Abstract

Totally 15 isolates of endophytic fungi were obtained from leaves and young stems of a medicinal plant Albertisia papuana Becc.(Menispermaceae). The antibacterial screening of the ethyl acetate extract derived from the fungi cultures in potato dextrose broth (PDB)were performed on a non eluted Thin Layer Chromatography Bioautography assay (TLC bioautography assay). From the screening test, itwas found that the ethyl acetate extract of the fungus Xylaria sp. DAP-KRI-5 culture showed strong antibacterial activity against Staphylococcus aureus. The isolation of active metabolite based on the bioautography guided assay from ethyl acetate extract of the Xylaria sp. DAP-KRI-5, led us to identify phloroglucinol as a main antibacterial compound. The chemical structure of phloroglucinol was deduced from it spectral data, including UV-Vis, 1H and 13C-NMR, GC-MS and published data elsewhere.

Abstract

Peroxidase has been successfully extracted and partially purified from Brassica juncea L. Czern root with protein content and specific activity of 0.747 mg/mL and 0.244 U/mg, respectively. Then, it was applied as biocatalyst for green oxidative coupling reaction of eugenol to produce an orange crystal, dimeric form of eugenol (84% yield). Structure elucidation was done by using FTIR, UV-Vis and GC-MS instruments. The optical activity of dimeric eugenol was found to have Ra-configuration. Furthermore, biological activity of the product as antioxidant has been evaluated resulting in IC50 values of 32.14 and 42.43 ppm for dimeric and monomeric eugenol, respectively.

Abstract

Green chemistry aspect is the chemical approach that has been studied in the past two decade. One of the principles is the development of green synthesis process that is friendly for the environment. This research showed that papain can be used as catalyst for Knoevenagel reaction with 3 kinds of substituted-benzaldehyde and malononitrile as substrates in aqueous medium. The best reaction condition with 80% yield was reached by utilizing of 25 mg papain/mmol substrate. Reaction was conducted at ambient temperature and pressure for 30 min. Products were as yellowish to yellow needle crystals and successfully characterized by melting point, UV-Vis, IR, mass spectra, and 13C & 1H-NMR, named as 2-(4-hydroxybenzylidene)-malononitrile); 2-(3-hydroxybenzylidene)-malononitrile); and 2-(4-hydroxy-3-methoxybenzylidene)-malononitrile).
