Abstract

Tea (Camellia sinensis) is an agricultural product that is widely issued by people around the world. Some types of tea in the world are green tea, black tea, oolong tea and white tea. Kombucha tea is brewed water for tea and sugar then given bacteria and fungi and fermented for 7-10 days. The purpose of this study was to determine the quality of kombucha of several different types and determine the organoleptic value of fermented kombucha tea using different types. This research was conducted in April-July 2019 in the Microbiology Laboratory of the Department of Biology FMIPA Padang State University. This research is an experimental study with 5 treatments and 1 repetition (K: kombucha starter, P1: kombucha starter + green tea, P2: kombucha starter + black tea, P3: kombucha starter + oolong tea, P4: kombucha starter + white tea. Parameters Calculated are the number of microbes, nata thickness, pH of kombucha tea, total acetic acid, and organoleptic tests of kombucha tea.The results of this study indicate differences in the types of tea used in kombucha fermentation, the quality of kombucha tea produced, kombucha tea using black tea. The organoleptic test requested by the panelists was kombucha tea from white tea.

Abstract

Thermophilic bacteria are bacteria that can produce thermostable enzymes and used in various industries. Thermostable enzymes that can be produced by thermophilic microorganisms one of them is xylanase enzyme. Xylanase produced by microbes has optimum temperature characteristics and more diverse optimum pH on various substrates, which will affect the activity of the resulting xylanase enzyme. The influence of temperature greatly determines the activity of the enzyme at the time of catalyzing a reaction. At optimum pH conditions, the enzyme has an active side conformation that is substrate-like so that it can form a complex of appropriate enzymes and produce the product to its full potential. One of the thermophilic bacterial habitats of hot springs in West Sumatera is the Aro Sapan River hot spring located in Koto Parik Gadang Subdistrict in Ateh, South Solok District. The Saw Aro River hot spring has a temperature of 75 ° C and is pH 8 or alkaline. The aim to this research was to know the profile of the growth of thermophilic bacteria. This research is a descriptive method. The data obtained were analyzed descriptively. The conclusion is the best activity of xylanase thermophilic bacteria was at the sixth time incubation.

Abstract

 Abstract -- Endophytic bacteria are microorganisms that exist in the system of plant tissues such as leaves, twigs, roots and they can form colonies without causing damage to the plant. The endophytic bacteria may produce secondary metabolites in accordance. One potential medicinal plant has endophytic bacteria are plant greeting (Syzygium polyanthum Wight). Part of the plant Salam particularly frequently used is part of the leaf, which serves as an antibacterial and antifungal. Because the leaves contain compounds bay leaf tannins, flavonoids and essential oils. The porpuse of this research was to find of to obtain isolates of endophytic bacteria and determine the antimicrobial activity of bay leaf (Syzygium polyanthum Wight).This research is descriptive that has done on January - February 2015 of Microbiology Laboratory at Biology Departement FMIPA, UNP. The parameters observed in the determination of isolates is a form colonies, colonies edge, elevation colonies, colony color, and Gram staining of  leaves (Syzygium polyanthum Wight). The antimicrobial activity was observed with the formation zone of inhibition around the paper disc. Obtained were 11 isolates of endophytic bacteria from leaves, 5 isolates of endophytic bacteria have the ability to produce antimicrobial.Isolates had the highest antimicrobial activity that isolates DSA 1-1 with a 6.42 mm diameter inhibition zone. Keywords: antimicrobial, endophytic bacteria, bay leaf.

Abstract

Biogas is a gas produced from the decomposition process of organic materials by microorganisms in anaerobic conditions. Waste of red onion (Allium cepa L.) and buffalo dung contain organic substances and nutrients that can be used as biogas. The purpose of this study was to identify fungi in biogas mixed with buffalo dung with scallion waste (Allium cepa L.).The study was conducted from April to September 2019 in the Microbiology Laboratory and Integrated Research Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, State University. This research is a descriptive study by observing macroscopic and microscopic characters as a basis for identification produced by fungi in buffalo dung biogas (Pariaman, West Sumatra) and onion leaf waste from Alahan Panjang, West Sumatra.The results of this study obtained 5 genus of fungi on biogas substrate from a mixture of buffalo dung with scallion waste (Allium cepa L.) including: Aspergillus, Trichoderma, Candida, Mucor and Humicolla. Biogas adalah gas yang dihasilkan dari proses penguraian bahan-bahan organik oleh mikroorganisme dalam keadaan anaerob. Limbah daun bawang merah (Allium cepa L.) dan kotoran kerbau mengandung zat organik dan unsur hara yang dapat dijadikan sebagai biogas. Tujuan penelitian ini untuk mengidentifikasi jamur pada biogas campuran kotoran kerbau dengan limbah daun bawang merah (Allium cepa L.). Penelitian dilaksanakan dari bulan April sampai bulan September 2019 di Laboratorium Mikrobiologi dan Laboratorium Penelitian Terpadu Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Negeri. Penelitian ini merupakan penelitian deskriptif dengan mengamati karakter makroskopis dan  mikroskopis  sebagai dasar identifikasi yang dihasilkan oleh jamur pada biogas kotoran kerbau (Pariaman, sumatera barat ) dan limbah daun bawang merah dari alahan panjang, sumatera barat. Hasil dari penelitian ini diperoleh 5 genus jamur  pada substrat biogas dari campuran kotoran kerbau dengan limbah daun bawang merah (Allium cepa L.) diantaranya yaitu: Aspergillus, Trichoderma, Candida, Mucor dan Humicolla.

