Abstract

Sucrose transporter (SUT) is a protein that functions to translocate sucrose from source to sink. Protein activity is determined by the accumulation of sucrose in sugarcane stem. The level of sucrose accumulation is also influenced by the activity of  sucrose phosphat synthase (SPS) and neutral invertase (NI) in conjunction with the synthesis and hydrolysis of sucrose. This aim of this study is to determine the sucrose transporter protein expression in Genetically Modified Product (GMP) of sugarcane.Genetically Modified Product GMP sugarcane events 1, 2, 3, 4, 5, 18, 20 and non GMP were used as materials. The analysis was performed by Western Blot method to observe the presence of SUT protein expression, stem sucrose content by resorcinol method to observe the translocation of sucrose, the SPS enzyme activity and NI that play a role in determining the accumulation of sucrose.The results showed that (1) an increase in SUT protein content in GMP plants events 2 and 18 with a protein band that is thicker than the non GMP plants, (2) SPS and invertase activity at all events GMP crop plants to increase compared to non GMP. Sucrose content of the stem at all events GMP plant to increase with age segment. The older age segments, the higher the sucrose  content. Abstract

Encapsulation is a method for embryogenic tissue coating system by Na-alginate which has synthetic seed preservation purpose. To produce sugarcane synthetic seed need Coleoptillar embryogenic callus and apical bud that it is encapsulated by encapsulation media (MS0 + 4% Na-alginat+ 100 mM CaCl2). In this study, PEG (0%, 1%, 2%, 3%) and PBZ (0 ppm, 1 ppm, 2 ppm, 3 ppm) add to media encapsulation which are give desiccation effect (PEG) and Giberellin synthesis blockade system. Both of PEG or PBZ is used to sugarcane synthetic seed growth retardation that it is implied as storage period. The result are show that PEG 2% is the most effective concentration which is added in encapsulation media. Dessiccation effect of PEG 2% can make synthetic seed storage period until 3 weeks, synthetic seed regeneration produce 89,32 planlets and planlets clorophill concentration is 26,03 ????g/mL. PBZ 1 ppm is the most effective concentration to make storage period until 5 weeks, synthetic seed regeneration produce 39 planlets and planlets clorophill concentration is 35,81 ????g/mL.

Abstract

Aquaponic is considered one of alternative culture system to improve production and quality of leafy vegetables by combining soil less culture and fish farming. Utilization of fish manure contained in waste water provides nutrient sources for growing plants. Three kinds of leafy vegetables including water spinach (Ipomoea aquatica F.), green coral lettuce (Lactuca sativa L.), and bok choi (Brassica rapa L.) were grown using both aquaponic and hydroponic system at the Green House in Serut village, Panti District, Jember Regency, from April to June 2019. Productions of each leafy vegetable were evaluated on both culture systems. Water spinach represented 29.12% higher production in aquaponic than hydroponic system, shown by significant value in plant height, number of leaves, root volume, and leaf fresh weight. Otherwise, bok choi production displayed 17.03% increased in hydroponic than aquaponic system. However, leaf production of lettuce was not significant between hydroponic and aquaponic system. Lettuce production in aquaponic only 1.90% lowest than hydroponic system indicated that lettuce was appropriate to cultivate in both farming systems. Abstract

Induction of in vitro sugarcane through somatic embryogenesis technique influenced by addition of plant growth regulator. The objective of this research was to determine appropriate formulation medium for indirect somatic embryogenesis induction on two potential sugarcane SUT Event 02 and PS 881. This research carried out in three steps, callus induction, callus proliferation, and shoot regeneration. Explants taken from basal of in vitro plantlet one month SUT Event 02 and PS 881 resulted from shoot regeneration previously. Five different medium formulas applied for callus induction and one formula for proliferation and shoot regeneration. Research using completely randomized design (CRD) factorial with five different formulation induction mediums. The result showed that the best respond of indirect somatic embryogenesis on SUT Event 02 and PS 881 was medium containing  3 mgL-1of  2,4-D.

