Abstract

ABSTRAK Latar belakang Perbedaan gambaran morfologi antara sel mesotel reaktif dan adenokarsinoma pada efusi pleura masih merupakan salah satu tantangan dalam diagnostik sitologi. Sejauh ini, telah banyak penanda tumor yang digunakan pada efusi pleura dengan berbagai derajat efektivitas. Penelitian ini bertujuan untuk mengetahui tingkat positifitas carcinoembryonic antigen (CEA) dan calretinin pada adenokarsinoma dan sel mesotel reaktif dari sitologi cairan pleura. Bahan dan cara Tujuh puluh empat sampel arsip slaid dari efusi pleura dengan diagnosa adenokarsinoma atau sel mesotel reaktif diperoleh selama periode waktu 1 Februari 2010-31 Januari 2011. Semua slaid berasal dari spesimen sitospin dengan pulasan Papanicolaou dan Diff Quik kemudian dipulas ulang dengan menggunakan antibodi CEA dan calretinin. Ekspresi kedua penanda tumor ini dievaluasi untuk menilai tingkat positifitas, korelasi antara kedua antibodi dianalisis dengan uji korelasi non parametrik Kendall’s. Hasil Pulasan positif untuk CEA dan calretinin masing-masing sebesar 81,6% (31/38) dan 39,5% (15/38) pada kasus adenokarsinoma Untuk sel mesotel reaktif, positifitas CEA dan calretinin masing-masing sebesar 36,1% (13/36) and 88,9% (32/36). Koekspresi positif kedua penanda tumor dijumpai sebanyak 25% (9/36) pada kasus sel mesotel reaktif. Abstract

Leprosy is a chronic granulomatous infectious disease that attacks the peripheral nerves, skin, and other organs. The cytokine IL-10 can deactivate macrophages, inhibit IL-12 production, which also inhibits IFN-ϒ production. IL-10 directly inhibits CD4+T cells and antigen-presenting cell (APC) function in cells infected with M. leprae. The purpose of this study was to determine the differences in IL-10 levels in leprosy patients and non-patients at dr. Mohammad Hoesin Palembang Hospital. This study used a case-control design for comparing 2 groups. Respondents in this study were 80 people. The case samples were all leprosy patients who were treated at the dr. Mohammad Hoesin Hospital, Palembang for the period of January 16 - February 16, 2019, while the control group was medical personnel and paramedics who worked at the same hospital. The examination was carried out using the ELISA Sandwich method. The results showed that there was a significant difference in IL-10 levels between leprosy patients and non-leprosy leprosy (p=0.000). The average IL-10 level in leprosy patients (13.24 pg/ml) was lower than that in non-patients (40.15 pg/ml). It is concluded that there is a difference in IL-10 levels between leprosy patients and non-leprosy patients in leprosy sufferers are lower than non-leprosy persons.

Abstract

Background Cervical cancer is the third most common type and the fourth most deadly cancerian women which remains a significant health problem worldwide especially in developing coumtry. Ki-67 is a prototype protein in nuclear which influence cell proliferation rate, hence it can be used as one of parameter to evaluate progressivity of tumor cells in carcinoma cases as the cervical squamous cell carcinoma (SCC). This study aims to determine the relationship between clinical staging of cervical SCC and histopatological grading of the cervical SCC with the expression of Ki-67. Methods This was an observational study with a cross sectional design. Sixty cervical squamousa cell carcinoma archive histopathological slide samples (45 biopsy specimens and 15 radical hysterectomy tissues) were collected for this study from the Anatomical Pathology Department of RSUP dr. Mohammad Hoesin Palembang (January 2008 to July 2013). The samples were stained with Ki-67 antibody and analyzed with respect to patient’s clinical stage and histopatological grade. Results The positivity of Ki-67 expressions were more common in the age of ≤ 50 year-old group, in the non-keratinizing type, in the late clinical staging (stage IIIB) and in the moderatelly differentiated histopathological grade. High scores of the Ki-67 expressions (Ki-67>20%) was predominantly found in the late clinical stage and moderatelly differentiated histopathological grade cases. Statistically, there was a significant relationship (p=0.034) with a strong correlation (r=0.714) between the Ki-67 ecpression and the clinical stage of cervical SCC, but not histopathological grade. Conclusion The higher clinical staging of cervical SCC, the higher expression of Ki-67. Key words: cervical squamous cell carcinoma, clinical staging, histophatological grade, Ki-67.

