Abstract

The purpose of this research is to study the pathogenicity of Nomuraea rileyi (Farl.) Sams. to eggs and larvae of Spodoptera exigua Hübner (Lepidoptera: Noctuidae).The fungi were isolated from insect cadaver of S. exigua. Egg clusters and several instar larvae with different conidial concentrations (106 – 109 conidia/ml) were used in the experiment. The results of experiment showed that there was no effect of all concentrations of fungi on egg mortality. Mortality of S. exigua larvae was dependent on the instar and conidial concentration. Generally with increasing conidial concentrations, the mortality were also increasing.

Abstract

The objectives of the research were to study the genetic variability of various Beauveria bassiana (Bals.) Vuill. isolates andvirulence to Crocidolomia pavonana. Thirteen isolates of B. bassiana were isolated from Nilaparvata lugens (Stål),Leptocorisa oratorius (F.), Thrips sp., Hypothenemus hampei (Ferr.), Crocidolomia pavonana (F.), Spodoptera litura (F.),Chrysodeixis chalcites (Esp.), and Riptortus linearis (L.) pests from different geographical locations in Indonesia. Fourteendifferent random primers were used to amplify DNA. Clustering of isolates were obtained using Unweight Pair GroupMethod Avarage (UPGMA). The results showed that only OPA08 primer generated more polymorphic DNA fragments.Result of clustering analysis showed that B. bassiana isolates forms two groups having genetic similarity equal to 55%.Clustering of isolates does not correlate to host type nor geographic origin. Mortality of second instar C. pavonana larvaewas dependent on the fungal isolates. At a concentration of 108 conidia/ml, isolate Bb-La2 had the highest virulence whichcaused 65% mortality of 2nd instar larvae. Isolates of B. bassiana is virulent against C. pavonana larvae present in the samegenotype group.

Abstract

Metarhizium spp. is one of the entomopathogenic fungus that can be used to control Spodoptera litura. The purpose of this research was to study the pathogenicity of Metarhizium spp. to Spodoptera litura eggs. The isolates were collected from rhizosphere of different crops i.e., cabbage, onion, leek and chili. The results showed that there was effect of all isolates on egg mortality. Mortality of S. litura eggs depend on the fungal isolates, ranged between 19.79%-75.70%. First instar larvae was also died 3 days after eclosion. The maximum mortality of first instar larvae was 58.65%. At a concentration of 108 conidia/ml, isolate Mt-kb had the highest virulence which caused higher mortality of eggs and first instar larvae.

Abstract

Virulence of various Metarhizium spp. (Deuteromycotina: Hyphomycetes) isolates on cocoa pod borrer pest, Conopomorpha cramerella Snell. (Lepidoptera: Gracillariidae). In Indonesia, Conopomorpha cramerella Snell. (Lepidoptera : Gracillariidae) is a serious pest of cocoa. Entomopathogens such as Metarhizium spp. (Deuteromycotina: Hyphomycetes) can be used to control this pest as an alternative to chemical insecticides. The purpose of this research was to study the virulence of isolates of Metarhizium spp. to pupae of C. cramerella. Nine isolates of Metarhizium spp each with 107 conidia/ml rate were used in the experiment. Pupal mortality was recorded daily. The results showed that isolate MetApCi from chili rhizosphere had the highest virulence causing 96.67% mortality of C. cramerella pupae and with an LT50 of 3.04 days.

Abstract

Crocidolomia pavonana merupakan salah satu hama penting yang membatasi produksi tanaman kubis. Alternatif pengendalian dengan penggunaan mikrorganisme yang berpotensi menekan perkembangan hama ini diantaranya dengan memanfaatkan rizobakteri. Tujuan penelitian adalah untuk mendapatkan isolat rizobakteri yang virulen terhadap larva C. pavonana. Penelitian dilaksanakan di Laboratorium Pengendalian Hayati, Fakultas Pertanian, Universitas Andalas, Padang menggunakan Rancangan Acak Lengkap (RAL) dengan 10 perlakuan dan 5 ulangan. Perlakuan terdiri dari beberapa isolat rizobakteri diantaranya 3 isolat Bacillus thuringiensis (KJKB2, KJKB3 dan BAKB), Bacillus subtilis (KJTSB), 3 isolat Serratia marcescens (AR1, AR2 dan RK10), Stenotrophomonas maltophilia (LMTSA) serta kontrol negatif (Aquadest steril) dan kontrol positif (insektisida Sipermetrin). Uji Virulensi dilakukan dengan metode perendaman pakan dalam suspensi yang mengandung rizobakteri dengan kerapatan populasi 10⁸ sel/ml. Variabel yang diamati adalah mortalitas larva, persentase pembentukan pupa dan imago serta preferensi oviposisi. Data dianalisis dengan sidik ragam dan dilanjutkan dengan uji lanjut BNT pada taraf 5%. Hasil penelitian menunjukkan bahwa semua isolat rizobakteri dapat mematikan larva C. pavonana dan mampu menghambat perkembangan biologi serangga tersebut. Bacillus thuringiensis KJKB3 menunjukkan hasil yang lebih baik dengan nilai mortalitas paling tinggi (64%) dan nilai LT₅₀ yang paling pendek (6.31 hari). Semua perlakuan tidak berpengaruh terhadap preferensi oviposisi, namun perlakuan isolat S. marcescens mampu menurunkan jumlah telur yang diletakkan pada tanaman perlakuan.