Abstract

Characterization of native orchid (species) in Indonesia is important for preservation of germplasm and selection process to obtain dominant character of orchids as potential parental. Morphological characterization is a method to obtain scientific information in phenotypic manner. Characterization of plants was carried out on 10 native orchids of Dendrobium genus to obtain specific character. Characterization process based on UPOV guidebook and morphological observation of pollinia was also carried out on 10 native orchids to obtain information about the structure and physiology of pollinia. The results of morphological characterization on 10 native orchid showed several qualitative characters such as flowering position, cross section of leaf, pseudobulb firmness, and leaf arrangement in D. Canaliculatum was different from 9 other native orchid. Morphological pollen of 10 native orchid shows the diversity structure of pollinia and the physiological conditions of mature pollen which are ready to pollinate are pollinia with a golden yellow colored. Knowledge and information about parental relationships is the main point for developing ornamental plant breeding programs. Selection of native orchids as potential elder is important where selected elders will be used as a source of genetic diversity. The results of this study are expected to provide scientific information and phenotypic selection for the development of hybrid orchids.

Abstract

Mass propagation technology through somatic embryogenesis has become an alternative for producing sugarcane seedlings rapidly.Application of proper plant growth regulator and concentration contribute to support somatic embryogenesis development. This study applied the combination of liquid and solid culture during proliferation stage to promote cell dispersion of embryogenic callus, rapid  embryo somatic production, and improve regeneration potency of somatic embryo. Application of 2,4-D and coconut water during proliferation may expected as proper combination for accelerating somatic embryo development and regeneration.Development of somatic embryogenesis in sugarcane var. Bululawang during proliferation were described in this study. Embryogenic callusfrom induction media were transferred to proliferation media containing MS Basal + vitamin supplemented with sucrose different level of 2,4-D (1 mgl-1, 2 mgl-1, 3 mgl-1, 4 mgl-1 ) and coconut water (0% and 5%).Result showed that low concentration of 2,4-D induced optimum somatic embryogenesis development in proliferation and regeneration. Concentration of single 2,4-D 1 mgl-1 without coconut water  induced rapid development of scutelar and coleoptilarduring proliferation and resulted in better shoot regeneration. In other way, 4 mgl-1 of 2,4-D concentration  affected to inhibit scutelar and coloeptilar formed as the result of failure callus differentiation.

Abstract

Phalaenopsis merupakan anggrek yang mempunyai nilai ekonomis tinggi karena warna yang menarik. Permintaan selalu meningkat sehingga  tehnologi kultur jaringan sangat dibutuhkan dalam perbanyakan anggrek ini. Perbanyakan tanaman anggrek menggunakan sumber eksplan protocorm-like body (PLB) merupakan salah satu cara yang tepat untuk menghasilkan anggrek dalam jumlah banyak. Penelitian ini bertujuan untuk mendapatkan kombinasi NAA dan Kinetin terbaik pada perkembangan PLB di kultur cair serta PLB yang disubkultur di media regenerasi. Bahan tanam menggunakan PLB umur 2 bulan yang ditanam pada media Vacin & Went (VW) cair dengan perlakuan kombinasi NAA dan Kinetin masing-masing pada konsentrasi 2,5 mg/L, 5 mg/L dan 7,5 mg/L. Media regenerasi menggunakan VW padat dengan suplemen 15% air kelapa, 5% pisang, 5% kentang dan 0,2% arang aktif. Percobaan menggunakan Rancangan Acak Lengkap dengan tiga kali ulangan. Hasil penelitian menunjukkan bahwa kombinasi NAA dan kinetin tidak berpengaruh terhadap jumlah dan berat PLB. Sementara itu, warna PLB hampir semua menunjukkan warna yang sama yaitu hijau kekuningan (5GY 7/6) hanya pada kombinasi perlakuan NAA 5 mg/L + Kinetin 7,5 mg/L terjadi klorosis berwarna kuning pucat (5Y8/4).  Perubahan warna planlet terjadi dari hijau kekuningan terang (7.5GY8/8) menjadi hijau kekuningan (7.5GY7/6) dan hijau kekuningan tua (7.5GY6/10) masing-masing pada awal aklimatisasi, serta pada dua dan empat minggu. Bibit anggrek tumbuh dengan baik dan menjadi tanaman dewasa setelah dipindahkan pada media moss hitam.

