Abstract

The development of the Internet of Vehicle (IoV) is overgrowing toward driving comfort, safety, and efficiency. Autopilot or a car without a driver is one implementation of IoT. To run the full function of IoV, a reliable communication network is needed because the risk will be substantial due to the low quality of network infrastructure services. In this research, we will analyze the quality of network infrastructure services. The analysis can be done through two approaches: direct measurement in the field using a GPS Tracker-based system and modeling using the mathematical function of transmit power of mobile communication transmitting devices. The test is carried out in a specific area with a sample of high traffic and low-density areas. This method was chosen to find the right pattern in measuring the quality of communication network services. The tests carried out produce dynamic data on service quality in certain areas. For areas of low density, the service quality tends to be below as well, and many areas without signal are found, while for areas with high density, the quality of service is found to be good, but network overload conditions often occur.

Abstract

The objective of this work is to clone flanking DNA derived from Tn-5 mutagenesis of wild type strain Xanthomonas axonopodis pv. glycines as first step to clone and to identify the gene involved in pathogenicity mechanism. We have localized the flanking DNA fragment from a nonpathogenic mutant of Xag M715. Southern hybridization analysis using 2.8 kb EcoRI from pYR103 as a probe showed that the fragment is located within 2.0 kb PstI fragment. A 0.7 kb flanking DNA was amplified using inverse PCR technique, and inserted into pGEM-T Easy vector generating a 3.7 kb recombinant plasmid (pAA01). Southern hybridization analysis of the wild type (YR32) with pAA01 as a probe indicated a hybridization signal located at approximately 3.0 kb PstI fragment. DNA sequence analysis revealed that the DNA fragment has a 64% identity to a vir gene of Bacillus anthracis.

Abstract

A rapid polymerase chain reaction (PCR)-based procedure was developed for detection of Xanthomonas axonopodis pv. glycines, the causal agent of bacterial pustule disease on soybean. A set of primers was designed from partial sequence of the pathogenicity gene of X. axonopodis pv. glycines strain YR32. Specific PCR product of 490 base pairs was produced from strains of X. axonopodis pv. glycines originally from Indonesia as well as from Taiwan. No other pathovars and bacterial species among those tested showed amplification product under optimized PCR conditions. Shaking infected soybean leaves in phosphate buffer saline during six hours was proved to be an essential in order to increase cell number of the bacterial. The procedure was applicable and reliable for detecting of pathogens in infected plant materials. The procedure was proved to be more effective than that of conventional detection and could be of great help for monitoring of pustule bacterial disease in the soybean fields. Key words: Xanthomonas axonopodis pv. glycines, bacterial pustule disease, rapid detection, PCR, specific primer

Abstract

Introduction turfgrass varieties require fertilization with high dosage, but it can contaminate environment.  The increasing  efficiency of fertilizer absorbtion was expected to reduce fertilizer requirement.  The experiment was conducted to study the effect of arbuscular mycorrhizae and Azospirillum sp. on nutrient uptake, fertilization efficiency, growth and visual quality of turfgrass.  The experiment consisted of two factors, i.e. the dosages of fertilizer and inoculant types.  The dosages of fertilizer were the relatif dosage from recommended dosage (RD) i.e 100%RD, 75%RD, 50%RD, and 25%RD.  The recommended dosage was 0.5 kg N + 1.5 kg P2O5 + 0.5 kg K2O per 100 m2/month by compound fertilizer (15-15-15).  The treatment of 100%RD without inoculant was used as control.  The inoculant types were no inoculant, arbuscular mycorrhizal fungi (AMF), Azospirillum sp., and AMF+Azospirillum sp.  Factorial experiment was arranged in Randomized Block Design with three replications.  The results showed that interaction between inoculant type and dosage of fertilizer affected nutrient uptake, fertilizer efficiency, growth, and visual quality.  AMF inoculation and 25%RD increased shoot N uptake and N fertilizer efficiency, compared with control.  Azospirillum sp. inoculation at 75% RD increased shoot N concentration, but did not affect on shoot N uptake and N fertilizer efficiency compared with control.  Inoculant did not affect shoot P concentration, shoot P uptake, and P fertilizer efficiency.  Azospirillum and AMF+Azospirillum inoculation increased shoot K concentration.  AMF+Azospirillum inoculation and 100%RD increased shoot N concentration and shoot N uptake, compared with control.    Key words : Arbuscular mycorrhizal fungi, Azospirillum, fertilizer efficiency, nutrient uptake, turfgrass.

Abstract

Geminivirus infecting pepper: symptom variation and transmission study Infection of geminiviruses has been reported to cause signz3cantyield loss on various crops. Polymerase chain reaction was able to detect geminivirus infection on pepper samples around Bogor and Cipanas, West Java. Symptom variation and transmission of the geminivirus infectingpepper was studied using five solanaceae plants with three dzgerent transmission mannec i.e. mechanical inoculation, side grafirng, and insect vector. The transmission study showed that mechanical inoculation was not able to transmit the eminivirus, while side grafting and insect vector, Bemisia tabaci, caused infection with symptoms varies from yellow mosaic, leaf curl leaf distortion, to stunting of the plant. Higher infection was observed on pepper var. Hot Chilli through B. tabaci (8009) and side grafting (71.4%) than those on chilli pepper and tomato. DNA fiagment of - 1.7 kb was amplified wing PCRfim those plants showing symptoms, but no DNA fiagment was observedfrom symptomless eggplant and tobacco var. White Burley. Thus it can be concluded that the last two kina3 ofplants were resistant to the geminivirus infectingpepper.

Abstract

Kota Banyumas dahulu ibu kota karesidenan Banyumas, menjadi pusat pemerintahan HindiaBelanda.
