Abstract

Aim of the research was to find out the viability ofPediococcus acidilactici U318 in inoculum powder duringstorage. The filler material used in the production ofinoculum powder was three kinds of flour, namely maizena,rice flour and tapioca. The research was done in laboratoryscale as experiment research which was designed byrandomized block design with factorial experiment. Twofactors were experimented, namely the kinds of flour as afiller material as the first factor and the storage period as thesecond factor.Result of the research showed that the treatment of flouras a filler material did not influence the viability of P.acidilactici U318 in inoculum powder. Any filler materialused in production of the powder gave same protective actionon its viability. The viability of P. acidilactici U318decreased during storage period. After storage of 4 weeks theaverage of viable P. acidilactici U318 was about 2,54x1010cfu/g which decreased one log cycle from the initial ofstorage.

Abstract

PENGARUH UKURAN PARTIKEL DAN SUHU EKSTRAKSI TERHADAP KARAKTERISTIK EKSTRAK PEWARNA ALAMI BUNGA KENIKIR (TEGETES ERECTA L.). Bunga kenikir marigold berpotensi sebagai sumber pewarna alami. Warna kuning hingga oranye diperoleh melalui proses ekstraksi. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh ukuran partikel dan suhu ekstraksi terhadap ekstrak pewarna alami bunga kenikir serta menentukan perlakuan terbaik untuk mendapatkan ekstrak pewarna alami bunga kenikir marigold. Rancangan dalam penelitian ini menggunakan Rancangan Acak Kelompok dengan dua faktor. Faktor I adalah perlakuan ukuran partikel yang terdiri dari tiga taraf yaitu 40, 60, dan 80 mesh. Faktor II adalah perlakuan suhu ekstraksi yang terdiri dari tiga taraf yaitu 30±2 ºC, 45±2 ºC, dan 60±2 ºC. Hasil penelitian menunjukkan bahwa perlakuan ukuran partikel dan suhu ekstraksi berpengaruh sangat nyata (P<0,01) terhadap rendemen, total karotenoid, tingkat kecerahan (L*), tingkat kemerahan (a*), dan tingkat kekuningan (b*). Interaksi kedua perlakuan berpengaruh sangat nyata (P<0,01) terhadap rendemen, total karotenoid, tingkat kecerahan (L*), tingkat kekuningan (b*) dan berpengaruh nyata (P<0.05) terhadap tingkat kemerahan (a*). Ukuran partikel 60 mesh dan suhu ekstraksi 45±2 ºC merupakan perlakuan terbaik untuk menghasilkan ekstrak pewarna bunga kenikir dengan karakteristik rendemen 8,79%, kadar total karotenoid 21,52%, tingkat kecerahan (L*) 21,05, tingkat kemerahan (a*) 11,43, dan tingkat kekuningan (b*) 19,66.

