Abstract

Plasma concentration of cortisol and hematological profile on broiler chickens can be used as heat stress indicators. This study was conducted to investigate the effect of administration of n-hexane extract of jaloh bark (EHJ) on heat-stressed broiler chicken. Thirty broilers, aged 20 days (strain Cobb), were randomly divided into 5 groups. The first group was external control (tCP) i.e. chickens without heat stress and without EHJ administration. The second group was internal control (CP) representing chickens given heat stress without EHJ administration. The third, fourth, and  fifth groups were chickens given heat stress and EHJ administration at doses 5, 10, and 20 mg/kg BW, respectively symbolized as CP+EHJ5, CP+EHJ10, and CP+EHJ20. The experimental broilers were exposed to heat stress in constant temperature of 33 ± 1oC for 4 hours daily for 5 and 10 days. The EHJ was given 1 hour before temperature in the cage was raised and was given in daily basis.  The level of cortisol in feces were measured from feces collected 1-2 hours before  treatment, 3-4 hours after heat stress exposure, and 2-3 hours after heat stress termination. This research indicated that heat stress at temperature 33 ± 1oC during 2 and 4 hours increased cortisol  excretion in feces (P<0.05) and increased ratio H : L (P<0.10). The EHJ at dose of 10 mg/kg BW relatively gave more protection from heat stress impact on broilers. Key Words: Cortisol, Triiodothyronine, Heat Stress, Salix, Hematology

Abstract

The function of Ito cells is expanding from a fat-storing site to a center of extracellular matrix metabolism and mediator production in the liver. Immunohistochemical reactivities of Ito cells were examined in eight livers of broiler chickens affected with spontaneous cholangiohepatitis and six chicken livers with malformation of extrahepatic biliary tracts. The livers in both groups revealed severe diffuse fibrosis. Ito cells expressing HHF35 muscle actin and desmin actively proliferated in the fibrotic foci of the all livers. The immunoreactivities of Ito cells to antibodies were enhanced compared with those in normal livers. There were no immunohistochemical differences between the Ito cells of two groups. From these findings, it was suggested that Ito cells actively proliferate and show enhanced immunoreactivities in the livers affected with cholangiohepatitis and malformation of extrahepatic biliary tracts.Key words: Cholangiohepatitis, immunohistochemistry, ito cell. broiler chicken

Abstract

The second group was internal control (CP) representing chickens given heat stress without jaloh extract. The third, fourth, and fifth groups consisted of chickens given heat stress and 10 mg/kg BB of hexane (CP+Hek), ethyl acetate (CP+EtOAc), and ethanol (CP+EtOH) fractions, respectively. Heat stress and jaloh extract were given every day. Extract of jaloh were given 1 hour before temperature in the cage was raised. On 10th day of the experiment, 3 chickens were killed and necropted. Liver and kidney tissues from each treatment were analyzed for histological condition. The results indicated that heat stress can induce injury degeneration, necrosis, and infiltration of inflammation cells of liver and kidney. It is concluded that the hexane fraction of jaloh extract could protect the detrimental effects on heat stress. Key Words: Heat Stress, Extract Salix, Liver, Broiler Kidney

Abstract

Neuroblastic tumors, which are uncommon tumors of nerve cell origin, are classified into neuroblastoma, ganglioneuroblastoma, and ganglioneuroma depend-ing on the degree of maturation of neoplastic neuroblasts and development of schwannian stroma [2,4,7].  Ganglioneuroma is a rare neuroectodermal tumor of the peripheral nervous system. Ganglio-neuromas generally are considered to be benign neoplasms composed of both mature ganglion cells and nerve fascicles with axons, Schwann cells, fibroblasts, and other connective tissue elements [1].  The aim of this study is to clarify the pathomor-phological feature of a subcutaneous ganglio-neuroma which occurred in a Sumatran tiger.

Abstract

Antigen-antibody reaction is an important tool for the analysis of localization of target molecules, including antigenic protein within worm tissues. The purpose of the present research was to demonstrate the ability of immunoglobulin yolk (IgY) anti-excretory/secretory recognized the antigen in the tissue of Ascaridia galli by mean of immunohistochemistry method. The excretory/secretory protein was procured from A. galli and concentrated by mean of vivaspin 30,000 MWCO. IgY was produced by egg yolks of immunized chickens with excretory/secretory, and purified using fast protein liquid chromatography (FPLC) method. A. galli adult worms were cut in transversal and longitudinal section of the center and anterior region. Slides were incubated with both primary IgY for overnight at 4 oC and secondary antibody rabbit anti-chicken IgY HRP-conjugate for one hour at room temperature. The slides were stained with 3-amino, 9-ethylcarbazole (AEC) chromogen, counterstained with Lillie Mayer Haematoxylin, and mounted in glyserin aqueous mount.  Antigen-antibody reaction was investigated under a microscope. The result showed that antigen was appeared in the tissues such as cuticle, epicuticle, buccal cavity, and eggs inside the uterine of A. galli. This research concluded that IgY stimulated by the excretory/secretory was able to recognized the antigen scattered in the tissues of A. galli so the IgY could be applied for immunodiagnostic.
