Abstract

Protoplasts regeneration was studied on 3 cultivars of eggplant (pupel, green, ang white fruits) and the regenerant were tested for their resistance to bacterial wilt. The researh was conducted in different steps, including protoplast isolation, protoplast purification, protoplast culture, cell suspention dilution, shoot regeneration, plantlet aclimatization, and testing for the resistant to bacterial wilt. Observations were made for protoplast number of each cultivar, protoplast cell wal formation, cell division, number of cell colony, microcalli, and shoot, percent of plantlet survival, in acclimatization, and the resistance of regenerants to bacterial wilt. Data was analized by calculating the average, percentage, and standart deviation. Results show that the combination of 0.5% cellulase Onozuka RS, 0.5% macerozyme R-10, 0.05% MES, and 9,1% mannitol in CPW solution was able to isolateds alot of sprotoplast (10 ) in each cultivars. The first culture incubation with out lighty in both (KM8P and VKM) media and reach which 0,2 mg/1 2,4-D, 0,5 mg/1 zeathin, 1 mg/1 IAA, promoted the formation of cell wall and cell division developed to microcalli. Regeneration media of MS, applied with vitamin MW, 0,1 mg/1 NAA, and 2 mg/1 zeathine produced 13 shoots.. From 21 regenerants acclimatized only 15 regenerants survive. There were differences among the regenerants in their resistance to bacterial wilt,3 regenerants resistant, 2 regenerant recovery (tolerant) and 10 regenerants were susceptibel.

Abstract

Tetraploid citrus are important for interploidal hybridization to create triploid seedless citrus.  Colchicine is the most commonly used as antimitotic agent to induce polyploid plants.  Tetraploid induction by colchicine in Pummelo ‘Nambangan’ was conducted in vitro using different types of explants.  The aim of this research was to induce tetraploid pummelo ‘Nambangan’ by colchicine treatment using germinated seed, shoot tip and cotyledonary node as explants.  Tetraploid shoot induction was conducted by soaking germinated seeds, shoot tips and cotyledonary nodes in 0.1% colchicine for 1, 3 and 5 h.  Regenerant shoots were grown on MS medium and their growth was observed after four weeks in culture.  Ploidy level was determined using flow cytometry analysis.  Stomata density, length and width of stomatal guard cell were also recorded. The results showed that shoot elongation was inhibited by colchicine treatment.  Soaking of shoot tip explants in 0.1% colchicine for 1 h resulted in 66.66% of putative tetraploid shoots.  Compared to diploid shoots, tetraploids had lower stomata density but bigger in guard cell size. Keywords: colchicine, tetraploid, pummelo (Citrus maxima (Burm.) Abstract

Untuk mengetahui beban traffic yang ada di diskominfo tenggarong. Penelitian ini tujuan yang ingin dicapai yaitu dengan pembagian jalur koneksi yang seimbang, maka diharapkan traffic dapat berjalan optimal, memaksimalkan throughput, memperkecil waktu tanggap dan menghindari overload pada salah satu jalur koneksi, menggunakan teknik load balancing dengan metode equal cost multi path (ECMP) pada diskominfo tenggarong. Pengujian parameter yang dilakukan ada 3, (1) Quality Of Service, (2) Round Trip Time, (3) Failover. Metodologi penelitian yang digunakan adalah menggunakan PPDIOO, (prepare, plan, design, implement, operate, and optimize). Hasil dari penelitian dapat disimpulkan bahwa, ISP 1 dengan kecepatan internet untuk download 7.36 megabit/s dan untuk upload 1.62 megabit/s. ISP 2 dengan kecepatan download 6.85 megabit/s dan untuk upload 2.24 megabit/s. Hasil pengujian quality of service, variabel jitter dengan niilai rata-rata 1,24 ms "Katagori Bagus". Variabel packet loss dengan niilai rata-rata 0,33% "Katagori Sangat Bagus". Variabel throughput 121% "Katagori Sangat Bagus". Berdasarkan standar TIPHON. Dengan nilai rata-rata pengujian quality of service adalah 40,8567 standar TIPHON "Sangat Memuaskan". Dalam pengujian round trip time, mempunyai nilai rata-rata 41,77777778 dan jumlah request time out (RTO) adalah 2. Hasil pengujian failover dihari senin,selasa dan rabu berhasil.