Abstract

BackgroundColorectal carcinoma has become one of primary problem that caused mortality and morbidity around the world. This malignancyhas contributed almost more than 9% of all cancer incidence. Lymphocyte infiltration density (TILs) is one of prognostic factors toevaluate colorectal cancer. Although some studies have found that there still have variations in sassociation lymphocyte density withclinicopathology of colorectal carcinoma which related to patient prognosis. This study was aimed to identify and analyze theassociation of lymphocyte density with clinicopathological colorectal carcinoma patients in Dr. Mohammad Hoesin General HospitalPalembang.MethodsA total of 41 HE stained slides which were diagnosed as colorectal carcinoma were observed under the light microscope.Lymphocytes were counted in five different areas that showed the highest lymphocytes density. The mean values of the lymphocytewere received from those five different areas. Determination of cutoff point was counted from the median of all mean value of thelymphocytes and being classified to become two categories, high-density lymphocytes and low-density lymphocytes.ResultsOut of 41 subjects, most patients have tumor size more than/or equal to 5 cm (63.4%), the tumors are often found in the left-sided ofcolon (39.0%), have moderate differentiated degree (65.9%), are included into T3 in tumor staging (48.8%), and no regional lymphnodes metastasis (39.0%). The most histopathological classification is adenocarcinoma (41.5%). Bivariate analysis showed thatlymphocyte density has correlation with tumor differentiation degree (p=0.021) and histopathological classification (p=0.004).ConclusionLymphocyte infiltration density has correlation with degree differentiation and histopathological classification of colorectal carcinomain Dr. Mohammad. Abstract

Sistem reproduksi wanita merupakan sistem kompleks yang meliputi semua alat reproduksi wanita dan akan berubah sesuai dengan siklus perkembangan. Siklus perkembangan reproduksi wanita berlangsung secara alamiah mulai dari menarche sampai menopause. Sebelum terjadi menopause ada masa transisi yang disebut premenopause. Perubahan organ reproduksi pada masa premenopause ini ditandai dengan terjadinya gangguan pada menstruasi. Perpanjangan dan pengurangan siklus menstruasi dapat dipengaruhi oleh beberapa faktor seperti Anti Mullerian Hormon (AMH) dan Indeks Massa Tubuh (IMT). Tujuan mengetahui hubungan kadar AMH dan IMT terhadap panjang siklus menstruasi premenopause di BPM Ranting Seberang Ulu I.Penelitian ini dilakukan di Laboratorium Biomolekuler Kedokteran Universitas Sriwijaya dengan desain penelitian observasional analitik. Sebanyak 74 orang sample dipilih secara Convinience Sampling yang dibagi menjadi 2 kelompok yaitu kelompok siklus mesntruasi normal dan tidak normal. Kedua kelompok diukur TB dan BB nya untuk mengetahui IMT dan diambil darah vena untuk dialkukan pemeriksaan AMH dengan metode ELISA. Hasil analisis data AMH dan siklus menstruasi dilakukan dengan uji Mann-Whitney. nilai p=0,004, IMT dan siklus menstruasi dilakukan dengan uji T Independen nilai p=0,484, AMH dan IMT dilakukan dengan uji Spearman nilai p=0,789 dengan nilai alpha 0,05 (p<?). Disimpulkan bahwa ada hubungan antara AMH dan siklus mestruasi, tidak ada hubungan antara IMT dan siklus mestruasi dan tidak ada hubungan antara AMH dan IMT.

Abstract

Formalin-fixed paraffin-embedded (FFPE) archival tissue presents a readily available resource in molecular study nowadays. The quality of nucleid acid, especially mitoconhdrial DNA (mtDNA) extracted from FFPE tissue could be affected by the storage time. Thus, this study investigated if the FFPE tissue’s storage time had an effect on the quality of the extracted mtDNA at Department of Pathology RSUP Dr. Mohammad Hoesin Palembang. DNA was extracted from 16 randomly selected archival FFPE tissues in Laboratory of Pathology Anatomy, RSUP dr. Mohammad Hoesin Palembang. The samples were grouped based on their storage time (less than 1 year and 1 to 5 yrs.’ old). The isolated DNA from each group was amplified using PCR with two primer pairs specifically designto amplify mtDNA of 320 bp and 142bp length, respectively. The PCR products were visualized by electrophoresismethod. None of the samples from both groups could be amplified witht the 320bp primers. However, the PCR result of the 149 bp primers showed positive for all of the samples from each study group. The study indicated that the storage time does not affect the quality of mtDNA isolated from the FFPE samples archived in Department of Pathology RSUP Dr. Mohammad Hoesin Palembang. Furthermore, the study showed that the mtDNA extracted from FFPE tissue has been degraded, therefore, the samples are not suitable for genetic studies require a long mtDNA amplicon.

