Abstract

Black pepper plantation area in Lampung province (the biggest producer area within Indonesia) has decreased from time to time, which might be related to unfavourable environmental condition. The aim of this study was to assess which environmental factors are positively or negatively correlated with productivity in particular yield of two local varieties of black pepper plant (Natar 1 and Natar 2) in Lampung province.  Two observational studies were carried out in the present study i.e. phenological development and productivity of black pepper productivity in related to various environmental factors. Our results showed that productivity of two varieties were significantly affected rainfall, light intensity and micro humidity. Dry matter yield was positively correlated with rainfall, both in Natar 1 and Natar 2. It was concluded that among environmental factors affecting productivity of black pepper, rainfall intensity was the most influencing factor with a positive relationship with the productivity. Keywords: Black pepper, climate, cultivar, environment, Indonesia

Abstract

The Special Efforts Program (Upsus) is an Indonesian government program to increase production/productivity of lowland rice to support the realization of food self-sufficiency. The Upsus targets are to increase the planting index by 0.5 and productivity by 0.3 tons/ha/GKP. The increase in planting index is achieved by the expansion of added planting, but the increase in productivity has not been reached optimally due to pest and disease attacks. The objective of this research was to determine the knowledge, attitudes, and actions of Upsus farmers in controlling pests and diseases, rice cultivation techniques, and implementation of Upsus in Karawang District. The research was conducted by distributing questionnaires to 60 farmers in Tempuran and Tirtajaya Sub-District at Karawang District. The respondents were selected by purposive sampling with the consideration of respondent being an active farmer, having at least 5 years of farming experience, and has ben a member of Upsus for at least 1 year. Most respondents have age in the range of 41-60 years, the highest education of elementary school, and have farming experience for 5-10 years. Pests and diseases are the biggest problems faced by the farmers. Upsus farmers used Ciherang and Inpari 32 varieties and the applications of pesticides were conducted according to the schedule. Most of the farmers could not distinguish between pests and diseases. The dominant pest and disease control carried out was the use of chemical pesticides. Abstract

Studies on endophytic bacteria has been increasing recently due to its potency to promote plant growth and induce plant tolerance to pests and diseases. The research was undergone to isolate endophytic bacteria from different varieties of upland rice from various growing regions in Indonesia, followed by evaluation of their biocontrol and plant growth activities. Isolation of endophytic bacteria was based-on surface-sterilized method using alcohol and NaOCl on trypsic soy agar (TSA) media. More than 200 isolates was successfully isolated and the selected isolates were subjected to in vitro assay for their ability to produce antibiotic and promote plant growth. The result showed 41 isolates of endophytic bacteria had antibiosis activity to Rhizoctonia solani, 25 isolates to Pyricularia grisea on potato dextrose agar (PDA) medium and 8 isolates showed antibiosis activity to both R. solani and P. grisea on both PDA and TSA. The result of plant growth test, showed 12 isolates of endophytic bacteria induced the growth of root and shoot of rice plant. This indicated that some of the endophytic bacteria have the potency as biocontrol agents and inducer of plant growth.Key words: endophytic bacteria, Pyricularia grisea, Rhizoctonia solani, upland rice

Abstract

Yellow disease caused by plant-parasitic nematodes are still a major problem on pepper production area in Bangka region. Significant yield losses occurred due to yellow disease. The objective of this research was to survey the yellow disease incidence and to study farmer’s knowledge, attitude and practices on the management of yellow disease of black pepper in Bangka district, Central Bangka and South Bangka. This study consisted of a survey of the incidence of yellow disease in the field and interviews to the farmers. The result showed that the yellow disease had infected nearly all of black pepper fields in the areas with incidence of as much as 20–60%. Almost all of the respondents reported that the yellow disease was the major disease of black pepper in Bangka region. Farmers’ level of knowledge about the causal of disease, symptoms, ecology and control methods of the yellow disease are still very limited. Various control measures have been done by farmers to combat the disease, especially by removing and burning the infected plants or by application of lime. A few farmers did not even controlling the disease. Pesticides have been used by a few farmers to control the yellow disease, but the application of pesticide does not in line with integrated pest management principles.

