Abstract

Soft coral Nepthea sp. grows in the seas of South-East Sulawesi, Indonesia. However, information on the chemical and pharmaceutical aspects of this genus is still limited. Therefore, this research aims to explore the chemical contents and biological activities of Nepthea sp. The sample was collected from the waters of Saponda Island by SCUBA diving. It was extracted by ethyl acetate and fractionated using vacuum liquid chromatography. The chemical content was analyzed by phytochemical screening, LC-MS/MS analysis, Total Phenolics Content and Total Flavonoids Contents. Antioxidant potency was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radicals and ABTS (2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid). Cytotoxicity property was analyzed by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays. The result showed that the fractionation of Nepthea ethylacetate extracts produced six fractions (A-F). Fractions A and B contain non-polar compounds. Based on LC-MS/MS data, the non-polar compounds in Fraction A and B include achillin, atractylenolide II, buthyl isobuthyl phthalate, rengyolester, 2a-acetoxycostic acid, ocotillol acetate, petasitolone and some unidentified compounds that are C33H58O4, C15H21NO, C21H33NO, and C16H20O4. In general, the antioxidant and cytotoxic properties of all samples are in the weak category, however, when examined for each sample, the antioxidant properties of fraction B is slightly better than fraction A based on the IC50 value of DPPH and ABTS. Cytotoxicity of Fraction A is better than Fraction B against Breast Cancer cell lines MCF-7. The non-polar fraction of Nepthea sp. can be developed as raw material for the discovery of new compounds, antioxidant and anticancer agents, especially breast cancer.

Abstract

Reactive oxidative stress (ROS) can lead to cell damage, and one of them is the kidney's cell. Etlingera elatior (Jack) R.M. Smith can be utilized as an agent that can protect the cell from ROS. This study aimed to investigate the protective effect of E. elatior fruit on the kidney's cell. We used experimental animals which were treated with Na-CMC (Group I), Na-CMC (Group II), the extracts of E. elatior fruit 200, 300, 400 mg/kg BW for Group III, IV, and V, respectively, for seven days. The blood was collected after treatment. At day 8, group I, III, IV, and V were induced by CCl4. At day 9, blood was collected and the kidneys were harvested for histopathology analysis. Blood collected were measured for albumin, total protein, urea and creatinine levels. After treatment, albumin and total protein showed no increased levels; urea decreased at doses of 200, 300, and 400 mg/kg BW, respectively, and creatinine levels only decreased at the dose of 400 mg/kg BW (p<0.05). The dose of 200 and 300 mg/kg BW showed protecting effects in the tubular cells of renal. Therefore, the ethanol extract of E. elatior showed a nephroprotective effect by normalizing the urea and creatinine levels of rats and protecting tubular cells of renal.

Abstract

The use of this mask has an important role in anticipating Covid-19 virus. If the community has knowledge about the use of masks, the community will also be obedient in the use of masks. Compliance is to describe the positive behavior of the people shown by using. Through this Thematic Community Service Program, the community can know the benefits of using masks in preventing Covid-19. the method used in the integrated community service activities of Thematic KKN is the method of socialization, the method for the environment of student residence, the method of media networking and reporting methods.Thematic KKN takes place in the city of Kendari, Sulawesi Tenggara
