Abstract

Excessive use of organophosphate pesticides is becoming more common these days. This can affect the level of pesticide residues in farmers' crops. Excessive spraying of pesticides can result in environmental pollution both small and large scale. If excessive exposure to these pesticides can cause damage to the ecosystem. The purpose of this study was to determine the level of organophosphate pesticide residues found in horticulture plants. This research method is a systematic review, by analyzing various studies regarding the contamination of organophosphate pesticide residues in horticultural crops. There are 20 studies from within the country. The results obtained in the form of levels of pesticide residues that are still below the threshold of the BMR (Maximum Residue Limit) of pesticides determined by SNI. Two other studies show that no pesticide residue content was detected. Conclusion: This shows that the sample tested is still safe for consumption. Socialization and education about the doses of pesticide use and how to reduce levels of pesticides in horticultural crops are needed to reduce the risk of exposure to pesticides

Abstract

The menstrual cycle is the distance between the first day of menstruation and the first period of the next menstruation. One of the faktors that influence the menstrual cycle is stres. Stres can stimulate the release of the hormone cortisol, which is a product of glucocorticoid adrenal cortex. This hormone affects the amount of progesterone in the body. This hormonal imbalance will cause changes in the menstrual cycle. The purpose of this study was to analyze the relationship between stres levels and the menstrual cycle of UIN Sunan Ampel Surabaya students. This research was an observational analytic study with a sampling design using simple random sampling with a total of 30 respondents. Data collection techniques use the DASS 42 questionnaire to measure stres levels and the menstrual cycle pattern questionnaire to measure the menstrual cycle. Data analysis using Fisher's exacs test and the results obtained p = 0.031, so it means that there is a relationship between the level of stres with the menstrual cycl.ABSTRAKSiklus menstruasi adalah jarak antara hari pertama menstruasi dengan hari pertama pada menstruasi berikutnya.

Abstract

Yeast is a microorganism that can be found in honeycomb. Yeast identification is a process to find and identify new species. One of which is molecular identification of yeast with rDNA sequences in the ITS region. Before carrying out molecular identification, it is necessary to optimize yeast DNA amplification methods to obtain good DNA sequences that ease the yeast identification process. The purpose of this study was to discover the optimum PCR (Polymerase Chain Reaction)protocols for the identification of yeasts isolated from Apis mellifera honeycomb based on the ITS rDNA. This study used 3 PCR (Polymerase Chain Reaction) protocols, i.e., from Kanti et al. (2018), Ediningsari (2008), and Maulana (2011). This results study shows that the optimum PCR protocol was from Maulana (2011), which produced clear and whole DNA fragment luminescences.

Abstract

Hightlight:Clinical conditions, CD4+ cell counts, and the viral copies number in the blood for AIDS/HIV were given antiretroviral therapy. The profile of CD4+ levels and plasma viral load in HIV patients receiving antiretroviral therapyThe lower CD4+ cell counts and higher viral loads happen in HIV-infected’s men. Abstract:Highly active antiretroviral therapy (HAART) is expected to reduce human immunodeficiency virus (HIV) morbidity and mortality. Antiretroviral therapy in HIV patients is given based on clinical conditions, CD4+ cell counts, and the number of viral copies in the blood. This study aimed to determine the profile of CD4+ levels and plasma viral load in HIV patients receiving antiretroviral therapy. This was a cross-sectional study conducted within six months at Voluntary Counseling and Testing (VCT) in Jayawijaya Hospital, Papua, Indonesia. The CD4+ levels were measured using CD4+ counter and viral plasma was checked using Polymerase Chain Reaction (PCR) for 90 patients. The results showed more female patients had a CD4+ level <200 cells/mm3, a higher number of copies of the virus in the blood plasma, and stages of disease 3 and 4. Statistically, there was a significant relationship between CD4+ levels and gender with a p-value = 0.00. HIV-infected males were more likely to have lower CD4+ cell counts and higher viral loads than females.

Abstract

Indonesia is a country with high variability of microorganisms, including bacteria, yeast, and fungi. Yeast isolates could be isolated from the honeycomb of Apis dorsata. Molecular approaches were used to identify yeast using ribosomal DNA gene sequences, called the ITS gene. The optimum condition for DNA extractions and amplifications are needed for the successfully of molecular identification. Therefore, it is necessary to optimize the DNA extraction and amplification of several protocols to obtain good identification results. This study aimed to compare the effects of DNA extraction with various temperatures and different amplification protocols. LIPI reference DNA extraction protocol with the boiling method and variations in incubation time of 10, 15, and 20 minutes at a temperature of 98° C. Meanwhile, for the amplification of yeast DNA using a variety of different amplification protocols. The results showed the optimal time of incubation was 10 minutes in K1 isolates with DNA purity of 1.896. meanwhile, for isolates K2, K3, and K4 each with a purity of 2.246, 2.335, and 1.748. optimal DNA amplification results were indicated by the presense of DNA bands for each sample K1, K2, K3, and K4, namely 503, 542, 492, and 526 bp. In this study, it can be concluded that the optimal incubation time for the extraction process is 10 minutes. In addition, the optimal amplification protocol was shown in the DNA bands in all sample.