Abstract

Penggunaan nutrisi organik untuk multiplikasi tanaman anggrek secara in vitro telah banyak digunakan untuk mendorong laju regenerasi yang tinggi pada tanaman anggrek. Setiap spesies anggrek membutuhkan penambahan sumber organik yang berbeda untuk merangsang regenerasi yang lebih baik. Pemilihan jenis sumber nutrisi organik yang sesuai pada media regenerasi anggrek sangat diperlukan untuk menghasilkan planlet anggrek yang berkualitas. Tujuan penelitian ini adalah mengevaluasi nutrisi organik yang tepat untuk regenerasi plantlet Dendrobium lasiantera x D. tiger twist, Phalaenopsis amabilis, dan Oncidium golden shower. Bahan yang digunakan antara lain air kelapa, ekstrak kentang, ekstrak tomat, dan ekstrak pisang. Penelitian dilakukan di Laboratorium Agrotechnopark Universitas Jember menggunakan Rancangan Acak Lengkap (RAL) dengan lima ulangan. Faktor perlakuan adalah kontrol (S1), ekstrak air kelapa (S2), ekstrak kentang (S3), ekstrak tomat (S4), dan ekstrak pisang (S5). Hasil penelitian menunjukkan bahwa pemberian nutrisi organik mempengaruhi ketiga genus anggrek yang digunakan. Penambahan ekstrak tomat 150 g/l menghasilkan pertumbuhan yang lebih baik untuk parameter tinggi tanaman, jumlah tunas, daun, dan akar pada angrek Dendrobium sp. Penambahan ekstrak kentang 150 g/l memberikan respon pertumbuhan tinggi planlet, jumlah tunas, dan jumlah akar yang baik pada anggrek Oncidium sp. serta jumlah daun dan jumlah akar yang baik pada anggrek P. amabilis dibandingkan sumber nutrisi organik lainnya.

Abstract

Sucrose‐phosphate synthase (SPS) is a key enzyme catalyzing the formation of sucrose‐6‐phosphate through the transfer of uridine‐diphosphate glucose (UDP‐G) as a donor to fructose‐6‐phosphate (F6P) as an acceptor. Plant SPS consists of three main domains: N‐terminal, glycosyltransferase, and C‐terminal domains. Among these, the N‐terminal domain is involved in regulating the allosteric activator glucose‐6‐phosphate (G6P). This study was directed toward determining the regulation and characterization of N‐terminal truncated SPS in transgenic tomato. In this study, the N‐terminal truncated mutant of sugarcane SPS (ΔN‐SoSPS1) and full‐length sugarcane SPS (FL‐SoSPS1) were expressed into tomato plants to verify the functional role and importance of the N‐terminal domain in plant SPS. Overexpression of ΔN‐SoSPS1 led to an up to 3‐fold increase in the specific activity of SPS compared to non‐transformant plants (WT), while the specific activity of ΔN‐SoSPS1 was higher than FL‐SoSPS1 in transgenic tomato plants. Unlike WT and FL‐SoSPS1, the ΔN‐SoSPS1 mutant was not allosterically regulated by G6P. These results indicated that deletion of the N‐terminal domain promotes the loss of allosteric activation by G6P and increases binding affinity between enzyme and substrate.

Abstract

Vegetables as a horticultural product is a food source that is always consumed by humans daily, including in urban areas. Assorted Vegetables can be eaten raw or processed and it takes as a decoration on the food. Needs vegetables including lettuce, green pakcoy and other vegetables from day to day continues to increase. Therefore, it needs supply of vegetables from the area around the town of Jember including Serut Village, District Panti Sub-District, Jember District. Serut village has the potential of non-agricultural land could be developed into a producer of vegetables. Empowerment yard area for the cultivation of vegetables will be achieved if supported by an efficient hydroponic cultivation techniques and the empowerment of human resources are adequate. Empowerment of rural communities is expected to increase the quantity and quality of vegetable production of lettuce, cabbage, and other vegetables as well as to support sustainable vegetable production. Through this hydroponic cultivation of fresh vegetables will be produced continuously, scheduled and able to add value to the revenue for the community.