Abstract

Elementary students have high playing activities while in school. Hot weather and feeling tired after playing, will cause thirst and to get rid of thirst Elementary school students will buy ice-based snacks. They do not realize, the danger of unhygienic snacks will cause diarrheal diseases, because ice cubes are made by home industries still traditionally without considering hygiene requirements. In some countries, the food industry has moved towards testing Enterobacteriaceae which shows enteric bacterial contamination as a condition hygiene. This study aimed to determine the microbiological hygiene quality of the Enterobacteriaceae numbers in ice-based school children snacks in Koto Tangah Subdistrict, Padang City. This study was a descriptive study. The study was conducted from April to May 2019 at the Laboratory of Microbiology at the Center for Drug and Food Control in Padang. Total samples taken were 20 samples. The results showed that the sample that fulfilled the microbiological quality requirements for sadness was 60% (12 samples) and 40% (8 samples) that did not meet the requirements, the highest results were obtained at SDN 44 Sungai Lareh with a value of 9.82 x 105 colonies/mL.

Abstract

The 2013 curriculum encourages students to be more active and independent when getting information about learning materials. Therefore, teachers are required to be able to increase students' learning motivation, one of which is by developing learning media that can attract students' learning interest. The purpose of this study was to see the needs of students for learning media with ESQ nuances about bacteria as a supplement to biology teaching materials for class X science SMAN 2 Payakumbuh. This type of research is descriptive quantitative research. The method used to collect data was done through distributing questionnaires to class XII students and interviewing two teachers, namely a biology teacher and an Islamic religious education teacher. In addition, the daily test results obtained by students were also used as data in this study. The results of the study revealed that students had difficulty in understanding biology learning about bacterial material due to the difficulty of observing micro learning objects plus less supportive learning media where the images on the learning media were of very low quality and colorless. This results in a lack of student motivation in participating in the learning process. For that we need a learning media that is easy to understand and has high quality images so that it can be studied independently by students.

Abstract

Cases of bacterial resistance to antibiotics are discussed with serious problems in the world of health. New antimicrobial compounds are needed which are more effective in treating infectious diseases. Isolate B.J.T.A-6 is an endophytic bacteria from Andalas plants (Morus macroura Miq.) which is known to be able to produce antimicrobial active compounds. Antimicrobial compounds can be produced by growing them on fermented media. The purpose of this study was to optimize the medium fermentation of Andalas endophytic bacteria of B.J.T.A-6  isolates in producing antimicrobial compounds. While medium fermentation is Nutrient Broth (NB), Muller Hinton (MH), and Luria Bertani Broth (LB). Antimicrobial activity tests were carried out by means of diffusion. The parameters used were the diameter of the inhibition zone against Staphylococcus aureus around the disc paper. The profile of medium fermentation optimization was analyzed statistically. The results showed that B.J.T.A-6  isolates could inhibit the highest growth of S. aureus by using LB fermentation medium.

Abstract

The number of death couse of infection continues to increase. Treatment of infections is increasingly difficult due to increased resistance of germs to existing antimicrobial compounds. A new source of antimicrobial substances is needed. One source of antimicrobial substances is endophytic bacteria that live in the stems of Andalas plants. The aim of this study was to compare the diversity of endophytic bacteria present in old and young Andalas plant stems. The method used to isolate endophytic bacteria from the stem of the Andalas plant is the streak plate method. Bacterial identification is done macroscopically and microscopically. The results of macroscopic observation showed that there are differences in the number of isolates found in old and young Andalas plant stems. 11 isolates of endophytic bacteria from Andalas plant stems, each of which consists of 9 endophytic bacteria from old Andalas stems and two isolates from young Andalas stems. A microscopic identification was found that of the 11 isolates isolated, eight isolates were gram-positive and three isolates were gram-negative bacteria

Abstract

 Tea can be used as a fermented drink called kombucha tea. Based on the oxidation process tea is classified into white tea, green tea, oolong tea and black tea. The compounds produced from the fermentation of kombucha tea have a fairly good antibacterial effect. This study aims to determine the antibacterial activity of kombucha tea fermentation from several variations of tea types against Escherichia coli and Staphylococcus aureus. This research is an experimental study with a completely randomized design (CRD) with 5 treatments and 3 replications conducted in May-June 2019 at the Microbiology Laboratory and the Integrated Research Laboratory of the Department of Biology FMIPA UNP. Kombucha tea used in this study was white tea, green tea, oolong tea and black tea. Antibacterial activity test was carried out by paper disc diffusion method against microbial test of S. aureus and E. coli bacteria. Research results showed that the highest antibacterial activity of kombucha tea fermentation in E.coli was found in black tea with a value of 1.64 cm and the lowest in tea white with a value of 1.04 cm. In S. aureus the highest value was obtained in black tea with a value of 2.48 cm and the lowest in white tea with a value of 1.39 cm.

Abstract

Halophilic bacteria is microorganism that live in high salinity environments up to 30%. The bacteria can be found in foods preserved by salting like salted fish. Salted fish is preserved foods are processed by salting and drying. The bacteria that live and ruin salted fish are halophilic bacteria and heterotoleran bacteria like halobacterium, micrococcus, bacillus, pediococcus, peseudomonas and vibrio. This observation aims to determine type of halophilic bacteria found in talang fish.  This is descriptif observation held january up to february 2015 is microbiology laboratory FMIPA UNP and balai veteriner laboratory Bukittinggi. Research methods is cup statterplot methods. Sample is talang fis from Aia Bangih, west pasaman. Based on observation found isolate halophilic bacteria. Its belong to gram positve like coccus. From both, the isolates not capable produce endosopra. Talang fish from Aia Bangih, west pasaman contain two isolate halophilic bacteria consist of a genus and two species.Keywords : talang salted fish, halphilic bacteria, biochemical test
