Abstract

Antiangiogenesis (inhibition of new blood vessels formation) has become a strategy to inhibit cancer development. The aim of this experiment was to investigate antiangiogenic effect  of  Gynura  procumbens  (Lour)  Merr  focusing  on  the  decreasing  of  the  number  of macrophages  and  heterophils  on  chick  embryo  chorioallantoic  membrane.  Nine-days-aged-eggs  were  divided  into  six  groups  (eight  eggs  each  group).  Group  I  (positive  control)  eggs were  induced  with  bFGF+Tris  HCl.  Group  II  (negative  control)  eggs  were  treated  with DMSO+Tris  HCl.  Group  III  (treatment  I)  eggs  were  induced  with  60  ng  bFGF  and  treated with  ethanolic  extract  of  G.  procumbens  leaves  with  the  dose  of  60  µg.  The  following treatment  groups,  i.e.  group  IV  (treatment  II),  group  V  (treatment  III),  and  group  VI (treatment IV) were treated with increasing dose of extract, starting from 75 µg, 90 µg, and the last was 110 µg. Eggs were incubated until they reach the age of twelve days to observe macrophages, while  to observe heterophils, eggs were incubated until  the  age of seventeen days. Based on haematoxylin-eosin staining, macrophages in the treatment groups were less than the control positive group (bFGF+Tris HCl), but based on giemsa staining, the effect of Gynura procumbens  in decreasing the number of heterophils could not  be observed  because some blood smears. These analysis suggest that  the  ethanolic extract of  Gynura procumbens leaves can perform as antiangiogenic agent decreasing the number of macrophages.Keywords: antiangiogenic, macrophages, heterophils, Gynura procumbens

Abstract

Ethanol extract Kencur (Kaempferia galanga L.) has been proven to have antiinflammatory effect as well as to decrease the side effect of NSAIDs drug use, to develop the formulation of ethanol extract of kencur, then made in the dosage form of topical patch. Topical patch preparations are expected to avoid effects on the gastrointestinal tract, first- pass metabolism, and facilitate the use of the patient thereby increasing patient compliance. In the patch formulation, the most important things to note is the penetration enhancer/ enhancer, and enhancer used in this study is sodium lauryl sulphate. This study aims to determine the effectiveness of sodium lauryl sulphate in the preparation of a topical patch of kencur extract as anti inflammatory on the number of neutrophils in the blood. The research method uses canrrageenan as an inflammation inductor and neutrophil count observation was conducted using blood smears, then was observed on the first, third and seventh. Group of treatment which was given sodium diclofenac gel as positive control, group of treatment which was without enhancer as the 1st group, groups of treatment which were givenenhancer 1%, 3%, and 5% as the 2nd, 3rd and 4th group. The result was analyzed by one way ANOVA, followed by Post Hoc Duncan. The experimental results prove that there was a significant difference between the number of neutrophils and macrophages in the negative control group who were not given enhancer and the treatment group were given enhancer sodium lauryl sulphate values ( p < 0.05 ). As well as the group treated with the addition of enhancer sodium lauryl sulfate showed a greater amount of decline compared to the treatment group that contains only ethanol extract kencur.

Abstract

Abstract : Sea urchin (Echinometra mathaei) contains a compound of Echinochrome A, which is a polyhydroxy naphthoquinone that is known to be potential antioxidant. The antioxidant effect correlated with the antidiabetic activity that associated with oxidative stress conditions. The aim of this study was to determine the effect of 70% Ethanol Extract of sea urchin (Echinometra mathaei) on blood glucose levels in alloxan induced rats. This research was an experimental study using 30 male white rats, divided into 6 groups, i.e the negative control group (normal rat were only given CMC Na 0,5%), Positive control group (hyperglycemic rats were only treated with CMC Na 0,5%), metformin group as a refference standard group (hyperglycemic rats that treated with metformin) and the treatment group (hyperglycemic rats were treated with ethanol extract 70% of Echinometra mathaei dose of 100 mg/kgBW, 200 mg/kgBW and 400 mg/kgBW). Alloxan monohidrat 120 mg/KgBW was used as diabetogen agent. The parameters examined in this study were blood glucose levels of rats after 7 days of treatment. The observational data were analyzed using the One Way ANOVA (P < 0.05) and continued with LSD. The results of this study showed that all doses of ethanol extract significantly reduced blood glucose levels in rats compared to the control group. The most effective dose in reducing blood glucose levels and compared to metformin was 400 mg / kgBW. Key word: alloxan induced, Echinometra mathaei, ethanol 70%, hiperglycemia, marine product.