Abstract

ABSTRAK Latar belakang Teknik potong beku maupun sitologi imprint dan scraping sudah digunakan sebagai konsultasi patologi intraoperatif dalam mendiagnosis tumor ovarium. Penelitian mengenai keakuratan diagnosis potong beku intraoperatif pada tumor ovarium pernah dilakukan namun terhadap sitologi imprint dan scraping belum pernah dilakukan di RSUP. Dr. Moh. Hoesin Palembang. Tujuan dari penelitian ini adalah untuk mengetahui akurasi potong beku, sitologi imprint dan scraping dalam mendiagnosis tumor ovarium dibandingkan histopatologi sebagai baku emasnya. Bahan dan cara kerja Penelitian ini merupakan uji diagnostik pada 105 sampel arsip tumor ovarium dari bulan Januari 2009 sampai Desember 2010. Hasil pemeriksaan potong beku, sitologi imprint dan scraping serta histopatologinya dibaca ulang oleh dua orang Spesialis Patologi Anatomik. Data dianalisis untuk menghitung nilai akurasi pemeriksaan potong beku serta sitologi imprint dan scraping. Hasil Didapatkan nilai akurasi potong beku sebesar 99% dan akurasi sitologi imprint dan scraping sebesar 97,14%. Kesimpulan Pemeriksaan potong beku serta sitologi imprint dan scraping merupakan pemeriksaan dengan nilai akurasi yang tinggi. Pemeriksaan sitologi imprint dan scraping dapat menjadi alternatif pemeriksaan intraoperatif untuk rumah sakit yang tidak mempunyai fasilitas potong beku. Kata kunci: Tumor ovarium, potong beku, sitologi imprint dan scraping, akurasi. ABSTRACT Background Frozen section or imprint and scrape cytology technics have been used as an intraoperative consultation measure for ovarian tumor diagnosis. Study about the accuracy of frozen section diagnosis for ovarian tumor had already been done in Dr. mohammad Hoesin General Hospital Palembang but not for imprint and scrape cytology. The aim of this study is to evaluate the accuracy of frozen section, imprint and scrape cytology in ovarian tumor diagnosis compared with histopathology as a gold standard. Methods This study was a diagnostic tes of one hundred and five samples from the archives of ovarian tumors from January 2009 to December 2010. The frozen section, imprint, scrape cytology and histopathological results were evaluated by two pathologists. The data were analized for the determination of the accuracy of frozen section and imprint and scrape cytology. Results The overall accuracy rate for frozen section, imprint and scrape cytology is 99% and 97,14% respectively. Conclusion Frozen section and imprint and scrape cytological examination revealed high accuracy, therefore imprint and scrape cytology are strongly recommended as an alternative measure for intraoperative consultation instead of frozen section mainly for hospitals that do not have facility of frozen section. Abstract

Tuberculosis (TB) is an infectious disease caused by the bacterium Mycobacterium tuberculosis which is still a health problem in the world including in Indonesia. TB infection arises due to interference with the regulation of the body's defense system. Cellular immune system plays a role in fighting TB infection, namely the role of T lymphocytes that differentiate into Th1 cells secrete pro-inflammatory cytokines IFN-? and Th2 cells that secrete IL-10 anti-inflammatory cytokines. The EC610 fusion antigen is specific and has a strong antigenicity to T cell stimulation. The aim of the study was to determine differences in the mean levels of IFN-? and IL-10 after EC610 antigen stimulation in patients with active TB and latent TB. The design of this study was a quasi-in vitro experiment with PBMC culture stimulated by EC610 antigens in the active TB and latent TB groups. The research and examination were carried out at the South Sumatra Province SSR and the Health Research and Development Laboratory Center, Central Jakarta. There were 21 subjects with active TB and 28 subjects with latent TB who met the inclusion criteria. Measurement of IFN-levels? and IL-10 was performed using ELISA-Reader. The results showed the levels of IFN-? and IL-10 after EC610 antigen stimulation were higher in active TB than latent TB. High levels of IFN-? active TB patients showed protective immune responses against M.tb germs while high levels of IL-10 showed their role as anti-inflammatory. There is no significant difference in the average level of IFN-? in patients with active TB and latent TB (p = 0.769) and there was a significant difference in the level of IL-10 levels after EC610 antigen stimulation was higher in active TB than latent TB (p = 0,000).
