Abstract

Tujuan penelitian adalah untuk mengetahui dan menjelaskan pengaruh dari ektrak kulit buah rambutan terhadap adipogenesis jaringan lemak viserall tikus dengan melihat ekspresi igf-1. Tahapan penelitian yang dilakukan adalah ekstraksi kulit buah rambutan, perlakuan pada tikus, isolasi protein lemak viserall, separasi protein dan western blot. Tikus strain wistar jantan dibagi menjadi tikus berat badan normal dan obesitas. Perlakuan yang dilakukan antara lain kontrol, asam elagat, placebo, dan perlakuan ekstrak dengan dosis 5, 10, 15, dan 20 mg/kg berat badan. Hasil menunjukkan bahwa tren penurunan berat badan terjadi pada grup tikus obesitas dengan asupan pakan yang tidak berbeda signifikan (P>0,05). Dosis efektif terlihat pada perlakuan 10 mg/kg dan pada dosis 20mg/kg justru terjadi kenaikan berat badan untuk kedua jenis grup tikus. Profil protein antara tikus normal dan obesitas menunjukkan pola yang hampir sama dengan perbedaan intensitas tiap pita. Ekspresi igf-1 terpaut dengan ekspresi igfbp-1 pada 36,7 kDa dan terjadi pada seluruh perlakuan dengan perbedaan nyata pada kedua grup. Ekstrak kulit buah rambutan dengan dosis efektif 10mg/kg memiliki efek anti-obesitas pada tikus obesitas. Menariknya, ekspresi igf-1 menurun pada perlakuan 20mg/kg berat badan dibandingkan dengan kelompok lain. Hal ini diduga penghambatan obesitas tidak melewati jalur tyrosin kinase pada IR family.

Abstract

Rambutan is a tropical fruit that grow well in Indonesia and the peel is considered as waste. Many researchers’ showed that rambutan peel contains polyphenol that could be expected to avoid obesity.  The objective of this study was to explore the increasing production of local rambutan and to identify the promising phytochemical compounds on its peel as phytopharmaca candidate against obesity. Survey was conducted on the production of rambutan, potential plantation area, and marketing. Sample of rambutan peel collected from the sub-district Kanigoro, Blitar. Phytochemical compounds were analyzed using TLC, HPLC and FT-IR. Bioassay analysis used obesity rat models. The survey result showed a mean of rambutan production increased 2,6% in 2007-2012. Average production of rambutan 70-120 kg/tree. Vegetative multiplication usually done to maintenance of rambutan quality. The main compound of  Rambutan peel  extract (RPE) is flavonoids, tannins, ellagic acid and the major functional group of CH3, aliphatic CH3, and C=O. These compounds have a potential activity against obesity.  RPE 30 mg/kgBW dose was significantly inhibit the weight gain of obese rats and reducing the adipocyte size (p<0.05).Key words: potency, production, local rambutan, blitar, obesity

Abstract

Obesitas saat ini disebut sebagai “sindrom baru dunia” yang memerlukan terapi untuk mengatasinya. Terapi herbal adalah terapi yang paling banyak dilakukan penderita obesitas. Kulit buah rambutan mengandung polifenol yangmempunyai aktivitas antioksidan dan berpotensi untuk dikembangkan sebagai obat terapi obesitas. Hepar dapat terkena efek samping obat karena bertanggung jawab melakukan metabolisme obat, termasuk karena pemberian ekstrak kulit buah rambutan. Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak kulit rambutan (Nephelium lappaceum L.) terhadap nekrosis sel hepar tikus (Ratus norvegicus) obesitas. Penelitian ini merupakan penelitian eksperimental rancangan acak lengkap (RAL) dengan 4 perlakuan. Dua puluh empat tikus jantan strain wistar umur 14 minggu yang mengalami obesitas dikelompokkan secara acak menjadi 4 kelompok, diberi ekstrak kulit buah rambutan melalui sonde lambung dengan dosis 15, 30, dan 60 mg/kgbb dan kelompok kontrol diberi akuades. Setelah 12 minggu, tikus didekapitasi dan dikoleksi organ heparnya untuk dibuatpreparat histologi dengan pewarnaan Hemotoksilin-Eosin (HE). Tiap sayatan diamati dalam 3 lapang pandang, diamati 100 sel hepatosit, dihitung persentase sel yang mengalami nekrosis menggunakan hand tally counter. Analisisdata menggunakan analisis varian tunggal pada taraf kepercayaan 95%, dilanjutkan dengan uji BNT dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa ekstrak kulit buah rambutan yang berpengaruh signifian terhadap nekrosis sel hepar tikus adalah 60 mg/kgbb, yaitu meningkatkan persentase piknosis, karyoreksis, karyolisis, serta total nekrosis

