Abstract

One of the ways to establish a diagnosis of pulmonary tuberculosis is through laboratory investigations. The quality of the specimen is known to greatly affect the results of the examination. In order to ensure good quality specimens, sputum from pulmonary tuberculosis patients should be examined immediately after collection. However, conditions in the field sometimes result in delays in testing. This study aims to determine whether or not there is an effect of storage time for sputum specimens at room temperature on the results of acid fast bacilli assessment. The research was conducted at Udayana Tingkat II General Hospital Denpasar in January-April 2020. Type of research was a pre experimental design with a one-shoot case study approach. Sputum storage was carried out for 24 hours and 48 hours at a temperature of 22-250C. Acid fast bacilli examination was carried out using the Ziehl Neelsen staining technique and acid fast bacilli assessment was carried out based on the IUATLD scale. The research data were analyzed for normality of the data and Friedman's test. The results showed that there were differences in the results of the acid fast bacilli assessment on specimens immediately examined and those stored at room temperature for 24 hours and 48 hours. Based on the Friedman test, it was found that the P value was 0.00 (P <0.05) so that it was concluded that there was an effect of storage time for sputum specimens at room temperature on the results of acid fast bacilli assessment.

Abstract

ABSTRAK Pendahuluan: Bolu kukus merupakan jajanan pasar yang digemari oleh semua kalangan masyarakat. Jajanan ini memiliki daya simpan yang singkat. Penyimpanan bolu kukus lebih dari 3 hari dapat meningkatkan faktor resiko pertumbuhan jamur dan bakteri kontaminan. Tujuan penelitian ini adalah untuk mengetahui cemaran Angka Lempeng Total (ALT) dan Angka Kapang Khamir (AKK) pada bolu kukus yang di produksi oleh prodeusen di Desa Bebalang Bangli dengan lama penyimpanan 3 hari pada suhu ruangan (270C) dan mengidentifikasi jamur apa saja yang mengkontaminasi bolu kukus. Metode: Penelitian yang digunakan adalah eksperimental, sampel yang digunakan adalah bolu kukus yang dibuat oleh produsen yang kurang dari 18 jam. Jumlah sampel yang digunakan adalah 15 buah bolu kukus dari 3 produsen rumah tangga. Hasil: Angka Lempeng Total (ALT) pada produsen A sebanyak 897×104 CFU/mL, Produsen B sebanyak 86×104 CFU/mL, Produsen C sebanyak 167×104 CFU/mL.Hasil penelitian Angka Kapang Khamir (AKK) didapatkan hasil yang melebihi batas standar SNI 7388-2009, menyatakan standar Angka Kapang Khamir (AKK) pada roti yaitu batas maksimum 1×104 koloni/g. Hasil Angka Kapang Khamir (AKK) Produsen A sebanyak 1,85×104 CFU/mL, Produsen B sebanyak 8,4×104 CFU/mL, Produsen C sebanyak 12,5×104 CFU/mL. Hasil pengamatan jamur kontaminan didapatkan 3 spesies jamur yaitu Aspergillus flavus, Aspergillus fumigatus dan Rhizopus stolonifer. Diskusi: Hasil penelitian yang telah dilakukan didapatkan hasil yang melebihi batas standar SNI 7388-2009 yang sudah ditetapkan yaitu ALT dan AKK batas maksimum 1×104 koloni/g. Kata kunci : Angka Lempeng Total (ALT) Angka Kapang Khamir (AKK), bolu kukus, Aspergillus flavus, Aspergillus fumigatus dan Rhizopus stolonifer. Introduction: Steamed cake or “bolu kukus” is one of the most favorite cake in traditional market. Steamed cake has a short shelf life, in more than 3 days of storage it may increase the risk factors of bacteria and mold contamination. The aim of this research is to know bacterial and mold contamination of traditional steam cake using standard plate count and yeast and mold plate count after 3 days of storage (270C) and also to identify type of mold in sample. Method: Type of this study is experimental study, using 15 steamed cake as a sample. Sample was taken from 3 traditional steamed cake producer in Bebalang Village, Bangli and stored in room temperature (270C). Result: Meanwhile in sample the total bacteria using standard plate count are 897x104 colony/ g (A producer), 86x104 colony/ g (B producer) and 167x104 colony/g (producer C). Yeast and mold plate count also show that contamination are more than SNI 7388-2009 (1x104 colony/g). The result shows that yeast and mold contamination are 1.85 × 104 colony /g (A Producer), 8.4 × 104 colony/g (B producer) and 12.5 × 104 colony/g (C Producer). Identification result shows that mold contaminant in traditional steamed cake is Aspergillus flavus, Aspergillus fumigatus and Rhizopus stolonifer. Discussion: The result of this study show that total bacterial and fungi of steamed cake after 3 days of storage are more than standard using Standar National Indonesia 7388-2009 (SNI 7388-2009). The maximum contaminant level of bacteria using SNI 7388-2009 is 1x104 colony/g. Abstract

