Abstract

Mangosteen (Garcinia mangostana Linn) is one of the most well-known plants in Indonesia. Mangosteen contains many derivatives of oxygenated and prenylated xanthones from phenolics which exhibit diverse biological activities such as antioxidants, antimalarials, anti-allergies, anti-tumors, antiviral, antibacterial, anti-inflammatory, anti-fungal, anticancer, and antidiabetic. Modification of xanthone compounds is known to increase antidiabetic activity and it is known that α- and β-mangostin acetylated can be produced from the modification of α- and β-mangostin using acetic anhydride. In this study, as many as 1.43 grams (1.59%) of the γ-mangostin compound were successfully isolated from the ethyl acetate extract of mangosteen pericarp. Modification of γ-mangostin through the acetylation reaction produces acetylated γ-mangostin in the form of 3,6,7-tri-methylester-γ-mangostin as much as 32.9 mg (63%). Antidiabetic test results showed γ-mangostin had an IC50 value of 8.55 μM, while the IC50 value of 3.6.7-tri-methylester-γ-mangostin was 1.82 μM. Acarbose as a positive control has an IC50 value of 4.48 μM. This shows that modification can increase antidiabetic activity.

Abstract

The material used in this research was dry powder of pericarp mangosteen (Garcinia mangostana Linn.). which is a family plant of Clusiaceae. This taxa is known as the main source of prenylated xanthones derived from phenolic compounds. The compounds separation process was carried out by maceration method, 8 kg of sample macerated using n-hexane solvent, 90 g of n-hexane extract were produced. The n-hexane extract was fractionated by vacuum liquid chromatography (VLC) using silica gel, eluted by increasing the polarity of the solvent, which is a mixture of n-hexane solvent: dichloromethane (10%, 20%, 30% and 50%), resulting in three combined fractions, namely the first fraction 13,92 g, the second fraction 18,27 g, and the third fraction 23,24 g. The refraction process was then carried out in the third fraction, producing two yellow crystals with melting points respectively, 155-156 °C and 172-173 °C. The structural elucidation method was carried out using UV-Vis, IR, HR-ESI-MS, and NMR spectroscopy techniques. Based on the analysis of spectrum data from two known compounds, 8-deoxygartanin (1) and β-mangostin (2) antidiabetic bioactivity test was carried out by the method of inhibiting the enzyme α-glucosidase in vitro. The test results obtained by compound (1) with an IC50 value of 38,5 μM and compound (2) with an IC50 value of 157,9 μM indicate that the two compounds are included in the inactive category. This is indicated by the IC50 value which is much higher when compared to the acarbose as a positive control with an IC50 value of 4.5 μM.

Abstract

Garcinia husor is one of the folk medicines in Maluku-Indonesia. This species has been used for the treatmet of Malaria disease. The phytochemical contents and antiplasmodial activity not reported yet. In this study we evaluated the quantitative phytochemicals, in vitro and in vivo antiplasmodial activity of stem bark ethyl acetate extract. In vitro assay was done using P. falciparum 3D7 strain sensitive of chloroquine. For in vivo analysis, four groups of M. musculus were infected by P. berghei and their parasitemia levels were for 7 days of treatment with ethyl acetate extract; hematological and biochemical parameter were analyzed at the end of experiment. The result showed ethyl acetate extract with the TPC (169.47 mg GAE/100 g ±0.61) and TPC (167.37 mg QE/100 g ±1.05) was active against P. falciparum 3D7 strain (IC50 value of 0.31±0.43 μg/ml). The animal treated with extract showed suppression of parasitemia to 87.57±1.41% compared with the P. berghei infected-mice (negative control), ED50 value of 22.30 mg/kg BW. The dose of extract in 200 mg/kg BW was reduce parasitemia of infected mice with P. berghei more potential. The ethyl acetate of the stem bark G. husor with has antiplasmodial properties and future investigation are necessary to elucidate its mechanism of action.