Abstract

The aim of the research was to characterize of the six species of Lactic Acid Bacteria (LAB) indigenous (Lactobacillus acidophilus KBc, Lb. salivarius KBd, Lb. brevis KBa, Lb. delbrueckii KBe, Lb. plantarum KBb and Lactococcus lactis KBf) isolated from the fermented horse milk of Bima. The tests were done to cover safety aspect, ability to survive on low pH condition, bile salt tolerance, ability to adherence and colonization, and effect on cholesterol level reduction. The six LAB species were not pathogen and invasive, and were able to grow well on media which contained bile salt of 0.75 %. Lactobacillus  acidophilus KBc and Lb. brevis KBa had ability to survive on media pH 2.5 for 3 hours and were able to adherence and colonization on gut mucosa epithel. Lactobacillus acidophilus KBc and Lb. brevis KBa had ability to reduce cholesterol level of blood serum of rabbits on hypercholesterolemia condition significantly up to 53.74 % and 51.70 %, respectively. Based on the characterization of the six species LAB, only Lactobacillus acidophilus KBc and Lb. brevis KBa can be used as probiotic.ABSTRAKPada penelitian ini telah dilakukan karakterisasi terhadap enam spesies bakteri asam laktat  (BAL) indigenous (Lac- tobacillus acidophilus KBc, Lb. salivarius KBd, Lb. brevis KBa, Lb. delbrueckii KBe, Lb. plantarum KBb dan Lac- tococcus lactis KBf) yang diisolasi dari susu kuda liar Bima terfermentasi. Pengujian yang dilakukan meliputi aspek keamanan, ketahanan hidup pada media pH rendah, kemampuan tumbuh pada media yang mengandung garam em- pedu, kemampuan menempel dan berkolonisasi, dan efek terhadap penurunan kadar kolesterol serum darah. Keenam spesies BAL tersebut tidak patogen dan invasif, serta dapat tumbuh baik pada media yang mengandung garam empedu 0,75 %. Lactobacillus  acidophilus KBc dan Lb. brevis KBa tahan hidup pada media pH 2,5 selama 3 jam serta dapat menempel dan berkolonisasi pada epitel mukosa usus. Kedua bakteri ini mampu menurunkan kadar kolesterol serum darah kelinci dalam keadaan hiperkolesterolemia secara nyata masing-masing sebesar 53,74 % dan 51,70 %. Berdasar- kan karakterisasi keenam spesies BAL tersebut, Lb. acidophilus KBc dan Lb. brevis KBa dapat dipergunakan sebagai probiotik.

Abstract

Candidacies in female reproductive tract are mainly caused by Candida albicans. This infection often causes serious problems, particularly on their reproductive tract (genital part). Until recently, control of this infection has relied on the use of antibiotics. However due to numerous bad side effects of antibiotics, lactic acid bacteria have been proposed as an alternative method to control the growth of Candida albicans. Therefore, this research was aimed to isolate, screen, and characterize lactic acid bacterial isolates (LAB) antagonistic against Candida albicans (the causative agent of candidacies infection in reproductive tract of human). LABs were isolated from various fermented foods, such as tape ketan and kimchi. Isolation of LABs was conducted by applying dilution and spread plate method on MRS agar medium supplemented with BCP indicator to distinguish LABs from non acid-producing bacteria. Colonies with indication to produce acid were screened for antagonistic activity against C. albicans on MRS agar and followed by characterization of those isolates (Gram stain, catalase production test, oxydase production, gas production test, resistance test to low pH conditions and to high level of NaDC (sodium deoxicolic), and test for ability to convert colic acid (CA) into deoxicolic acid (DCA)). The results showed that 46 LAB isolates were successfully isolated from samples of tape ketan and kimchi. Among those, 7 isolates showed antagonistic activity against C. albicans in in vitro tests. All these 7 candidates were also found to be resistance to low pH conditions (up to pH 2) and to high level of NaDC (up to 0.6 mM). Four most potential isolates were further testes for ability to convert colic acid into deoxycolic acid and none showed positive result, indicating that they all showed initial potential and safe for future human probiotic development (especially to be used to treat patients infected by C. albicans).

Abstract

Lactic Acid Bacteria (LAB) is a bacterium that has an ability to inhibit a pathogenic bacteria growth, including isolates 18A. The aim of this study is to determine the species of LAB isolates 18A by conventional tests and API 50 CH test. This study was a descriptive observational study, with lactic acid bacteria isolates 18A were obtained from bali cattle’s colon as an object. LAB isolates 18A were re-cultured on MRS media, catalase test, and Gram staining. Identification test were performed by CO2 production test, growth in the presence of 15% NaCl, growth at 10°C, and growth at pH 9.6. Confirmation test were performed using API 50 CH (Standard Analytical Profile Index). The results indicated that LAB isolate 18A were able to grow well on MRS broth media, catalase-negative, Gram-positive, and spherical-chain in shape. Identification test revealed that the bacteria is homofermentative, were able to grow at 10°C, and do not grow at pH 9.6. Fermentation test using API 50 CH showed that LAB isolates 18A were able to degrade 15 sugar components, consist of no: 5, 10, 11, 12, 13, 22, 24, 25, 26, 27, 28, 29, 30 , 31, and 39. Based on conventional identification results, it can be concluded that BAL isolates 18A is a Lactococcus. In addition, the result of API 50 CHL test showed that BAL isolates 18A is a Lactococcus lactis ssp lactis 1.

