Abstract

This research aims to evaluate the safety of dexamethasone use as an anti-inflammatory and analgesic on the handling of femur fracture of rats by bone graft method. The sample used was 32 male Wistar rats aged 2 months were divided into two groups, 16 rats for each. Group I was fractured rats and fixed with intramedullary pin without bone graft, whereas Group II was fractured and fixed with intramedullary pin and demineralized freeze-dried bovine bone xenograft (DFDBBX). Group I and II were then divided into two groups for each, Group IA, IB, IIA, and IIB, eight rats for each. Group IA and IIA were not injected with dexamethasone, whereas Group IB and IIB were injected with dexamethasone 0.5 mg/kg BW/ day intramuscularly. At the end of the first week, blood was collected from eight rats (two rats for each) of Group IA, IIA, IB, and IIB for total leucocytes and blood Calcium analysis. Urine was also collected for urine Calcium analysis. Rats were then killed to collect the femur for histopathologic analysis using HE staining and immunohistochemistry to analyze the expression of TGF?-1 using Streptavidin-biotin method. Subsequent sampling was conducted at the end of 2nd, 4th, and 8th weeks of treatment, respectively. The total data of leucocytes, blood, and urine Calcium were statistically analyzed using a 2x2x4 factorial method, while the histopathologic and immunohistochemistry specimens were analyzed descriptively under the microscope based on the alteration of the bones. The statistical analysis showed that dexamethasone increased the concentration of urine Calcium (p<0,05), the concentration of blood Calcium insignificantly (p>0,05) affected by dexamethasone but cause homeostatic calcium disorder and the total leucocytes significantly (p<0,05) affected by grafted materials. Histopathologic analysis of the femur showed that there was an increased infiltration of inflammatory cells in the fracture area from the group untreated with dexamethasone. Whereas the immunohistochemistry analysis showed positive results from all groups. Based on the whole analysis, it can be concluded that although dexamethasone depressed inflammation process, it can also disturb the fracture healing process.

Abstract

House flies (Musca domestica L.) are one of the major pests found in poultry farms resulting in not onlyannoyance and indirect damage to the poultry production but also transmitting many infectious organismsmechanically and biologically. A highly pathogenic avian influenza virus H5N1 (HPAIV H5N1) collectedfrom field house flies in Java Island, have fully succeeded identified and isolated in 2008. The AIV H5N1isolates were further used in the present study to determine the presence and persistence of the AIV H5N1in the laboratory infected house flies. One hundred house flies from a free AIV poultry farm in Yogyakarta,Indonesia were used in this study. The collected house flies were fasted for 12 hours then divided equallyin control and treated groups. The treated group was allowed to imbibe Dulbecco’s modified eagle medium(DMEM) containing AIV H5N1 while the control group imbibed medium without virus for one hour. Theflies from each group were collected at 12 and 24 hours post-exposure, respectively. All flies were immobilizedat 4°C, immersed in absolute ethanol for a few seconds and dissected under the stereomicroscope to collectthe gastrointestinal (GI) tracts. Based on the RT-PCR results, it is concluded that AIV H5N1 remainedexist in GI tracts of house flies for at least 24 hours post-exposure.

Abstract

The objective of this study was to analyze the influence of chitosan on coronary artery wall thicknessand lumen diameter of Sprague Dawley rats induced by high fat ration. The animals for this study weretwenty adult male rats divided into four groups, i.e. group I as the control was fed with basal rationcontaining normal fat for 3 months, group II was fed ration containing high fat for three months, group IIIwas fed ration containing high fat and given chitosan 180 mg per kg body weight per day orally in 2 mLaquadest for 3 months, group IV was fed ration containing high fat for three months and after one monthgiven chitosan 180 mg per kg body weight per day orally in 2 mL aquadest for two months. Each groupconsisted of five animals. After 90 days, the rats were necropsied and the hearts were collected tohistopathological. The difference between treatments was statistical analysis by Anava, and continued byLowest Significant Difference Test to locate the difference. Normal fat ration did not influence coronaryartery wall thickness and coronary artery lumen diameter in rats. High fat ration increased coronaryartery wall thickness and chitosan could be decreased coronary artery wall thickness in rats. High fatration + chitosan that given simultaneously could be increased coronary artery lumen diameter in rats.Based on the result of this study, it was concluded that high fat ration was a major factor able to causeatherosclerosis and chitosan was able to prevent atheroma plaque formation.

Abstract

The objective of this study was to analyze the influences of chitosan on C-Reactive Protein expression in aorta and coronary artery of Sprague Dawley rats by high fat induction. The animals for this study were 20 adult male rats divided into four groups, i.e. group I as the control was fed with basal diet containing normal fat for 3 months, group II was fed diet containing high fat for 3 months, group III was fed diet containing high fat and given chitosan 180 mg per kg body weight per day orally in  2 ml aquadest for 3 months, group IV was fed diet containing high fat for 3 months and  after 1 month given chitosan 180 mg per kg body weight per day orally in 2 ml aquadest for 2 months. Each group consisted of five animals. After 90 days, the rats were necropsied and the hearts were collected to histopathological and immunohistochemical analysis by immunohistochemistry streptavidin-biotin method. C-Reactive Protein expression in aorta was negative. Chitosan was able to prevent atheroma plaque formation in coronary artery and CRP may involve in atherosclerosis.

Abstract

Disorder of carbohydrate metabolism in the Diabetes Mellitus patients resulted in increasing of cholesterol synthesis. This condition increases the risk of heart attack or coronary heart disease, therefore it is recommended to lower fat intake. One of the alternatif to lower fat intake is by replacing fat with fat replacer in the diet. The aim of the present study is to evaluate the effect of replacing 75% of fat with SOPE on the glucose and lipid profile in diabetic-hipercholesterolemic rats. Eihgteen Sprague Dawley rats were devided into 2 groups of 9 rats. They were fed with hipercholesterol diet and were diabetic induced with alloxan IM injection. One group was fed with 75% SOPE substitute the fat in the standard diet and the other group was fed with standard diet. The result indicates that the oil substitution of standard diet of Sprague Dawley rats for sorbitol-oleic polyester as much as 75% gives an advantage to rats which suffer hiperglikemic-hypercholesterolemic because it decreased the glucose, total cholesterol, LDL cholesterol, and LDL/HDL cholesterol ratio, and did not decrease the HDL cholesterol, but increased triglyceride serum.

Abstract

Cats are pets that many people, including in Yogyakarta, are keeping. Various kinds of diseases can attack cats, but not all owners are able to check their cat's health status. This community service aims to carry out health checks for cats owned by the community especially animal owners who are customers of the Veterinary Clinic of Internal Medicine, FVM-UGM. The service was carried out from April to October 2022 by checking the cat's health status directly. The service has been conducted and can carry out examinations and therapy of 20 cats. Based on the examination, it was found that 17 cats (85%) were diagnosed as sick and three healthy cats (15%). The three main diseases found were skin, digestive and urinary systems. Through medical examinations by students accompanied by veterinarians in the Department of Internal Medicine, cat owners have been invited to watch and given information regarding disease diagnosis, therapy, and how to administer medicine. The owner already understands the cat’s health status, agree to the action and treatment and can provide medicine for the sick cat. It was concluded that the community service for cat health examination had been successfully carried out on 20 cats, and the owner knew the condition of the cat and was able to give the medicine given.