Abstract

The lady beetle, Menochilus sexmaculatus is one of the biological control agents that can be used to control aphids. Presently, there is a lack of information about the effectiveness of this beetle to control aphids. The objective of this research was to study the functional response of M. sexmaculatus on Aphis gossypii at different ages of plants. All preys were exposed to one M. sexmaculatus adult for an hour at five different densities of aphids (10, 20, 30, 40, and 50 individuals) is four different ages of chilli plants (2,4,6, and 8 weeks). Data were analyzed by ANOVA and logistic regression to determine the type of functional response. The results showed that the ability of M. sexmaculatus to prey was not significant at 2 to 4 and 6 to 8 weeks old chilli plants. At 2 week old chilli plants M. sexmaculatus showed type I of functional response, while at 4, 6 and 8 weeks the functional response were type III.

Abstract

Penelitian ini bertujuan untuk mempelajari karakter fisiologi isolat B. bassiana yang berasal dari berbagai inang dan virulensinya terhadap Spodoptera litura. Lima isolat B. bassiana yang diuji diisolasi dari hama Leptocorisa oratorius (F.) dan Hypothenemus hampei (Ferr.), rizosfer tanaman bawang daun buah kakao dan batang gandum. Karakter fisiologi yang diamati adalah daya kecambah konidia, pertumbuhan koloni, dan sporulasi, Cendawan diperbanyak pada media SDAY dan uji virulensi dilakukan terhadap larva instar II S. litura. Hasil penelitian menunjukkan bahwa daya kecambah konidia, pertumbuhan koloni dan sporulasi bervariasi antar isolat. Isolat B. bassiana yang diisolasi dari L. oratorius dan buah kakao memiliki karakter fisiologi yang lebih baik dibandingkan dengan isolat lain. Kedua isolat ini juga lebih virulen terhadap larva instar II S. litura dengan mortalitas sebesar 80 – 81,67%.

Asian J Agric 1: 1-5. Metarhizium spp is an entomopathogenic fungus which has wide host range. Metarhizium spp can not only infect insect pests, but can also infect beneficial insects such as predators. The pathogenicity of four isolates of Metarhizium spp. was investigated against Menochilus sexmaculatus predator. Isolates of Metarhizium spp were isolated from rhizosphere of cacao, cabbage, chili, and rubber crops. The experiment was conducted by treating the fourth instar larvae with an appropriate conidial suspension of 108 conidia mL-1. The results showed that all isolates of Metarhizium spp were pathogenic toward beetle predator M. sexmaculatus. Mortality of larvae within seven days after application of conidial suspension varied between 27.50 to 67.50% and there were statistically significant differences among the tested isolates. Metarhizium spp. had also a significant affect in reducing pupation and adult emergence of M. sexmaculatus to below 30% and 3%, respectively. These studies indicate that entomopathogenic fungus Metarhizium spp was pathogenic to beetle predators, M. sexmaculatus.

Abstract

Beauveria bassiana (Balsamo) Vuill. is an entomopathogenic fungi can live endophytic on different kinds of plants. B. bassiana also play a role as growth promoters that improves seed germination and growth of plant. The effect of increased duration of seed treatment with Beauveria bassiana on seed germination and seedling parameters in chili were assessed in laboratory and field conditions. Chili seeds were soaked in the conidial suspension of B. bassiana in a concentration of 1 x 108 conidial / ml or in sterile distilled water for 3, 6, 9, and 12 h. Seed germination percentages and the vigor index were significantly affected by the application of B. bassiana. Overall, seed germination, the vigor index and plant growth of chili were significantly enhanced as seed treatment duration increased. B. bassiana can also accelerate seed germination of chili and increase of seedling growth. Soaking the seeds with a B. bassiana suspension for 9 hours is the best time to increase of seed germination and seedling growth of chili

Abstract

The aim of this study was to identify the chemical compound methanol extract of entomopathogenic fungus Beauveria bassiana from insect walang sangit and the chemical compound roots of red chili plants that were able to stimulate the growth of chilli plants after being applied with entomopathogenic fungus   B. bassiana through seed immersion inoculation using GCMS method. The chemical compound identified as a growth booster in   B. bassiana fungus isolates from the insect walang sangit is an Acetic acid Ethanoic acid Ethylic acid Glacial acetic acid CH3COOH, Hexadecanoic acid, methyl ester (CAS) Methylpalmitate Uniphat A60, n-Hexadecanoic acid Hexadecanoic acid n-Hexadecoic acid Palmitic acid, Dianhydromannitol and Ergosta-5,7,22-trien-3-ol, (3.beta., 22E) - (CAS) Ergosterol (CAS), while the chili root contains Acetic acid Ethanoic acid Ethylic acid Glacials acetic acid CH3COOH, Hexadecanoic acid, methyl ester (CAS) Methyl palmitate Uniphat A60, n -Hexadecanoic acid Hexadecanoic acid Palmitic acid, 8, 11- octadecadienoic acid, methyl ester (CAS) METHYL 8, 11-OCTADECADIENOATE, (23S)-ethylcholest-5-en-3.beta.-ol.s It can be concluded that some of the chemical compounds in methanol extracts identified in chili roots have similarities with chemical compounds that exist in   B. bassiana entomopathogenic fungus which are potential as stimulers of chili plant growth
