Abstract

Barred loach is wild fish in which there is no information related to its enzymatic digestive capacity yet. Therefore research on this subject needs to be conducted.  The spectrophotometry method was used to study protease, lipase, and amylase activities in barred loach. The study used 89 fish with an average weight of 3.61 ± 0.26g, and 1.68 ± 0.21g.  The results showed that pH 7.0 was the highest protease activity. Similarly, lipase activity found  between pH 7.0 and pH 8.1. Still, the amylase activity found to be high at pH 8.1. Protease activity also found to be high in the posterior intestine than in the hepato-pancreas and anterior intestine. Lipase and amylase activities did not found differences between hepato-pancreas, anterior intestine, and posterior intestine. In conclusion, the protease, lipase, and amylase activities of barred loach found along the gastrointestinal tract in both large and small fish, which shows that barred loach can digest protein, fat, and carbohydrates in diets better.

Abstract

Studies on morphology, growth, and reproduction have been carried out on wild Javaen barb, but there was no information on its digestive capacity; therefore, the research was conducted to determine protease and amylase activities in the digestive tract. This study used a total of 50 barbs with body weights between 13.56 -128.93g / fish. The measurement of enzyme activity was carried out using the spectrophotometer method. The results showed that differences in fish size resulted in differences in protease activity, but not for amylase. Fish with a small size have a higher protease activity than fish with a larger size. The protease activity did not differ between pH 6.9 to 10.0 but was higher than pH 12.5. Protease activity also did not vary between the anterior and posterior intestine and between 30-50°C. Amylase activity also found no difference between the anterior and posterior intestine, but there was a difference in activity between temperatures of 30-50°C. In conclusion, protease activity occurs in a neutral to alkaline environment, and there were differences in protease activity between different body sizes but not between intestinal segments. Amylase activity occurs throughout the intestine and decreases at temperatures of 50°C.

Abstract

Suatu penelitian untuk mengetahui karakteristik enzim digesti, protease dan amilase pada ikan gurami, Osphronemus gouramy Lac., telah dilakukan dengan metode survey. Jumlah ikan yang digunakan untuk penelitian sebanyak 25 ekor yang dikelompokan menjadi tiga kelompok ukuran13,29, 35,86 dan 91,86 g/ekor. Hasil penelitian menunjukkan bahwa aktivitas protease digesti ikan gurami berbeda secara signifikan pada segmen usus dan pH buffer yang berbeda (P<.05). Aktivitas amilase digesti tidak berbeda secara signifikan diantara segmen usus yang diuji (P>.05), namun berbeda secara signifikan diantara ukuran ikan yang berbeda (P<.05), dan aktivitas amilase tertinggi dijumpai pada ikan dengan ukuran terkecil. Kesimpulan yang dapat diambil adalah bahwa aktivitas protease dan amilase dijumpai sepanjang saluran digestinya baik pada ikan yang berukuran kecil maupun besar. Aktivitas protease umumnya tinggi pada suasana asam dan netral pada usus depan dan tengah. Aktivitas protease ikan yang berukuran besar lebih rendah dari pada ikan yang berukuran lebih kecil. Aktivitas amilase tidak terdapat perbedaan diantara segmen usus yang diuji, namun aktivitas amilase tertinggi dijumpai pada ikan dengan berat rata-rata 13,29 g/ekor