Abstract

Penyakit jantung koroner (PJK) terutama disebabkan oleh aterosklerosis karena hiperkolesterolimia. Oksidasi low density lipoprotein (LDL) oleh reactive oxygen species (ROS) sebagai penyebab utama proses aterogenik dapat dicegah dengan kehadiran antioksidan seperti vitamin E. Buah dan sayuran banyak mengandung vitamin. Salah satu buah yang mengandung vitamin E adalah buah merah (Pandanus conoideus Lam.). Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak buah merah terhadap kadar LDL darah pada tikus putih (Rattus norvegicus L.). Jenis penelitian ini adalah penelitian eksperimental laboratorium yang didesain mengikuti Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 4 kali ulangan dengan parameter penelitian adalah kadar LDL darah tikus putih. Data dianalisis menggunakan one way anova dilanjutkan dengan uji Duncan 95%. Berdasarkan hasil penelitian, minyak buah merah memiliki kemampuan untuk menurunkan kadar LDL darah tikus putih.

Abstract

The purpose of this study is to examine the effect of Good Corpotare Governance’s mechanism that consist of Board of Directors, Independent Commissioner, Manajerial Ownership, and Institusional Ownership on the existence of Enterprise Risk Management and the effect of the Enterprise Risk Management on firm performance. In addition, this study also examines the intervening role of Enterprise Risk Management on the relationship between Corporate Governance and firm performance. The population of this study was manufacturing companies listed in Indonesia Stock Exchange for the 2012 to 2014 financial year. By using purposive sampling method, 25 firms were selected as a sample. This study used data from company’s annual report. The analysis technique used multiple linear regression analysis using SPSS computer program. The results of analysis showed that Board of Directors, Independent Commissioner, Manajerial Ownership, and Institusional Ownership affect the existence of Enterprise Risk Management and the existence Enterprise Risk Management affects firm performance. The result also proved that Enterprise Risk Management act as an intervening variable among Board of Directors, Independent Commissioner, Manajerial Ownership, and Institusional Ownership on firm performance.Keyword : Good Corporate Governance, Enterprise Risk Management and Firm Performance

Abstract

ABSTRAK Bawang putih tunggal telah banyak digunakan masyarakat sebagai obat tradisional salah satunya untuk meningkatkan stamina. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak bawang putih tunggal sebagai imunostimulan melalui pengujian aktivitas dan kapasitas fagositosis makrofag serta berat limpa mencit (Mus musculus) yang diinduksi dengan bakteri Escherichia coli. Mencit jantan (Balb-C, umur 12 minggu, BB 20 ± 5 g) diberi ekstrak bawang putih tunggal selama 28 hari berturut-turut dengan dosis ekstrak bawang putih tunggal N 0%, P1 0,25%, P2 0,5%, P3 1%, dan P4 2%. Pada hari ke 29, mencit diinduksi dengan bakteri E.coli yang diberikan secara intraperitoneal (1 ml, 1,5 x 105 cfu/ml). Aktivitas fagositosis makrofag dihitung dari sediaan apusan cairan peritoneum. Ekstrak bawang putih tunggal meningkatkan jumlah dan kapasitas makrofag aktif secara nyata pada perlakuan P2, P3, dan P4 (p=0,000;α=0,05) dengan berat limpa yang berbeda secara signifikan (p=0,000;α=0,05). Hasil penelitian menunjukkan semakin tinggi ekstrak bawang putih tunggal yang diberikan maka semakin tinggi pula aktivitas dan kapasitas fagositosis makrofagnya dengan berat limpa yang semakin kecil. Kata kunci: bawang putih tunggal, fagositosis makrofag, imunostimulan. ABSTRACT Single garlic has been used as traditional medicine for increasing stamina. This research aim to knew the effect of single garlic as immunostimulant through activity and capacity fagocyte of macrophag test and mice weight spleen that induced by E.coli. Male mice (Balb-C, 12 weeks old, weight 20 ± 5 g) has been given sigle garlic extract for 28th days with various dosage (N 0%, P1 0,25%, P2 0,5%, P3 1%, and P4 2%). on day 29, mice ware induced by E.coli through intraperitoneal injection (1 ml, 1,5 x 105 cfu/ml). Fagocyte activity of macrophag counting by peritoneum liquid smear. Single garlic extract increase amount and capacity active macrophage in P2, P3, and P4 (p=0,000;α=0,05) with significant various spleen weight (p=0,000;α=0,05). The result show that the higher sigle garlic extract can make higer activity and capacity fagocyte of macrophag with lower spleen weight. Keywords: single garlic, macrophag, immunostimulant.

