Abstract

In general, local goat productivity in Indonesia is relatively still lower than that from the temperate area. Yearling body weight in local goat about 14-17 kg. However, the local goat have superiority, such as adaptability and a good reproductive performance, which have litter size 1,49-1,61. Increasing local goat productivity may be done by breeding program (cross breeding and selection) and the environmental improvement. Both cross breeding program and selection that involve mating regulation will be efficient by reproductive technology application, such as artificial insemination (AI). Application of AI in goats is limited by four main factors that influence simultaneously, viz. poor quality of frozen semen, the variety of female reproductive performance, improper technical and insemination time, and poor management. To overcome these constraints his obligatory to pay more attention, so that AI will be able to be used as one of tools to increase the population and genetic quality in goats.   Key words: Goat, reproductive efficiency, artificial insemination

Abstract

Indonesia has agreat variety of roosters, either indigenous type as well as exotic and cross breed. The purpose of this experiment was to study the characteristics of semen from three types of Indonesian local roosters such as Merawang, Kampung and crosses Sentul Kampung with Kedu (SK Kedu). A total of 15 roosters consist of  Merawang roosters,  Kampung, and SK Kedu roosters were 5 each. The semen was collected 3 times a week by dorso-abdominal and cloaca massage method. The parameters evaluation was macroscopic characteristics consist of volume, color, consistency, and pH. Microscopic evaluation of semen such as a mass movement, sperm motility, live sperm, sperm abnormality and sperm concentration. Results of this experiment showed that semen volume of  Merawang  (0.40±0.26 mL) was higher (p<0.05) compare to Kampung (0.24±0.12 mL) or  SK Kedu (0.16±0.10 mL) but no difference on semen color, consistency and semen pH. There were no difference in the mass movement, sperm motility and live sperm as well as on sperm abnormality among three types of roosters.  Sperm  concentration of Merawang (4490 million mL-1) was significantly higher than Kampung (3245 million mL-1) and the SK Kedu roosters (3751 million mL-1). Its was conclude that Merawang roosters had good semen quality better than Kampung and SK Kedu roosters

Abstract

Kajian terhadap profil hormon-hormon steroid merupakan kunci penting dalam upaya memahami aspek fisiologis satwa. Beberapa dekade terakhir telah dikembangkan metode alternatif untuk mengetahui profil hormon steroid, yaitu melalui pengukuran metabolit hormon steroid yang diekskresikan melalui ekskreta tubuh seperti feses. Metode tersebut dikenal sebagai metode non-invasif. Metode ini memungkinkan pengumpulan sampel secara terus menerus dalam jangka panjang dengan meminimalisasi gangguan terutama pada satwa liar. Kajian terhadap profil metabolit hormon steroid yang terukur dapat diaplikasikan antara lain untuk mengetahui status reproduksi, penentuan jenis kelamin, studi perilaku hingga monitoring tingkat stres satwa. Berbagai kajian dengan memanfaatkan metabolit hormon steroid telah berhasil diaplikasikan pada berbagai taksa vertebrata. Tulisan ini memberikan gambaran terkini mengenai aplikasi metode non-invasif untuk kajian profil metabolit hormon steroid dari sampel feses.

Abstract

Implantation is the most critical stage in the establishment of pregnancy. In mammals, it has been estimated that between 25% and 60% of conceptuses are lost before or at the time of implantation. The objectives of  this study were to investigate  the ability of blastocyst hatching and attachment of trophoblast cells in in vitro, the outgrowth and differentiation of trophoblast cells in in vitro culture of hatched and non hatched blastocyst, the activity of mitochondria NADH-CoQ reductase and the pattern of mitochondrial distribution. Blastocysts were collected from mice cornua utery at day-4 of pregnancy and were divided into 3 groups: blastocysts undergo hatching within 24 hours, 48 hours and non hatching. Embryos were cultured in DMEM medium in 5% CO2 incubator at 37°C for 10 days. The trophoblasts monolayer were processed for Giemsa staining and histochemistry analysis of NADH-tetrazolium reductase activity. The outgrowth of trophoblast cells were measured using eyespiece micrometer. The results showed the ability of blastocyst hatching in in vitro were different. The trophoblast outgrowth diameter of 24 h hatched blastocyst was significantly different with the 48 h hatched and non hatched blastocyst. The activity of NADH-CoQ reductase of 24 h and 48 h hatched blastocysts showed higher intensity than the non hatched blastocysts (P<0,05) and the distribution of mitochondrial in trophoblast cell cytoplasma of 24 h and  48 h hatched blastocyst were homogen around nucleus whereas thoes of non hatched blastocyst were cluster and heterogen. In conclusion, in in vitro study the failure of blastocysts hatching and implantation was due to the failure of outgrowth and differentiation of the trofoblast cells and the impairment of NADH-CoQ reductase activity in complex I mitochondria to produce energy and the pattern of mitochondrial distribution.Key words: mitochondrial distribution, NADH- tetrazolium reductase, trophoblast, implantation