Abstract

Rhoeo discolor has been known to have a good adaptation to various environmental conditions. This character might be due to mutualistic association with endophytic bacteria. The objective of this study was to isolate endophytic bacteria from roots of R. discolor and to evaluate their potency as biocontrol agents and plant growth promoters. The methods to isolate endophytic bacteria involved the following methods, sterilization of root surface, grinding of root tissues, dilution, and plating in the medium tryptone soya agar (TSA). A total of 21 isolates of endophytic bacteria were isolated from the roots of R. discolor. Based on hypersensitivity test on tobacco leaves, 19 isolates showed negative reaction (no necrosis symptom) and only 2 isolates showed positive reaction (necrosis was developed). The results on biocontrol and growth promoters assay showed that 7 isolates were able to inhibit the growth of Fusarium oxysporum under in vitro test and 12 isolates were able to increase the growth of rice seedlings.

Abstract

Root knot nematodes (RKN), Meloidogyne spp. is one of the most important rice pathogen in some regions in the world. Information of RKN on rice plants in West Java is very important because West Java is one of the largest national rice production center. The purpose of this study was to determine and identify the presence of Meloidogyne spp. on rice plants in West Java. Rice plant samples were taken from 7 rice-growing districts in West Java. Typical symptoms of infected rice plants by Meloidogyne spp. are stunting and the formation of root galls. RKN was found to infect rice plants in several areas in Bogor, Cirebon, and Sukabumi. All stadia of the RKN development which include eggs, second, third, and fourth stage jeveniles, adult females and males were found inside the rice root tissues based on nematode staining observation. Morphometric measurements of the body and stylet length, maximum body width, length of the second stage juveniles, and female perineal pattern, indicated that Meloidogyne species found was M. graminicola.

Abstract

Meloidogyne sp. is one of the main constraints of tomato production in Indonesia. Endophytic bacteria may be considered as biocontrol agents for controlling Meloidogyne sp. The objective of this study was to isolate endophytic bacteria from forestry plants and to evaluate its potential for controling Meloidogyne sp. on tomato. Endophytic bacteria were isolated from roots of mahoni (Swietenia mahogany), trambesi (Albizia saman), gaharu (Aquilaria malacensis), and meranti (Shorea sp.). Isolation of bacterial endophytes from plant tissue was conducted using surface sterilization method with 70% alcohol, 3% NaOCl and sterile water on medium trypsic soy agar. Endophytic bacteria was separated and purified based on shape and color of the colony. A total of 33 isolates of endophytic bacteria were isolated from roots of mahoni (11 isolates), trambesi (5 isolates), gaharu (7 isolates), and meranti (10 isolates). The bacteria was tested for the hypersensitivity reaction on tobacco plants and the result showed that 22 isolates did not cause necrosis, indicated they are not pathogenic. Ten isolates of endophytic bacteria was selected for further experiment, i.e. to evaluate their potential as biocontrol agent for Meloidogyne sp. and as growth promotor for tomato plants. The experiment was conducted in the screenhouse using seed treatment. The result showed that two isolates of endophytic bacteria, i.e. MSJ1H and AGS1F were able to increase the growth of tomato plants up to 60% and reduce the number of root knot caused by Meloidogyne sp. Endophytic bacteria isolated from forestry plants have the potential as a biocontrol agents to plant parasitic nematode Meloidogyne sp.

Abstract

Blast disease (Pyricularia oryzae) in Indonesia is initially known to cause problem on upland rice, but since 2000’s blast disease was also reported occurred on lowland rice. Application of endophytic fungi is very potential to be used as disease control method. This study was conducted to isolates endophytic fungi from lowland rice, and determine its capability to reduce  blast disease severity. Isolation of endophytic fungi was done from root, tiller, and the leaves of lowland rice.  Kencana Bali variety was used for in vivo inhibition test due to its most susceptible response against P. oryzae. Forty seven endophytic fungi isolates were obtained from Bogor, Sukabumi and Blitar. Based on colony morphology, endophytic fungi can be differentiated into 9 morphotype. Four out of fourteen endophytic fungi showed antibiosis activity in in vitro inhibition test against P. oryzae. Inhibition test conducted on Kencana Bali variety in the green house showed that four isolates was able to suppress blast disease development by 30-70%.
