Abstract

Pepper plants showing Begomovirus-like symtoms, consisting of yellowing, leaf curling, and distortion, werecollected from fields located in the Padang, West Sumatra. The aim of this research is to identification ofbegomovirus associated with yellow leaf curl diseases on pepper. Total DNA was extracted from infected leaftissue according to Doyle and Doyle (1999) with slight modification. Polymerase chain reaction (PCR) was used toamplify the coat protein region of the virus using universal degenerate primers pAV494 and pAC1048. The PCRamplified DNA product (approx. 560 bp) was sequenced. The nucleotide and amino acid sequences and BLASTsearch revealed highest homology with pepper yellow leaf curl Indonesia virus isolated pepper, tomato andAgeratum conyzoides from Java, but differences from those of tomato yellow leaf curl virus. The isolate was thententatively called pepper yellow leaf curl Indonesia virus-Padang (PYLCIV-Pdg).

Abstract

Rhizobacteria was reported to have a potential role as inducer of systemic resistance in several host plants. Five isolates of rhizobacteria (RbTD1-3, RbTD1-8, RbPdGN-3, RbAg1-5, RbLPK1-9) were isolated from rhizosphere of chillipepper plants in West Sumatera. Identification of rhizobacteria was conducted by polymerase chain reaction using primers 27F and 1525R which amplified 16S rRNA region. DNA fragment of 1.5 kb was succesfully amplified from 4 isolates (RbLPK1-9, RbPdGN-3, RbTD1-3 and RbTD1-8) and subjected to direct sequencing. Phylogenetic analysis based-on 16S rRNA sequences showed that isolate RbLPK1-9 has a close relationship with Pseudomonas fluorescen Biotipe G (92%), whereas isolates RbTD1-3, RbTD1-8 and RbPdGN-3 have a close relationship with Bacillus subtilis 10407 (57%).Key words: polymerase chain reaction, rhizobacteria, 16S rRNA

Abstract

Endophytic bacteria reside within plant hosts without causing disease symptoms. Bacterial endophytes promote plant growth and yield, suppress pathogens, may help to remove contaminants, solubilize phosphate, or contribute assimilable nitrogen to plants. The aim of this study were to obtain indigenous endophytic bacterial isolates, which have the ability to control bacterial pustule cause by Xanthomonas axonopodis pv. glycines and to increase growth and yield of soybean. The bacterial endophyte isolates were introduced as seed treatment (108 cfu/ml) and soil drench on 3 weeks old soybean seedling. Xanthomonas axonopodis pv. glycines were inoculated on one month old of soybean by pricking leaf methodes. The effect of bacterial endophyte on disease incidence, disease severity, plant growth and yield of soybean were evaluated. We have found two selected bacterial endophyte isolates from soybean’s rhizosphere (St4E1.1 and St1E1.1.), with the ability to control bacterial pustule and to increase growth and yield of soybean.Key words: bacterial endophyte, bacterial pustule, in planta technique, soybean

Abstract

Tobacco leaf curl disease (“kerupuk”) is a new disease on tobacco in West Sumatra. This disease has spread throughout the tobacco planting area in District Limapuluh Kota with intensity of 2.5–26.3%. DNA fragment of 1600 bp was successfully amplified using polymerase chain reaction method with primer pairs for Begomovirus PAL1v 1978/PAR1c 715 from tobacco leaves showing symptoms. This result indicated Begomovirus infection. Sequence analysis based on NCBI BLAST program showed that Begomovirus infecting tobacco in West Sumatera has the highest homology of 89% and 91–97% with Pepper yellow leaf curl Indonesia Virus infecting tomato and chili pepper in West Sumatera, respectively. This is considered as the first report of Begomovirus infection on tobacco in West Sumatra.

Abstract

Plant rhizosphere is rich of microbes, one of these is a group of fungi. This research aimed to obtain fungi isolates from rhizosphere of healthy Chrysanthemum, followed by evaluation of their potential as biocontrol agents of Fusarium oxysporum. Isolation of fungi rhizosphere was based-on serial dilution method. As many as 6 isolates of rhizosphere fungi was succesfully isolated with diverse characters. Pathogenicity test showed that 6 isolates were not pathogenic because it did not cause wilt symptoms on Chrysanthemum. The selected isolates were subjected to in vitro assay for their potential as biocontrol agents, 4 isolates had antagonist ability (46.42–51.61%) and antibiosis activity against F. oxysporum. Fungi isolates having the potential as biocontrol agents was identified as Trichoderma sp. (2 isolates), Penicillium sp., and Paecillomyces sp.