Abstract

This study aimed to determine the ability of galactomannan extract from coconut pulp to stimulate the growth of lactic acid bacteria and to determine the concentration of galactomannan from coconut pulp that could stimulate the growth of lactic acid bacteria with the best growth rate. The experiment was conducted in vitro where the growth medium used was MRS Broth without glucose but added with galactomannan from coconut pulp. MRS Broth media with glucose was used as a control with a concentration of 1%. The galactomannan concentrations used were 0%, 0.5%, 1%, 1.5%, and 2% (w/v). Determination of the confirmed time was carried out by measuring the optical density (OD) on 1.5% galactomannan extract media once every 3 hours. The results showed that galactomannan from coconut pulp can stimulate the growth of lactic acid bacteria. Total lactic acid bacteria related to the degree of acidity and total acid. The more total bacteria, the lower the pH value, thus the total acid will increase. In the media that was added galactomannan from coconut pulp, the best total lactic acid bacteria, pH and total acid were obtained from a concentration of 1% (w/v) with 9 hours incubation, the highest total lactic acid bacteria was 4,5 CFU/mL. Abstract

Dietary fiber with excellent fermentability can be categorized as prebiotics, one of which was green grass jelly. The objective of this research was to know the concentration of green grass leaf extract powder to stimulate the growth of Lactic Acid Bacteria (Lactobacillus casei subsp. rhamnosus). In vitro experiments were performed in laboratory with a concentration treatment of green grass leaf powder and glucose as a control. The medium used was yeast-peptone medium added with 1% glucose, 0% green grass leaf extract powder, 0.1% green grass leaf extract powder, 0.2% green grass leaf extract powder, 0.3% green grass leaf extract powder, 0.4% green grass leaf extract powder, and 0.5% green grass leaf extract powder. Fermentation was carried out at 37oC with a fermentation time of 9 hours. The experimental results showed that the concentration of green grass leaf powder extract of 0.2% was the best, in performance to stimulating growth of LAB. This concentration of the extract was the critical concentration, since the process of medium solidification was occured at the extract concentration of more than 0.2%. Abstract

Tabah bamboo shoot pickle is one of the fermented food which is the source of lactic acid bacteria. Lactic acid bacteria (LAB) is beneficial to health because it has the ability as a probiotic. Lactic acid bacteria that have probiotic criteria should have resistance to low pH and bile salts. This study aims to determine isolates of lactic acid bacteria isolated from tabah bamboo shoot pickle resistant to low pH and bile salts (NaDC). Lactic acid bacteria were tested to low pH by using MRS broth that have different pH (pH 2, pH 3, pH 4 and pH 6.2 as a control) incubated at 37ºC for 3 hours. isolates were survive in low pH then continued in bile salt resistance test with 0.3% bile salt concentration for 15 minutes, 30 minutes, 45 minutes, 60 minutes and 24 hours. The results showed that three isolates out of 88 isolates had ability to grow in low pH and in medium supplemented by NaDC 0,3%. Abstract

This research aimed to isolate some of potential bacteria which could degrade dibenzothiophene (DBT). The bacteria were isolated from the soil which has been contaminated by petroleum for many years in Buluh Telang Langkat North Sumatera. This experiment was also conducted to find out the isolate that had the highest degradation rate of DBT. The results showed that the isolated bacteria had a different ability to degrade dibenzothiophene with OD660 ranged from 1.100 to 1.137 and the degradation rate ranged from 42.62% to 69.88%. The isolate of LSU20 had the highest ability to degrade DBT. The isolated bacteria which was incubated for 96 hours at 45 o C in shaking condition at 150 rpm could degrade 69.88% of 200 ppm DBT in tetradecane.
