Abstract

Aim: This study investigated the potential effect of nanocurcumin on the increase of SOD enzyme expressionand decrease of caspase-3 in lead acetate-induced rats ovarian granulosa cells.Materials and Methods: Forty five female rats were divided into 5 groups, the negative control group (ratsreceiving corn oil, one hour later receiving distilled water), positive control group (rats receiving corn oil,one hour later receiving lead acetate of 40 mg/kg bw) and experimental groups 1, 2 and 3 (rats receivingnanocurcumin 50 mg, 100 mg and 200 mg/kg bw). One hour after administering the nanocurcumin, therats received 40 mg/kg of lead acetate. All groups received oral treatment once a day for 26 days. Onday 27 the rats were sacrificed and the expression of SOD and caspase-3 enzymes were measured usingimmunohistochemical methods.Results: This study found that lead acetate decreased SOD enzyme expression and increased caspase-3. Incontrast, nanocurcumin increased SOD enzyme and decreased caspase-3 expression in lead acetate-inducedrats ovarian granulosa cells.Conclusion: Nanocurcumin has potential as a strong natural antioxidant by affecting the increase of SODand the decrease of caspase-3 cells in lead acetate-induced rats ovarian granulosa cells.

Abstract

The antioxidants can be used for protective in oxidative stress that is one of the important mechanisms of 2-methoxyethanol (2-ME)-induced testis toxicity. The current study was carried out to evaluate The protective effect of xanthone via Malondialdehyde (MDA) and Superoxide Dismutase (SOD) expression on mice Sertoli cell induced by 2-Methoxyethanolon the Sertoli cell number induced by 2-ME in mice. The study used 35 male mice divided into 5 groups: control group (mice were given daily with water purified by distillation); 2-ME group (mice were given daily with 2-ME 200 mg/kg BW orally once in a day for 35 days); and the treatment group ( mice were given the xanthone 60 mg, 120 mg, and 240 mg/kg BW orally once in a day for 38 days, and on the 3th day, were given 2-ME 200 mg/kg BW one hour after the xanthone administration). After 38 days, the testis tissues were collected to evaluate the histological of Sertoli cell number, and also evaluated the immunohistochemical of MDA and SOD expression on Sertoli cell. The result showed that 2-ME administration significantly increased MDA expression, and decreased both SOD expression and the number of Sertoli cells. However, the treatment of xanthone significantly decreased MDA expression, and increased the expression of SOD of the Sertoli cell in the immunohistochemical. Xanthone also significantly increased the Sertoli cell number in histopathological evaluation. In conclusion, From the results of this study demonstrated that xanthone is able to protect Sertoli cell number in mice treated with 2-ME through decreasing MDA and increasing SOD.

Abstract

Red dragon fruit skin extract is used as an antioxidant to lead acetate toxicity. This study aimed to prove the difference in morphology of mice testicular spermatozoa exposed to lead acetate. Twenty-five mice were divided into 5 groups (5 mice per group). K- group was the control group without lead acetate and dragon fruit skin extract administration, K + group was treated with 8 mg/KgBW lead acetate administration for 14 days, P1 group was treated with the administration of 8 mg/KgBW lead acetate for 14 days + 250 mg/kgBW red dragon fruit skin extract for 21 days, P2 group was treated with the administration of 8 mg/KgBW lead acetate for 14 days + 500 mg/kgBW red dragon fruit skin extract for 21 days, and group P3 was a treatment group with the administration of 8 mg/KgBW lead acetate for 14 days + 1000 mg/kgBW red dragon fruit extract for 21 days. The results showed that there was significant difference (P<0.05) in normal sperm morphology among K-, K +, P1, P2, and P3. In conclusion, the administration of red dragon skin extract at a dose of 500 mg/kgBW (P2) is the optimal one which can be used as therapy to increase motility, morphology, and concentration of mice spermatozoa exposed to lead acetate.

Abstract

The purpose of this research is to prove that pare leaf extract (Momordica charantia L.) has anthelmintic activity against Raillietina sp. in vitro. This study consisted of four treatments and each treatment contained six replications, each of which consisted of six worms. The treatments in this research were K- with 1% CMC-Na, P1 with 2% extract concentration, P2 with 4% extract concentration, P3 with 8% extract concentration. Pare leaf extract has anthelmintic activity against Raillietina sp. in vitro with an effective dose of 8%. The higher the concentration of pare leaf extract, the higher the mortality rate of worms. The longer the immersion time, the higher the number of dead worms.

Abstract

The aim of this research is to know the effect of pomegranate (Punicagranatum L.) on the improvement of spermatozoa quality that is increase of percentage of capacitation and acrosome reaction in white rat (Rattusnorvegicus). This research used 20 healthy white rat (Rattusnorvegicus) with average age 2-3 months old, 200-300 g. Research design using completely randomized design (CRD) with four treatments and 5 replications. Rats were randomly divided into 4 treatment groups. Rat in the negative control group not treated just kept alone, solvent control group were given CMC Na 0,5% and is exposed to heat, the treatment groups 1, 2, and 3 induced pomegranate extract standardized to 40% ellagic acid at a dose of 75mg / kg, 150mg / kg and 300mg /kg perorally with doses 1 ml/rat/day gastric for along 14 days and exposed to heat. Data were analyzed using One Way Analysis of Variant (ANOVA) followed by Duncan (p<0,05). The result showed that there was significant effect from therapy on the quality of rats spermatozoa especially to capacitation and acrosome reaction. The treatment from treatment 2 group showed the best result in maintaining the quality of rats spermatozoa with 27.6± 3.04 values for capacitation and 6,80 ± 1,30 for acrosome reactions. Extract of pomegranate (Punicagranatum L.) standardized to 40% ellagic acid has a significant change in improve the quality of spermatozoa, namely an increase in the percentage of capacitation and acrosome reactions in white rats (Rattusnorvegicus) exposed to heat.

Abstract

The aims of this community dedication: 1) Improve the economy of the villagers of Watubonang and Biting, Badegan Ponorogo district through changing mindsets to improve people’s welfare 2) Increasing the knowledge and skills of the villagers of Watubonang and Biting, Badegan Ponorogo district through technology transfer for making pecticide formulations, antiscabies drugs, antiseptik, compost manure disinfectant, dipping solution based on tobacco waste. The result of the communiy dedication implementation showed an increase in participants on how to manufacture various kinds of tobacco waste products from 6.7% to 93.3% and an increase in the skills of participants on how to manufacture various kinds of tobacco waste products from 6.7% to 93.3%. in addition to the increased knwoledge and skills of the community dedication participants in Watubonang and Biting villages, Badegan Ponorogo district, it showed a decrease in the average cost of using medicines, antiseptiks, and disinfectants per house from 210 thousand rupiah to 42 thousand rupiah. Form the results of community dedication in the villages of Biting and Watubonang, Badegan Ponorogo district, it is hoped that they will become fostered villages of FKH Unair.
