaboko with the addition of extracts of purun tikus (Eleocharis dulcis). This study uses experimental laboratory methods and data analysis is carried out descriptively. The parameters observed were sensory tests which included the appearance, texture and aroma of kamaboko with the addition of purun tikus extract. The results of sensory values with the appearance parameters and aroma of kamaboko with different extract treatment and storage duration showed the value of appearance and aroma of kamaboko without the addition of extract was higher than that of kamaboko with the addition of purun tikus extract. While the texture parameters showed kamaboko with the addition of purun tikus extract had a higher textur compared to kamaboko without the addition of extract.

Abstract

The aim of this study was to bacteria isolation and characterization proteases from water swamp. The water swamp samples collected fromIndralaya for microbial isolation. Three isolates showed proteolytic index >1.00. The optimum pH of extraceluller proteases from A6S3,A4S3 and A15S3 were 7.5; 8.0; 8.0, respectively. The optimum temperature of A6S3, A4S3 and A15S3 protease were 40; 50; 50 oC,respectively. Effect of metal ion (Fe, K, Mn, and Zn) shown Fe and K were inhibit protease A6S3, all metal ion were inhibit protease A4S3and K, Mn and Zn inhibit protease A15S3. Study on the effect of metals ion and spesific inhibitors indicated that all protease weremetaloprotease. Their moleculer weights were determined by using SDS-PAGE and zymogram technique for A6S3 isolate were 70, 88, 106and 121 kD respectively. Whereas for isolate A4S3 was 138 kD and isolate A15S3 was 131 kD.

Abstract

In an effort of obtaining indigenous protease producing bacteria, screening for bacterial protease was conducted from samples collected from Indralaya soil swamp, South Sumatera. Three of 31 colonies showed high protease activity with proteolytic index >1.00. T1S1 produced enzyme with the highest activity. The crude enzyme activity after 48 hours of incubation was 0.391 IU/ml. The optimum pH of the extracelull proteases from T1S1, T3S2 and T3S3 were 8.0, 8.0, and 7.5, respectively. The optimum temperature of T1S1, T3S2 and T3S3 proteases were 40, 50, and 500C, respectively. All metal ions tested (Na+, K+, Mn2+, Zn2+ and Fe2+) inhibited proteases except Fe2+ which activatesthe T3S3 protease at 5 mM. EDTA (1 and 5 mM) inhibited all proteases. Study on the effect of metals ion and spesific inhibitors indicated that all protease are metaloprotease. Molecular weights was determined using SDS-PAGE and zymogram technique. The molecular weight of T1S1 proteases was 121 kD,T3S2 proteaseswere 51, 71, and 119 kD whereas T3S3 proteaseswere 49, 70, and 116 kD.

Abstract

Beberapa bakteri patogen memproduksi enzim hidrolitik seperti protease dan hialuronidase yang berfungsi untuk mendegradasi komponen matrik ekstraseluler sehingga dapat merusak struktur jaringan inang. Tujuan penelitian ini adalah untuk melakukan karakterisasi protease dari bakteri Staphylococcus epidermidis yang diisolasi dari koleksi Rumah Sakit Pertamina Jakarta. Bakteri ditumbuhkan pada media Luria broth (LB) yang mengandung tryptone 1 %, NaCl 1 % dan yeast extract 0,5 %. Hasil karakterisasi menunjukkan bahwa protease S. epidermidis ini memiliki pH dan suhu optimum 8 dan 50 oC. Ion logam Mn2+ (5 mM) dan Ba2+ (5 mM) merupakan aktivator kuat protease S. epidermidis yang dapat meningkatkan aktivitas protease masing-masing 3 dan 2 kali lipat dari protease kontrol, sedangkan Na+ (1 mM), K+ (1 mM), Fe3+ (1 dan 5 mM), Zn2+ (5 mM), dan Ca2+ (1 mM) merupakan inhibitor ion logam yang kuat. Protease S. epidermidis digolongkan ke dalam serin metaloprotease karena dapat dihambat secara sempurna oleh PMSF dan EDTA. Protease tersebut mempunyai berat molekul sekitar 35 kD.Kata Kunci: bakteri patogen, karakterisasi, protease, Staphylococcus epidermidis.

