Abstract

Adanya kasus pandemi COVID-19 menyebabkan berkurangnya pendapatan masyarakat Desa Pulao Semambu.

Abstract

Lais (Krypyoterus apogon) is highly economic important catfish in South Sumatera. Investigating fish diversity of catfish is of importance for species conservation. Many species or subspecies extincted due to environment decreasing caused by habitat loss, pollution and over fishing. Cytochrome C Oxidase subunit I (COI) is one of mitochondrial DNA markers used for species barcoding in freshwater, brackishwater and marine fish. This research aim to explore the use of COI gene for species barcoding in investigating the diversity of COI nucleotide, constructing phylogenetic tree of lais, and knowing physical and chemistry characteristic of habitat at Musi and Kelekar River, South Sumatera. The methods used in the research consisted of DNA  extraction, PCR (Polymerase Chain Reaction) amplification and sequencing mtDNA COI gene of Kryptopterus apogon obtained from Musi and Kelekar River (South Sumatera). A 615 bp (sample code CM1) and 693 bp sequences (sample code CK1 and CK2) of parsial coding were sequenced from this fish. Nucleotide BLAST analyzes showed COI gen can distinguish lais at Musi and Kelekar River from others fish at GenBank database with high identity (92.29% and 92.21%) to Phalacronotus bleekeri (Thailand) and Phalacronotus apogon (Australia).Phylogenetic analyzes indicated that obviously, lais was at the same cluster with others genus in Siluridae family, but different cluster from Hemibagrus nemurus (Bagridae), Chanidae and Oreochomis niloticus (Cichlidae). Water quality criteria (water tranparancy, temperature, ammonia, pH and alkalinity) at Musi and Kelekar River were still in tolerance for maintainning survival rate of lais. Further study using more species in Siluridae family and various habitat are needed to investigate the diversity of DNA of Siluridae at South Sumatera water resources. Keywords: Catfish, DNA barcode, COI gene, Musi River, Kelekar River

Abstract

The transportation process can lead to stress which increases mortality in fish. Efforts to reduce mortality in the process of fish transportation can be conducted by minimizing the metabolic activities of fish during transportation. One of the natural substances that is used for fish transportation is tuba roots. The aim of this study is to determine the effect of different dosages of tuba root extract on the transport of catfish seeds with a closed transport system for 12 hours. This research was conducted at the Aquaculture Laboratory, Aquaculture Study Program, Faculty of Agriculture, Sriwijaya University. The research was conducted in April 2019. This study used a completely randomized design (CRD) method with four different dosage treatments (T) and three replications. The dosages of tuba root extract were: 0 ml.L-1 (T0), 0.05 ml.L-1 (T1), 0.10 ml.L-1 (T2), 0.15 ml.L-1 (T3). The parameters observed were blood glucose levels, water quality, survival of catfish after transportation and maintenance for 7 days. The results showed that T1 (0,05 ml.L-1) was gave the highest survival rate of 91.66±2.88%, with the lowest stress level indicated by blood glucose values 89±30.63. Abstract

The demand of live carp is higher and the price can be higher than the price of died carp. One of  ways to transport live fish was wet transportation system. The aim of this research was to know the effect of red guava leaf extract for reducing metabolism of common carp during transportation. This research used completely randomized design (CRD) with defferent concentration with four treatments and three replications i.e. of P. guajava var. pomifera leaves extract 0% (T0), 0.25% (T1), 0.50% (T2), and 0.75% (T3). The observed parameters were survival rate, glucose analysis and water quality (temperature, pH,  Dissolved Oxygen (DO), Total Ammonia Nitrogen (TAN) and carbon dioxide (CO2)). Addition of red fruit guava leave extract into transport media during transportation increased the percentage of survival rate of common carp during transportation. The survival rate of during transportation common carp was highest on T1, with the concentration of red fruit guava leave extract by 0.25%. The value of average in blood glucose levels was found in T2 (0.50%). Addition of P. guajava var. pomifera extract was able to reduce TAN values during transportation and the addition of P. guajava var. pomifera extract did not cause further effects on the survival rate of carp during maintenance post-transportation.