Abstract

Postpartum treatments have been used by local women in Indonesia for some time now. One commonly used postpartum treatment is the consumption of a traditional herbal medicine called PHM-1 during the first 40 days after childbirth. In addition, a second medicine known as PHM-2 is taken for days 41-80. However, sufficient scientific evidence about the benefits of this postpartum herbal medicine does not exist. In this study, the antibacterial abilities of PHM-1 and PHM-2 therefore was evaluated and showed positive potential for both. Low IC50 values were obtained against pathogenic bacteria such as Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Rhodopseudomonas palustris, Ralstonia pickettii, and Staphylococcus epidermidis and compared with chloramphenicol as a positive control. The antibacterial activity of both PHM-1 and PHM-2 against R. pickettii had the highest inhibitory activity as evidenced by the lowest IC50 values of 11.16 and 6.26 μg/ml, respectively. Furthermore, an in vivo acute toxicity test showed that PHM-1 produced a low LD50 value of 3.28 g/kg BW for both of male and female mice. These results support the use of both PHM-1 and PHM-2 as antibacterial treatments for postpartum women.

Abstract

Dua senyawa murni berhasil diisolasi dari ekstrak metanol kayu batang tumbuhan Garcinia cylindrocarpa menggunakan metode kromatografi cair vakum, kromatografi kolom grafitasi dan kromatografi lapis tipis prparatif serta pemurnian menggunakan metode rekristalisasi. Karakterisasi senyawa menggunakan spektroskopi UV dan IR.

Abstract

Garcinia tetranda merupakan salah satu spesies dari famili Clusiaceae yang mengandung senyawa metabolit sekunder. Senyawa golongan santon telah berhasil diisolasi dari fraksi S ekstrak etil asetat kayu akar Garcinia tetranda Pierre. Senyawa golongan santon ini adalah α-mangostin 1 yang berupa padatan kuning dengan titik leleh 173-1740C. Metode isolasi yang digunakan yaitu Kromatografi Cair Vakum (KCV) menggunakan silika gel 60 GF254 dan dielusi dengan peningkatan kepolaran pelarut. Penentuan struktur senyawa dilakukan menggunakan analisis UV, IR, 1H dan 13C NMR, DEPT 135, korelasi HSQC dan HMBC serta dibandingkan dengan literatur.

Abstract

Penelitian ini bertujuan untuk mengisolasi senyawa flavonoid dari kulit akar Artocarpus elasticus yang berasal dari Alor, Nusa Tenggara Timur (NTT). Proses ekstraksi dilakukan dengan menggunakan pelarut metanol dan dilanjutkan dengan metode fraksinasi menggunakan Kromatografi Cair Vakum (KCV) dan Kromatografi Kolom Gravitasi (KKG). Pemurnian dilakukan dengan metode rekristalisasi. Senyawa yang berhasil diisolasi berupa serbuk berwarna kuning dengan titik leleh 205-206 °C. Penentuan struktur senyawa dilakukan menggunakan analisis UV, IR, 1H dan 13C NMR dan diketahui bahwa senyawa hasil isolasi merupakan artonin E (1) yang telah dilaporkan sebelumnya.

Abstract

Artocarpus kaya akan senyawa fenolat salah satunya adalah flavonoid. Senyawa ini memiliki sifat bioaktivitas dan telah lama digunakan sebagai sumber pengobatan tradisional di Indonesia. Penelitian ini bertujuan untuk mengisolasi senyawa flavonoid dari kulit akar Artocarpus elasticus yang berasal dari Pulau Alor, Nusa Tenggara Timur (NTT). Proses ekstraksi dilakukan dengan metode maserasi menggunakan pelarut metilen klorida dan dilanjutkan dengan metode fraksinasi menggunakan Kromatografi Cair Vakum (KCV). Pemurnian dilakukan dengan metode rekristalisasi. Penentuan struktur senyawa dilakukan menggunakan analisis UV, IR, 1H dan 13C NMR. Senyawa yang berhasil diisolasi yaitu artobiloksanton (1) berupa padatan coklat muda dengan titik leleh 211-212 °C yang merupakan senyawa yang telah dilaporkan pada penelitian sebelumnya.

Abstract

Senyawa sikloartobiloksanton (1) telah berhasil diisolasi dari kulit akar Artocarpus elasticus yang berasal dari Pulau Flores NTT. Senyawa berupa padatan kuning dengan titik leleh 205-207˚C. Penentuan struktur senyawa dilakukan menggunakan analisis UV, IR, 1H dan 13C-NMR, dan perbandingan senyawa. Senyawa tersebut merupakan senyawa santon turunan flavonoid yang pernah dilaporkan sebelumnya.