Abstract

Staphylococcus aureus is gram-positive bacteria that often infect the skin. S. aureus has been experiencing resistance to several antibiotics. One of the solutions to overcome the resistance is by using garlic that is believed by the society can overcome bacterial infection. The study aimed to find out the influence of the variation in concentration of local single clove garlic essential oil used on the inhibition and damage of morphological structure of S. aureus bacteria. The inhibition test was conducted using disc diffusion method. The experiment groups consisted of  1% DMSO as negative control, vancomycin 30µg/ml as positive control and single clove garlic essential oil (25 mg/ml, 50 mg/ml, 75 mg/ml and100 mg/ml) as treatment groups. The diameter of inhibition zone was measured using calipers. The morphological damage of the bacterial cells can be seen using Scanning Electron Microscopy (SEM) with magnification of 25000x. The result of Kruskal-Wallis test analysis indicated that the extract of local garlic essential oil has inhibitory activities against S. aureus bacteria (P<0.05). The damage to the morphological structure of bacterial cells with the administration of 30 µg/ml vancomycin was equal to 100 mg/ml single clove garlic essential oil extract. Single clove garlic essential oil can be used as an alternative treatment for skin infection diseases by inhibiting S. aureus growth.

Abstract

 Atherosclerosis is initiated by inflammatory response characterized by the accumulation of lipids in the arteries, causing blockage of blood vessels. The excessive of low-density lipoprotein (LDL) in the blood is a major factor in endothelial dysfunction and results in an inflammatory process. The inflammatory process in atherosclerosis is due to the expression of adhesion molecules, vascular cell adhesion molecules-1 (VCAM-1). This research aimed to evaluate the effect of single bulb garlic oil extract (SGBOE) against VCAM-1 expression in high-fat diet mice. Thirty male mice strain Balb/C were acclimatized for one week. Mice were divided into 6 groups (n=5): normal, high-fat diet (HFD), HFD + simvastatin, and HFD + SBGOE 12.5, 25, and 50 mg/kg BW respectively. HFD was given for 45 days. The SGBOE was given for four weeks orally. The aortic VCAM-1 expression was assayed using the immunohistochemical-flouresencemethod. The result of One-Way ANOVA showed that the treatment influenced significantly (p<0.05). The foam cells increase in the high-fat diet group compared than normal mice. Dose 12,5 mg/kg BW reduces VCAM-1 expression near the normal group. Allicin and S-allyl cysteine (SAC) compounds in single bulb garlic are antioxidant compounds that act to prevent oxidative stress and prevention mechanisms against lipoprotein modification.

Abstract

BackgroundHigh-fat diet (HFD) is highly responsible for the development of excessive reactive oxygen species (ROS). ROS and low-density lipoprotein (LDL) then trigger macrophage activation to secrete interleukin-6 (IL-6). Single bulb garlic or in Indonesian called bawang lanang is traditional medicine that possesses strong antioxidant properties. This study aimed to evaluate the effect of single bulb garlic oil (SBGO) on ROS, IL-6, and lymphocyte density in HFD-fed male mice.MethodsThis was an experimental study on 24 male mice randomly subdivided into 6 groups: one group was fed a normal diet, whereas the remaining five groups were fed HFD for 45 days and were then treated with single bulb garlic oil 0 mg/kg, simvastatin 26 mg/kg BW, single bulb garlic oil 12.5 mg/kg BW, 25 mg/kg BW, and 50 mg/kg BW respectively for the next 35 days. At the end of treatment, the mice were dissected for collection of (i) serum in order to measure ROS and IL-6 levels using ELISA, (ii) aortas to measure IL-6 expression using immunohistochemistry-fluorescence (IHC-F) and to obtain lymphocytes from bone marrow and spleen which were then counted using a light microscope.ResultsOur results indicated that SBGO decreased the ROS level, IL-6 level, IL-6 expression, and lymphocyte density in HFD mice. SBGO 12.5 mg/kg BW is the optimum dose in our study to reduce inflammation in HFD male mice.ConclusionSBGO is useful to reduce inflammation and improve antioxidant imbalance, thus might be a potential antiatherogenic agent.