Abstract

In the last decade, a concern on protease as medicinal target for overcoming bacterial and viral diseases has been rapidly increased because of the obvios involvement of this enzyme in the molecular of the diseases mechanism. The porpuse of this research was to characterize proteases from fish pathogenic bacteria Aeromonas hydrophila. The bacteria were grown in media containing triptone 1%, NaCl 1% and yeast extract 0,5%. The optimum production time of A. hydrophila was 48 h, the optimum pH was 7,5, the optimum temperature was 50oC. Study on the effect of metals ion and spesific inhibitors indicated that protease from A. hydrophila was serin metaloprotease.Keywords: protease, characterization, pathogenic bacteria

Abstract

Proteases constitute one of the most important groups of industrial enzymes, therefore screening of these proteases from bacteria is important. The aim of this study was to characterize proteases from plant swamp bacteria. The result of  proteolytic assays revealed three isolates possesed proteolytic index >1.00 (T1P4, H1P4 and K1P4). The optimum fermentation time of each isolate was 48 hours. The optimum pH of proteases from T1P4, H1P4 and K1P4 was 8.0; 7.5; and 8.0,  respectively. The optimum temperature of T1P4, H1P4 and K1P4 protease was 50 °C; 40 °C;  and 50 °C, respectively. Metal ions (Fe, K, Mn, and Zn) inhibited T1P4 protease except Fe2+ (5 mM), H1P4 protease except  Zn2+ (5 mM),  K1P4 protease except Fe (1 and 5 mM). Effect of inhibtor specific (EDTA) shown EDTA inhibit all  proteases. Study on the effect of metals ion and spesific inhibitors indicated that all protease were  metaloprotease. The moleculer weights of T1P4, H1P4 and K1P4 proteases were determinited by using SDS-PAGE and zymography technique were 151; 138; and 127 kD, respectively.Key words:  bacteria, characterization, protease, swamp

Abstract

The objective of this research was to make a protein hydrolysates from catfish(Pangasius pangasius) enzymatically using papain enzyme and analyzed the antioxidant activityof protein hydrolysates produced. The research used the method completely randomized designwith two replications the treatment were the difference concentration of the papain enzyme(0%, 1%, 2%, 3%, 4%, 5%, and 6%). The parameters of research were antioxidative activity usingDPPH (2,2-difenil-1–pikrilhidrazil), protein content, and molecular weight using SDS-PAGE(Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis). The results showed that catfishprotein hydrolysates prepared by papain enzyme has antioxidative activity. The highest degree ofhydrolysis was 71.98% at enzyme concentration of 6%. Based on the DPPH scavenging methodcatfish protein hydrolysates has the antioxidative activity with the value 37.85-67.62%. The proteincontent of catfish protein hydrolysates were 20.86-54.47 mg/ml. The molecular weight of catfishprotein hydrolyzates were 11.90-65.20 kDa.Keywords: Antioxidant, catfish, papain enzyme, protein hydrolyzate

Abstract

The complex formed between chitosan and sugar as a result of heating process which induces the Maillard reaction is known to have antibacterial and antioxidant properties. This study aims to compare the antibacterial activity of chitosan monosaccharide complexes against Bacillus subtilis, Listeria monocytogenes and Vibrio cholera in snakehead fish surimi as the food matrix model. The analysis was conducted onthe color intensity of the Maillard Reaction Products (MRPs) which was formed through the reaction of chitosan with glucose, galactose and fructose. The antibacterial activity of each MRPs was studied both invitro and in the food matrix using disc difussion and standard plate count method respectively. The results showed that the chitosan galactose complex had the highest color intensity with absorbance values at 420nm of 0.248. In vitro tests result showed that the chitosan glucose complex had the greatest inhibition on all three test bacteria. Whereas, when MRPs was applied in the food matrix of snakehead fish surimi, thegalactose chitosan complex showed the best inhibitory effect against B. subtilis and L. monocytogenes with a decrease in the number of bacteria of 0.63 and 2.13 log CFU/mL respectively compared to the control.On the other hand, the inhibitory activity of V. cholera in the food matrix was effectively performed by the glucose chitosan complex with the total bacterial count of 2.25 log CFU/mL l lower than the control. The addition of chitosan and chitosan monosaccharide complex to the food matrix generally reduced the total microbial count (TPC) by 1.53 log CFU/mL. This result confirms that the antibacterial activity of MRPs isconsistent .both in vitro and in the food matrix.