Abstract

Vascular streak dieback (VSD) symptoms was reported recently in several cacao plantations in West Sumatera.  Disease incidence reached 58.82–100% with disease intensity of 24.29–44.71%.  In some cases, dead plant was also found. Fungal isolation was performed to identify the agents associated with VSD.  Plant samples showing VSD symptoms was collected from 3 locations of cacao production center in West Sumatera, i.e. Limapuluh Kota regency, Padang Pariaman regency, and Padang city.  Small pieces of leaf and twig were plated on water agar and potato dextrose agar medium for fungal isolation.  Morphology of hifa, basidiocarp, and basidiospora observed from fungi colonies indicated the presence of Ceratobasidium theobromae on infected plant samples.  This is the first report on the association of C. theobromae on cacao in West Sumatera.

Abstract

Vascular streak dieback (VSD) is a deadly disease of cocoa because it attacks the vascular tissue growing point. RGBGP Technology (Root Growth Based Growth Plants) manure is designed to help control the disease while helping grow cocoa plants. This research aims to examine the behavior of cocoa farmers in applying RGBGP technology package of manure that is allegedly formed due to individual characteristics and innovation characteristics. This research was designed descriptively qualitative located in Limapuluh Kota District. Sampling was conducted by census, 43 people belonging to 4 selected farmer groups, namely Aroma, Inovasi, Buah Lobek, and Maju Sejahtera. The results show that farmers' behavior generally shows the willingness to implement RGBGP package of manure in the long term, supported by the relative advantages of innovation, accessible resources, easy to do and applied technology, observable results, and no significant constraints. However, attitudes are not related to individual characteristics of farmers consisting of formal education, nonformal education, experience of farming, land status, land area, main occupation, and gender.

Abstract

The use of rhizobacteria is an alternative control in suppressing plant disease attacks. The purpose of this study was to obtain indigenous rhizobacteria which have the potential as antagonist agents of Fusarium oxysporum f. sp. cubense. Sampling was carried out randomly selected on endemic lands. Isolation of rhizobacteria using serial dilution method and its antagonist test against Fusarium oxysporum f. sp. cubense uses the dual culture method. The isolation results from 3 banana cultivars (kepok, raja sere, and mas) obtained 24 rhizobacterial isolates with different morphological and physiological characters. The antagonist test results showed that 8 rhizobacterial isolates were able to suppress the growth of Fusarium oxysporum f. sp. cubense and is not plant pathogenic.

Abstract

Teknologi BP3T (Bakteri Perakaran Pemacu Pertumbuhan Tanaman) pupuk kandang dirancang untuk membantu mengendalikan penyakit VSD (vascular streak dieback) yang menyerang tanaman kakao sekaligus membantu pertumbuhan tanaman. Penetian ini bertujuan menganalisis perbandingan biaya penggunaan pupuk kandang konvensional dengan teknologi BP3T Pupuk Kandang. Penelitian ini dirancang secara deskriptif kuantitatif yang berlokasi di Kabupaten Limapuluh Kota. Pengambilan sampel dilakukan secara sensus, yaitu 43 orang yang tergabung ke dalam 4 kelompok tani terpilih, yaitu Kelompok Tani Aroma, Kelompok Tani Inovasi, Kelompok Tani Buah Lobek, dan Kelompok Tani Maju Sejahtera. Biaya pupuk yang dikeluarkan petani sebelum menggunakan pupuk Formula BP3T lebih besar dibandingan dengan biaya penggunaan pupuk Formula BP3T. Dari kesimpulan yang ada, disaran kepada petani untuk menggunakan pupuk Formula BP3T dan mengembangkannya. Selain biayanya murah, pupuk Formula BP3T juga memilikik manfaat untuk meningkatkan ketahanan tanaman terhadap penyakit VSD.

Abstract

Yellow leaf curl disease of pepper has become an emerging important disease in West Sumatra since early 2000. Several attempts have been made, including disease survey and detection, in order to identify the causal agent of the disease. Pepper (Capsicum annuum) plants showing yellow leaf curl from West Sumatra were analyzed for presence of Begomovirus employing Polymerase Chain Reaction (PCR) with degenerate primers pAL1v 1978 and pARc 715. A DNA fragment of 1.6 kb was successfully amplified and subjected to direct sequencing. A stem loop region was found in the nucleotide sequence obtained, which contains the conserved nucleotide signature sequence TAATATTAC present in begomoviruses. Based on the stem loop region comparisons and phylogenetic analysis, the virus isolates from West Sumatra showed the closest relationship to Pepper yellow leaf curl Indonesia virus (PYLVIV) and Tomato yellow leaf curl Indonesia virus (TYLCIV). The sequence was different from other Asia Begomoviruses reported earlier. These isolates were divided into three groups which were tentatively called Pepper yellow leaf curl Indonesia virus-West Sumatra-[group 1], -[group 2] and -[group 3] {PYLCIV-WS-[group1], -[group2], and -[group3]}.