Abstract

The purpose of the research to determined the effect of soaking beef in various concentration of ginger blend and determined the best concentration of ginger blend on pH, water holding capacity, and cooking loss on beef. This research was conducted at the Laboratory of Animal Production and Reproduction, Department of Animal Science, Faculty of Agriculture, University of Lampung. Meat samples obtained from RPH Z-Beef Indonesia. The treatment used a completely randomized design (CRD) with four treatments and five replications. Treatments are in the form of concentration of ginger blend 0%, 10%, 20%, and 30% with immersion time for 50 minutes. The results obtained were analyzed by Analysis of Variant (ANOVA) at the 5% level and followed by the Least Significant Difference (LSD) test. The results showed that the addition of ginger blend with a concentration of up to 30% did not significantly influenced (P>0.05) on the pH value and water holding capacity, but had a significant effected (P<0.05) on the cooking loss of beef. Keywords: Ginger, Beef, Physic quality.

Abstract

The purpose of study is to know that physical quality of milk from Crossbreed Ettawa goats lactation period I—IV, those were organoleptic test, specific gravity test, and pH test in Sungai Langka Village Gedong Tataan Subdistrict Pesawaran District. This research was conducted method of purposive sampling with goat milk sampling as much as 250 ml on each lactation period I--IV and livestock recording data using questionnaire. Steps in this study begin from sampling through the milking process on each 15 Crossbreed Ettawa goats, followed by organoleptic test, specific gravity test, and pH test. The results of research, showed that quality milk which include organoleptic test in 1st lactation--4th lactation were color (white to yellowish white; white to yellowish white; white to yellowish white; and white yellowish), odor (Unpleasant to savory; Unpleasant to savory; Unpleasant to savory; savory), consistency (thin to slightly thickened; very thin to slightly thickened; thin to slightly thickened; and somewhat lumpy), taste (very bad to awful; very uncomfortable until a little tastier; very bad to awful; and tasty), receptivity (do not like to like; do not like to like; do not like to like; and like). pH test in 1st lactation--4th lactation that is 6,60; 6,59; 6,62; 6,54. Abstract

This research aims to know the efectivity of garlic by marination method on physical quality of broiler meat such as pH, water holding capacity, and cooking loss. Research was conducted on 8 Mei 2017 in Laboratory Animal Production and Reproduction, Animal Husbandry, Lampung University. The material used in this experiment was broiler meat from chicken slaughterhouse and chinese garlic blend. This research has done based Completely Randomized Design (CRD) with 4 treatments and 5 replications. The treatments is marinating with a blend of garlic 4% (T1), 8% (T2), 12% (T3), and non marinated (T0). Obtained data were analyzed with the assumptions of variance by 5% or 1%, then the result were significantly different in the advanced test using Least Significant Different (LSD). Results showed that marinating blend of garlic significantly affected (P<0,01) to water holding capacity and cooking loss, but did not significantly affect (P>0,05) to pH. Marinated broiler meat with garlic blend at a concentration of 12% yields the best water holding capacity and cooking loss. Keywords: garlic, broiler meat, marination, pH, water holding capacity, cooking loss.

