Abstract

Oocyte vitrification is the important part of gamete preservation for further purpose. The objective of this study was to evaluate the response or development of vitrified-mice oocyte following activation using various concentrations of Strontium Chloride (Sr Cl2). Oocytes were collected from superovulation-induced female mice. Oocytes vitrification was then performed using a gradual equilibration of 2 M Ethylene Glycol in 0.25 M sucrose and 7 M Ethylene Glycol on 0.5 sucrose. Subsequently, the vitrified oocytes were thawed and activated using various Strontium Chloride concentration in each group. Control 1 is unvitrified oocyte and without Sr Cl2. Control 2 is unvitrified oocyte then activated by 20 mM Sr Cl2. Zero (0) mM Sr Cl2 is vitrified oocyte without Sr Cl2. Group Ten (10) mM is vitrified oocyte then activated by 10 mM Sr Cl2. Group Twenty (20) mM is vitrified oocyte then activated by 20 mM Sr Cl2. The viability of vitrified-thawed oocytes was observed based on ooplasm integrity. Whereas the oocytes respond to artificial activation was observed based on pronucleus formation after 10 hours of activation. The result showed that 39% of oocyte degenerated following vitrification. The respond of vitrified-thawed oocytes following artificial activation using Strontium Chloride was significantly lower compared to fresh oocytes (p<0,05). Interestingly the highest percentage of activated oocytes (36.36%) was present in a group achieved 20 mM Strontium Chloride. As conclusion is Strontium Chloride 20mM has a best result (36,36%) to activate vitrified oocyte than 0 mM and 10 mM of Strontium Chloride.

Abstract

The aim of this study was to understand the effect of using Zinc (Zn) to oestrous synchronization in Indonesian Mixed Breed Goat. The number of 12 does aging 24-36 months and a buck were used in this experiment in order to natural copulation purpose. The experimental animals were devided into 2 groups. The first group was given oestrous synchronization using polyurethane sponge which contained of medroxi progesterone acetate 60 mg through intra vaginal for 14 days (control). The Second group was synchronized in the same method as the first group and supplemented with Zn 20 mg/head by peroral. The does that have been oestrous then mated naturally. The experimental results indicate the number of natural copulation in second group was higher than control (83% vs 50%). Oestrous uniformity occurred in both groups. The Oestrous onset in second group was earlier than control (36 vs 48 jam) and duration of oestrous was longer than control (12 vs 48 jam).Maximum oestrous quality score was higher than control (7.17±0.75 vs 6.67±0.52). The conclusion is the addition of Zn Supplement improves the quality of oestrous during oestrous synchronization by intra vaginal polyurethane sponge which contained of medroxi progesterone acetate 60 mg in Indonesian mixed breed goat.

Abstract

Peranakan Etawa (PE) goat farming is one of the growing livestock sectors in Indonesia. Indonesia needs a sustainable breeding system that can guarantee the availability of goats. Estrus synchronization is a necessity that can help uniform the breeding time and manage the time of pregnancy and birth. Artificial estrus synchronization techniques that use a combination of Prosterone and Zinc minerals give good results of estrus synchronization in goats. The Aim of this research is to understand the combination efectiveness of estrus synchronization technique with progesterone acetate and Zinc supplementation. The design of this study used two treatment groups, each group consisting of three does PE goats. Estrus synchronization of experimental animals in treatment 1 and 2 was carried out using polyurethane sponge implant method which contained medroxi progesterone acetate 60 mg. Group 1 was given Sponge Progesterone for nine 9 days while giving Zinc (Zn) mineral supplement (Elemental Zn 20 mg, Kimia Farma) orally given 7 times. The frequency of Zinc administration every day begins during intra vaginal sponge insertion. Group 2 was treated same as group 1 with 14 days of implant time and 7 times of Zinc supplementation.The results showed that both treatments occurred estrus simultaneously with good estrus quality. Both treatments show the results of the estrus 8 score assessment with the estrus 3-9 range. The maximum estrus quality’s value of group 2 was higher (8.33 ± 0.57) compared to group 1 (8.0 ± 1.0). The estrus duration of group 2 occurs for 24 hours and group 1 for 7 hours. Estrus onset of Group 2 is earlier (58 hours) than group 1 (75 hours). Both treatment groups showed an increase in temperature at period of the occurrence of estrus. Body temperature during estrus time was 38.7 ± 0.20 0C to 39.60 ± 0.26 0C. The conclusion of this study is that in both treatments of estrus synchronization using sponge progesterone and zinc supplementation can cause estrus synchronization effectively . The duration of estrus in group 1 (9 days of implant) was Shorter than group 2 (14 days of implant). The onset of estrus time of group 1 (9 days of implant) was longer than Group 2 (14 days of implant). The body temperature of the experimental animals increases with the occurrence of estrus.

Abstract

Peranakan Etawa (PE) Goat is a type of superior goat that is very potential to be maintained as a dairy goat. Food safety guarantee of animal origin becomes very important in order to prevent and spread disease from animal to human. So it takes an effort in terms of quality control of food of animal origin, especially goat's milk. This study aims to determine the quality of PE goat milk which is maintained by farmers in small farms. The study was conducted on 15 samples of goat milk collected in the region of Samigaluh, Kulon Progo. The research used a method of testing the quality of milk in the laboratory that includes testing the physical condition of milk and milk composition. Testing the physical condition of milk consists of testing of cleanliness, color, odor, and taste. Testing of milk composition consist of degree of acid, reductase, fat content, calculation of total bacteria by Total Plate Count (TPC) method. The data obtained are then analyzed descriptively. The results showed milk samples were clean, yellowish, typical odor of fresh milk, and had a slightly sweet taste. Mean of acid degree 7,1°SH, mean of reductase rate 2 hour, mean of fat content 4,53% and mean total bacteria 2,8 x 105 CFU ml. These results indicate that the quality of goat milk included in the category worth to be consumed with sufficient quality. The quality of goat's milk is influenced by the type of feed given and the nutrient composition in the feed type.

Abstract

Reproduction management Socialiszation to “Subur” Perananakan Etawah (PE) Goat Farmer Comunitee in Hargo Tirto, Kokap, Kulon Progo, Yogyakarta was focused on reproduction problems in PE Goat. The main issue that should be priority is Goat Reproduction. Good Dairy goat management reproduction will give positive effect to success of milk production. The back ground of animal husbandary comunitee is located in mountain range of Kulon Progo, their main job activity are as a coconut sugar farmer. The Subur Animal Husbandary Comunitee is grouped as a new dairy goat farmer. They still trying to learn how to develop their husbandary, do not have adequate knowledge of good management farming and good information about reproduction.  That condition are the reasons to choose Subur Animal Husbandary Comunitee as a target for Socialiszation in presentation and discussion to farmers. Specific explanation and direct practice in field and does oestrus observation. Distribution of feed, medicine, vitamin and mineral were given to Subur Animal Husbandary Comunitee. The results of this program give many positive impact to the farmers and dairy goats such as; the farmers understand and be able to detect signs of oestrus which is very important to them, the farmers have apply detection of oestrus by visual and buck as a natural oestrus detector, natural mating and pregnant occurrence increased after the farmers have applied oestrus detection in does of PE Goat.