Abstract

Madura and Bali cattle are two of Indonesian indigenous breeds. One possible method to increase their productivity is through artificial insemination (AI) using frozen semen. This study conducted at the Singosari National Artificial Insemination Center, East Java, Indonesia, to evaluate the semen characteristics of different bull breeds ranging in age from 7 to 10 years and to provide useful recommendations regarding the efficacy of older cattle. A total of 2016 ejaculation data recorded from Indonesian indigenous cattle (Madura and Bali bulls) and Bos taurus bulls (Simmental) were used in this study to analyze semen volume, sperm motility, and sperm concentration. Results revealed differences in semen volume, sperm motility, and sperm concentration among bull breeds (p < 0.05). Simmental bulls have the highest semen volume (p < 0.05) compared with Bali and Madura bulls. Base on sperm motility, Bali bulls showed the highest average value (p < 0.05) compared with Simmental and Madura bulls. Sperm concentration was the highest (p < 0.05) among Bali bulls compared with Simmental and Madura bulls. Another important finding was that the average semen volume, sperm motility, and sperm concentration of older bulls fulfilled the standard criteria established for frozen semen processing. Therefore, it would be possible to improve the performance of these breeds through artificial insemination using frozen semen of Madura and Bali bulls, which can be more widely distributed to farmers to increase their herd productivity. This study also indicates that Indonesian indigenous bulls have superior semen characteristics than Simmental bulls.

Abstract

Sumatran rhino is the most endangered rhino species. Its population is estimated less than 300 individuals remaining in the wild with highly declining rate to 50% in the last 15 years. The number of male rhinoceroses in the captivity are very few, therefore the assessment of its fertility is very important in order to support the breeding success since the captive breeding success is very poor. The objectives of this study were (i) to determine the male reproductive status, (ii) to establish a reliable semen collection method, and (iii) to assess semen parameters of the fresh collected sample. Three methods of semen collection were examined to determine its fertilizing potential, i.e. (i) stimulated combination of artificial vagina (AV), penile massage (PM) and accessory gland massage (AGM); (ii) AV and PM; and (iii) only with PM. The first method gave the best result with an ejaculation success of 85.71% (6/7, n = 7). The second and third methods obtained an ejaculation success rate of 50% (2/4, n = 4)) and 25% (1/4, n = 4), respectively. The collected ejaculates had a volume of 1.2-12.4 ml with whitish to cream turbid colour and pH 6.90-6.99. Sperm concentration was (143-333) x 103 sperm/ml. The quality of the sperm was low with only approximately 1% of them moved forward slowly. Approximately 80% of the spermatozoa were immature (prox. cytoplasmic droplet) with head (macro-, microcephalic) and tail abnormalities (broken tail). Semen quality increased after several collections and the amount of immature sperm decreased up to 5%. Electroejaculation procedure could produce 34 ml semen, but no sperm was found in the ejaculate. Hence, the combination of AV, PM, and AGM could get higher volume of ejaculate compared to other methods, but sperm concentration was better obtained using AV and PM only. Repeated semen collection increased semen quality, although the male has low fertilizing capacity due to low sperm concentration (oligozoospermia) and small volume of the ejaculate (oligospermia). Abstract

Estrone conjugate (E1C) and pregnanediol glucuronide (PdG) were predominant steroid metabolites of estrogen and progesterone in feces of most primates and could be used to evaluate ovarian function. These metabolites were determined along with records of genital swelling throughout 3-4 months period from three female Javan Gibbons (Hylobates moloch) maintained in pairing-typed cage at Schmutzer Primate Center, Jakarta (Ullah) and at Taman Margasatwa Taman Sari, Bandung (Donna and Citah). Following methanolic extraction of lyophilized fecal powder, samples were analyzed using enzyme linked immunosorbent assay (ELISA) for E1C and PdG.  In all of the three females observed, both hormone profiles did not indicate any regular cycle of ovarian function even though genital swellings were sometimes observed. In one female (Donna) the hormone patterns showed clear signs of cycle irregularities with extended luteal phase of 40 days and erratic pattern of follicular phase. Of the other two females, no ovarian cycle was found. The data indicate that the fecal steroids analysis is a practical and valuable diagnostic tool for providing reliable information on ovarian function in Javan Gibbon. Factors affected reproductive hormonal profile should be taken in consideration in trying to achieve success in captive breeding program for this species.

