Abstract

The propagation plants coffee is carried out in generative manner and needs be optimized with treating gibberellin hormone and water temperature in germination stage of coffee seeds. This research aimed to know the effects of changes in the physical condition of coffee seeds with concentration of gibberellin hormone (GA3) and different water temperatures. This research conducted at the Seed Technology Laboratory Faculty Agiculture, Andalas University from July to September 2020. The method used was Randomized Complete Design with 3 replications. The parameters observed were seed weight early (g), seed weight after treatment (g), seed moisture content early (%), moisture content seed after treatment (%) and membrane leakage (μmhos). Results showed that 200 ppm of gibberellin hormone concentration can increase weight of seed after treatment with 4.28 g. The best seed moisture content was at 90°C water immersion temperature with 48.28%.

Abstract

Objectives of experiments were to evaluate (1) in vitro inhibitory effects of plant extracts on Clavibacter michiganensis subsp. michiganensis (Cmm), (2) inhibitory effects of plant extracts on Cmm infected tomato seeds, and (3) effectiveness of seed treatment plus plant extracts to eliminate Cmm. After evaluating 20 plant extracts, curcuma rhizome and betel vine leaf extract, cinnamon and clove oil were selected for further test. Tomato seeds were artificially inoculated with Cmm to obtain high level of infection. Part of the seeds were dipped in either suspension of selected extracts for 20 minutes and the others were matriconditioned using a mixture of burned rice hull (at 22 °C and RH 60-70%) plus either of plant extracts oils, respectively. Elimination of Cmm level from infected seeds was observed at 10 days after treatments. Results of the experiment indicated curcuma extract, betel vine extract, cinnamon oil or clove oil showed in vitro inhibitory effects on Cmm. Moreover, dipping infected seeds in either 5% of curcuma, betel vine extract, or 0.5% of clove oil or matriconditioning plus these extracts oils were effective to eliminate Cmm from infected seeds. These treatments may potentially be used and developed commercially for eliminating seedborne Cmm on infected tomato seeds.   Keywords: Bacterial cancer, seedborne-pathogen, seed-treatments

Abstract

Identifi kasi ketahanan genotipe adalah langkah awal dalam pengembangan kultivar tahan terhadap serangan patogen. Tujuan penelitian ini adalah (i) mendapatkan cara inokulasi dengan jumlah dan konsentrasi inokulum Cmm yang efektif untuk mengevaluasi ketahanan tomat terhadap Cmm di rumah kaca, (ii)  mendeterminasi reaksi ketahanan berbagai genotipe tomat akibat inokulasi Cmm. Percobaan ini menggunakan 38 genotipe tomat yang terdiri dari 7 genotipe tomat lokal, 15 genotipe tomat komersial, dan 16 genotipe koleksi Pusat Studi Pemuliaan Tanaman IPB Bogor (PSPT/IPB). Agen penyebab penyakit yang digunakan adalah 6 isolat Cmm hasil percobaan sebelumnya. Cara inokulasi Cmm yang efektif terhadap tomat cv. Marta (sangat rentan),  uji reaksi ketahanan berbagai genotipe tomat terhadap Cmm telah dilakukan di rumah kaca.  Kesimpulan dari penelitian ini adalah (i) inokulasi dengan menyuntikkan inokulum Cmm 5 µl konsentrasi 106 cfu/ml pada beberapa tempat di ketiak daun (daun pertama, daun tengah dan pucuk) merupakan cara yang paling efektif mengevaluasi ketahanan tomat terhadap Cmm, (ii) berbagai genotipe tomat yang diuji belum ada yang tahan teradap Cmm, genotipe tomat lokal ada yang agak rentan dan agak tahan.Kata kunci: kultivar tahan, suspensi bakteri, tomat