Intestinal microbial dysbiosis is one of the triggers for metabolic syndrome. The increase in pathogenic bacteria triggers the high release of lipopolysaccharides which underlie the incidence of insulin resistance in diabetes mellitus. It is important to modulate the intestinal microbiota towards eubiosis through consumption of probiotics. A potential probiotic product is soy yogurt which contains live bacteria and high protein and isoflavones. The aim of this study was to examine the effect of soy yogurt supplementation on the abundance of cecum microbes (Lactic Acid Bacteria (LAB) and Escherichia coli) in diabetic rats. The research was conducted at the Laboratory of Stikes Wira Medika Bali. Type of research is experimental with a completely randomized design consit of 6 groups. The conditioning of diabetes mellitus was done by induction of nicotinamide (NA) 230 mg / kgBW and streptozotozin (STZ) 65 mg / kgBW. Abundance of cecum microbes analysis using Total Plate Count. The results showed soya yogurt supplementation in rat with diabetes mellitus had a positive correlation in improving the abundance of microbial cecum. The total abundance of LAB in diabetic rat was 7.36 ± 0.15 Log10 CFU/mL and was significantly different (p <0.05) compared to diabetic rat treated with soy yogurt supplementation, namely as much as 8.08 ± 0.1 Log10 CFU/ml. Meanwhile, the abundance of E. coli in diabetic rat was 9.42 ± 0.12 Log10 CFU/mL and was significantly different (p <0.05) compared to rats treated with soy yogurt supplementation, namely 8.14 Log10 CFU/ml.

Abstract

Soft rot bacteria infection in carrot tuber (D. carota L.) causes severe economic losses. Soft rot disease can be caused by various bacteria belonging to Enterobacteriaceae. This study aimed to isolate and identify bacteria as causal agent of soft rot disease in local carrot variety in Bali. Samples were collected at Badung Tradisional Market, Denpasar, Bali. Isolation was carried out by serial dilution method (Platting Method). Eight bacteria (BL1, BL2, BL3, BL4, BL5, BL6, BL7 and BL8) were isolated from soft rot tuber. BL6 isolate showed positive result in Postulat Koch test that caused soft rot on carrot tuber. The result of identification by Microgen™ GnA+B-ID System and identification book Bergeys’s Manual of Determinative Bacteriology Ninth Edition (Holt et al., 1994), BL6 was identified as Citrobacter.

Abstract

Perilaku merokok berkorelasi positif dengan meningkatnya angka kejadian infeksi oleh Methicillin Resistant Staphylococcus aureus atau dikenal dengan MRSA. Perokok dilaporkan memiliki tingkat kolonisasi MRSA lebih tinggi dibandingkan non-perokok. Hal tersebut akan sangat berbahaya karena infeksi MRSA dapat menyebabkan infeksi parah seperti infeksi aliran darah, infeksi paru-paru atau necrotising fasciitis (kerusakan jaringan). Penelitian ini bertujuan untuk melakukan deteksi awal keberadaan MRSA yang berasosiasi dengan perokok aktif. Responden pada penelitian ini adalah perokok aktif yang secara rutin melakukan konseling di Klinik Berhenti Merokok (KBM) Puskesmas 1 Denpasar Utara yang memenuhi kriteria inklusi dan eksklusi. Pengujian bakteriologi dilakukan di Laboratorium STIKES Wira Medika Bali dengan melakukan kultur terhadap spesimen apus tenggorokan responden. Identifikasi MRSA dilakukan dengan melakukan pengujian sensitivitas dengan metode Kirby Bauer menggunakan cakram antibiotik oxacillin OX 1 µg. Hasil isolasi dan identifikasi S. aureus pada sampel apus tenggorokan 13 responden didapatkan hasil sebanyak 4 isolat (30,8%) teridentifikasi sebagai S. aureus sedangkan sebanyak 9 (69,2,8%) sampel menunjukkan hasil negatif. Hasil uji sensitivitas terdapat 2 isolat yang memiliki daya hambatan intermediate yaitu SA1 dan SA2 dengan diameter daya hambatan rata-rata sebesar 12 mm dan 11,83 mm. Berdasarkan hasil tersebut disimpulkan bahwa tidak ditemukan bakteri MRSA pada sampel usap tenggorokan perokok aktif pasien konseling di KBM Puskesmas 1 Denpasar.

Abstract

Onychomycosis merupakan infeksi pada lempeng kuku yang dapat disebabkan oleh jamur dermatofita (Tinea unguium), non dermatofita atau yeast. Infeksi onychomycosis menyebabkan kerusakan pada kuku yang menyebabkan lempeng kuku menebal, rapuh dan mudah hancur. Penelitian ini bertujuan untuk gambaran onychomycosis pada lansia di Panti Sosial Tresna Werdha (PSTW) Wana Seraya. Jenis penelitian yang digunakan adalah penelitian deskriptif. Pengambilan sampel penelitian dilakukan di Panti Sosial Tresna Werdha (PSTW) Wana Seraya di Jl. Gemitir No.66, Kesiman Kertalangu, Denpasar Timur, Kota Denpasar, Bali. Sampel dianalisa di Lab. Parasitologi Teknologi Laboratorium Medis STIKes Wira Medika Bali. Pemeriksaan menggunakan teknik pengamatan langsung menggunakan pewarna kalium hidroksida (KOH) 10% pada kerokan kuku 15 orang responden yang memenuhi kriteria inklusi dan eksklusi. Hasil pemeriksaan menunjukkan terdapat 1 responden (6,67%) yang positif onychomycosis dengan ditemukannya hifa pada pengamatan mikroskopis preparat kerokan kuku. Sementara itu 14 sampel lainnya (93,33%) dinyatakan negatif. Hasil screening pada penelitian ini menunjukkan lansia di PSTW Wana Seraya telah memiliki hygiene diri yang baik khususnya dalam memelihara kebersihan kuku kaki dan tangan sehingga mengurangi faktor resiko terjadinya infeksi jamur kuku.