Abstract

Premenopause is part of natural aging process in women indicated by the decrease of ovarial estrogen production, and usually followed by development of menopausal syndrome. Susumeno, skim milk fortified with 100 mg of soy isoflavone and 8 mg of Zn was given to premenopausal women for decreasing menopausal syndrome. This research aimed to evaluate the effect of susumeno on menopausal syndrome in premenopausal women. About 33 premenopausal women aged 43-52 years old in Purwokerto were choosen as respondent. Their menopausal syndrome were identified by using questioner. The answers were scored from 1 to 5 ; i.e., 1 = not detected; 2 = less detected; 3 = mildly detected; 4 = detected; 5 = much detected. It was revealed that some of menopausal syndromes had significantly decreased, especially vaginal dryness from less detected to not detected (p=0,01); fatigue from mildly detected to not detected (p=4,07E-05); mindless from mildly detected to not detected (p=0,022); and dyspareunia from less detected to not detected (p=0,036). Abstract

The objectives of this experiment were to evaluate the effects of isoflavone-riched soybean flour with different treatment level of isoflavone on MDA level, SOD activity, and profile of Cu,Zn-superoxide dismutase (Cu,Zn-SOD) spermatogenic cells in the seminiferous tubules of male rat testes. Diet was given as isonitrogen and isocaloric with 10% of dietary protein. Twenty five males of Sprague Dawley rats were divided into five groups and treated with isoflavone-riched soybean flour by oral administration with different concentrations. The treatment was done for 2 months. The optimum concentration of isoflavone was 1.5 mg/day and resulted in : decreased level of MDA testes (2.29±0.05 nmol/g), mantained high SOD activity of rat testes (729.4±23.73 U/g), and increased content of Cu,Zn-SOD by immunohistochemical technique on spermatocytes and early spermatids. It was concluded that isoflavone-riched soybean flour diet (1.5 mg/day isoflavone) can prevent the formation of lipid peroxide and improve the antioxidant status of rat testes.

Abstract

Pengukuran estron terkonjugasi (E1C) dan pregnanediol glucuronat (PdG) di feses dilakukan pada tiga ekor Owa Jawa (Hylobates moloch AUDEBERT 1797) betina untuk mengevaluasi fungsi ovarium ketiga owa tersebut yang masing-masing dipelihara di kandang individu di Pusat Studi Satwa Primata IPB dan di Taman Margasatwa Ragunan, Jakarta.. Contoh feses dikoleksi secara regular pada jam 07.00-09.00, 5-7 kali seminggu selama 4-9 bulan. Setelah melalui proses pengeringbekuan, contoh feses diekstraksi menggunakan metanol dan asai E1C dan PdG dilakukan dengan enzyme immunoassay (EIA). Profil hormon yang diperoleh menggambarkan adanya satu betina yang bersiklus dengan panjang siklus 21-25 hari. Lama fase folikular yang diperoleh pada betina tersebut bervariasi antara 11-18 hari, dan fase luteal yang relative konstan dengan lama 8-12 hari. Akan halnya dua betina yang lain, tidak diperoleh adanya gambaran pola yang menunjukkan siklus ovarium yang regular. Pembengkakan organ kelamin luar yang berfluktuasi juga hanya teramati pada betina yang bersiklus dengan lama pembengkakan 3-5 hari, dan perdarahan menstruasi terjadi selama 2-3 hari. Hasil yang diperoleh menunjukkan bahwa pengukuran steroid di feses merupakan suatu metoda nonivasif yang dapat diterapkan untuk mengevaluasi fungsi ovarium Owa Jawa. Keadaan fisiologis dari individual betina mungkin menjadi bahan pertimbangan lain dibandingkan tipe perkandangan sebagai faktor yang mempengaruhi profil hormonal.Kata kunci: Owa Jawa, steroid di feses, pembengkakan organ kelamin luar