Abstract

Identifikasi dini terhadap jenis kelamin tanaman pepaya merupakan hal penting yang dapat membantu petani dalam budidaya tanaman pepaya. Identifikasi kelamin pepaya berdasarkan marka morfologi dan fisiologi telah dilakukan, namun beberapa hasilnya masih bias karena faktor lingkungan. Identifikasi kelamin tanaman pepaya menggunakan marka molekuler bisa lebih cepat dan akurat. Penelitian tersebut telah banyak dilakukan, salah satu di antaranya adalah marka berbasis sequence characterized amplified region (SCAR) dan beberapa primer SCAR telah dihasilkan untuk identifikasi kelamin pepaya. Penelitian bertujuan untuk menyeleksi primer SCAR yang efektif dalam mengidentifikasi seks tanaman pepaya. Penelitian  dilakukan pada bulan November 2018 sampai Juni 2019 di Laboratorium Molekuler dan Uji Mutu Kebun Percobaan Sumani Balai Penelitian Tanaman Buah Tropika di Solok. Primer SCAR yang diseleksi adalah W11,T12, PKBT5, Napf2, dan SDp. Tanaman referensi sebagai sampel umur 11–12 bulan  adalah tanaman betina, jantan, dan  hermaprodit masing-masing lima tanaman dari pepaya lokal dan Merah Delima. Hasil penelitian menunjukkan bahwa lima primer SCAR yang diuji  hanya dapat membedakan tanaman betina dengan tanaman jantan dan hermaprodit tetapi belum dapat membedakan antara tanaman jantan dengan hermaprodit. Konsistensi pola amplifikasi dihasilkan dari primer SCAR W11, Napf2, dan T12 dengan posisi 800 bp. Primer SCAR W11, Napf2, dan T12 selanjutnya dapat digunakan sebagai marka untuk identifikasi kelamin tanaman betina dengan tanaman jantan dan hermaprodit.KeywordsSCAR; Identifikasi; Pepaya; Jantan, Hermaprodit AbstractThe determination of sex expression of papaya plants is important to farmers in its cultivation. The identification of papaya plant sex based on morphological and physiological characters have been previously carried out, however, the results were still biased due to environmental factors. Many studies have been carried out to identify this plant sex, such as the use of molecular and SCAR markers, based on sequence characterization on amplified regions. This research aims to select the SCAR primers that are effective in identifying papaya plant sex. The study was conducted from November 2018 to June 2019, at Laboratory of Molecular and Quality Testing of the Indonesian Tropical Fruit Research Institute in Solok. The selected SCAR primers were W11, T12, PKBT5, Napf2, and SDp, using a total of five female, male, and hermaphrodite plants are reference aged 11–12 month from local papaya and cv. Merah Delima. The five SCAR primers tested were only able to differentiate females from male and hermaphrodite plants. The consistency of the amplification pattern was obtained from the SCAR W11, T12, and Napf2 primers at 800 bp. In conclusion, SCAR W11, Napf2, and T12 primers are used as markers to distinguish female plants from male and hermaphrodite.

Abstract

Tujuan dari kegiatan pengabdian pada masyarakat ini yaitu untuk memperkenalkan budiyaya pohon Aren (Arenga Pinnata Merr) dan manajemen serta organisasi usaha kelompok tani aren. Mitra yang menjadi subjek atalah petani aren di Nagari Persiapan Gaduik Barat, Kecamatan Tilatang Kamang, Kabupatan Agam, Sumatera Barat. Metode untuk memecahkan permasalahan dilakukan melalui penyuluhan budidaya aren dan manajemen serta organisasi usaha gula aren melalui kelompok tani. Penyuluhan dilakukan oleh tenaga ahli dibidang budidaya dari Universitas Andalas dan bidang manajemen dari Universitas Tarumanagara. Pelaksanaan penyuluhan dilakukan melalui pertemuan metode webinar dengan aplikasi Zoom Meeting. Para petani dikumpulkan di ruang pertemuan Kecamatan Kamang. Target luaran dari penyuluhan ini yaitu: (1) memberikan pengetahuan tentang Teknik budidaya pohon aren untuk meningkatkan produktivitas, dan (2) memberikan pengetahuan tentang manajemen dan organisasi usaha tani gula aren. Luaran yang dihasilkan berupa modul, publikasi, dan poster.