Abstract

The purpose of the research was determined the effect of using coconut water fermentation on physical quality of broiler meat.This research was conducted on March, 30th2018 at Production and Reproduction of Laboratory, Department of Animal Husbandry, Faculty of Agriculture University of Lampung.The materials of this research is used 20 pieces chest of broiler meat and coconut water fermentation. The experimental design used was Completely Randomized Design (RAL) with 5 treatments and 4 replications.Treatment given in this research is P0 (without coconut water fermentation, P1 (coconut water fermentation 25%), P2 (coconut water fermentation 50%),P3 (coconut water fermentation 75%),and P4 (coconut water fermentation 100%). The observed variables is value of pH, water holding capacity (WHC), and cooking loss.The data obtained in this study was analyzed by using variance analysis (ANOVA), the results of which have significant effect on continued test using Least Significance Different (BNT).  The results showed that use coconut water fermentationdid not significant effect (P>0,05) to WHC and cooking loss of broiler meat, but use coconut water fermentation at concentration 100% have significant effect on value of pH of broiler meat. The best value of broiler meat pH that is on treatment of P4 (coconut water fermentation 100%) with average value that is 5,599. Key words: Coconut water fermentation, Physical quality of broiler meat, and  Broiler meat.

Abstract

Breeding mangoes is a long term activity and complicated. One of difficulties is polyembryonic seeds. These factors make genetic improvement through conventional parental selection and breeding slow and unpredictable. Adoption of molecular markers and genomics-based breeding strategies will likely improve predictability and breeding efficiency. The objective of this study was to evaluate the occurrence of zygotic or nucellar in polyembryony mango cultivars Garifta Merah, Lalijiwo, Manalagi, Madu, Saigon Kuning, and Saigon Merah using RAPD and SSR markers. The type of embryo (zygotic or nucellar) was evaluated by comparing the exhibiting amplification patterns, if different from the mother plant considered as zygotic and identified as nucellar if they exhibited the same banding pattern as the mother plant. Out of the 16 SSR and 16 RAPD primers evaluated, 9 primers each amplified the largest number of allele and sharply defined band. Dendogram of the SSR primers divide into three groups while RAPD primers distinguish clearly into two major groups, zygotic group and nucellar group. Based on zygotic and nucellar size, number and position, indicating no relationship between the type of embryo and embryo size, number and the position.

Abstract

Penelitian bertujuan mengetahui profil peternak dan performa kuantitatif sapi Peranakan Ongole (PO) betina kelompok poel satu dan poel dua di Kabupaten Lampung Timur dan Lampung Selatan. Penelitian ini dilakukan di Sentra Peternakan Rakyat (SPR) di Provinsi Lampung dari bulan September sampai Desember 2017. Peubah yang diamati untuk profil peternak yaitu Umur (U), Tingkat Pendidikan (TP) dan Pekerjaan(P). Performa kuantitatif meliputi bobot badan (BB), lingkar dada (LD), tinggi badan (TB), dan panjang badan (PB).Materi Penelitian sebanyak 199 ekor sapi Betina PO berbagai tingkatan umur dan 53 peternak di kedua SPR. Penelitian dilakukan dengan metode survei. Pengambilan data menggunakan purposive sampling dan wawancara (deep interview) kepada peternak. Data performa ternak untuk membandingkan di dua SPR menggunakan uji t. Hasil penelitian menunjukkan Peternak Sapi PO di SPR Lampung Timur lebih baik dibandingkan Peternak Sapi PO di SPR Lampung Timur. Performa kuantitatif sapi PO betina poel satu di Kabupaten Lampung Timur (BB252,86±68,81kg, LD 124,10±13,26cm, TB 146,24±11,25cm, PB 125,90±13,86cm) masing-masing lebih tinggi (P<0,05) daripada sapi PO betina di Kabupaten Lampung Selatan (BB 252,78±33,14kg, LD 120,39±10,34cm, TB 136,65±9,69 cm, PB 119,08±9,60cm), kecuali pada BB yang tidak berbeda nyata (P>0,05). Performa kuantitatif sapi PO betina kelompok poel dua di Kabupaten Lampung Timur lebih tinggi (P<0,05) daripada sapi PO betina di Kabupaten Lampung Selatan. Kesimpulan dari Penelitian ini bahwa peternak dan performa kuantitatif sapi PO betina di SPR Lampung Timur lebih baik daripada sapi-sapi PO betina SPR di Lampung Selatan.