Abstract

The papaya plant has three types of flowering, male flowers, female flowers, and perfect flower (hermaphrodite). The difference in interest affects the shape of the resulting fruit. This study aimed to characterize morphology and cytology of papaya flowers. The study was conducted in Tropical Fruit  Research  Institute at Solok and Laboratory of SPT at the Biology Department of the Faculty of Mathematics and Natural Sciences, Andalas University, Padang, West Sumatra. The research material uses papaya flowers Merah Delima variety  from KP. Aripan Balitbu Troipka. The results showed the male flowers of male plants were smaller than the male flowers of the perfect plant. The difference of hermaphrodite flower of pentandria with hermaphrodite elongata flower, among others, the number of stamens, the length of the stem and the size of the ovary. Hermaphrodite pentandria has a number of stamens 5 while elongata has stamens 10

Abstract

The existence of Clavibacter michiganensis subsp. michiganensis (Cmm), the pathogen causing bacterial canker, on 22 lots of tomato seeds distributed commercially in Indonesia was evaluated. Isolation of suspected Cmm colonies were conducted by agar dilution plating on semiselective SCM medium followed by confirmation of colony identity on YDC medium. Identity of suspected Cmm colony was confirmed using IF test, pathogenicity test on tomato seedlings, hypersensitivity test on leaf of Mirabilis jalapa and Nicotiana tabacum, ELISA, and Cmm specific DNA amplification by PCR. After seed extraction and evaluation of the extract on semi-selective SCM medium and confirmastion by IF test, at least six tomato seed lots were contaminated with Cmm. After more confirmation using pathogenicity and hypersensitivity test, ELISA, and PCR amplification, at least three seed lots were confirmed possitively to carry Cmm.

Abstract

Bacterial canker, caused by Clavibacter michiganensis subsp. michiganensis is a newly introduced disease of tomatoes in Indonesia. Its existence was first officially reported in 2004. The objective of this research was to monitor the existence of C. michiganensis subsp. michiganensis in various tomato production centers in Sumatra and Java. Tomato samples showing symptoms of C. michiganensis subsp. michiganensis infection were collected from various tomato production centers in Sumatera and Java and the causal agents were isolated from these samples. Based on the occurrences of typical symptoms of C. michiganensis subsp. michiganensis infection in tomato, the incidence of suspected C. michiganensis subsp. Michiganensis infection ranged from 1-20%. From a total of 74 tomato plants sampled, 24 bacterial isolates exhibiting similar colony morphology to C. michiganensis subsp. michiganensis were obtained. After various physiological, hypersensitive response, and pathogenicity tests, 18 isolates derived from 14 tomato production centers in eight provinces in Sumatera and Java were identified as C. michiganensis subsp. michiganensis. Although the incidence was still low, results of these experiments further indicated that C. michiganensis subsp. michiganensis existed in Indonesia and had spread in a number of tomato production centers in Sumatra and Java. Positive results of C. michiganensis subsp. Michiganensis identification from suspected tomato samples should be a warning sign for all tomato production stakeholders in Indonesia, especially those responsible in regulating the seed trade, importation, and plant quarantine

Abstract

The objectives of the experiment were to evaluate effectiveness of seed treatments to eliminate Cmm of infected tomato seed lot, evaluate physiological quality of Cmm infected tomato seed lot after treatments with plant extracts. The research used curcuma rhizomes, betel vine leaf extract, cinnamon and clove oil to eliminate Cmm in tomato seed. Part of the seeds were dipped in either suspension of curcuma rhizomes, betel vine leaf extract, cinnamon and clove oil for 20 minutes and the others were matriconditioned using a mixture of burned rice hull (at 22 0C and RH 60-70%) plus either of plant extracts or oils. Elimination of Cmm from infected seeds was observed 10 days after treatments, and physiological seed qualities were evaluated before and after seed treatments. Results of the experiment indicated, dipping infected seeds in either 5% of curcuma, betel vine extract, cinnamon oil or 0.5% of clove oil suspension or matriconditioning plus either these extracts, eliminated 99% of infected Cmm on tomato seeds. Seed treatments using plant extracts for elimination of Cmm did not reduce seed germination, germination rate, vigor index, and did not increase time to reach 50% total germination (T50) of infected tomato seed except the seed treatment by using 5% cinnamon oil with or without matriconditioning.Key words: plant extracts, physiological quality, seed viability, vigor
