Abstract

Gene encoding GRA1 protein is potent DNA-vaccine candidate against toxoplasmosis. The aim of the research was to clone the gene encoding GRA1 protein of tachyzoite Toxoplasma gondii local isolate by DNA recombinant technology. Tachyzoite was grown in Balb/c mice in vivo. Messenger RNA was isolated from total RNA and it was used to synthesis cDNA. Complementary DNA encoding GRA1 protein of tachyzoite Toxoplasma gondii local isolate was amplified and cloned in a prokaryote cloning vector. The recombinant GRA1-encoding gene was then digesting using EcoRI restriction endonuclease and sequencing. The result showed that the recombinant GRA1- encoding gene consisted of DNA sequences encoding all signal peptide and mature peptide of GRA1 protein. Alignment of recombinant GRA1 sequence to gene encoding GRA1 protein of Toxoplasma gondii RH isolate showed 100% homologous.

Abstract

Recently,there are many hepatotoxicity case reports induced by energy drinks and its niacin content is assumed to be the cause of liver injury. Caffeine in energy drinks actually has a hepatoprotective effect. However, this hepatoprotective effect of caffeine is less potent in counteracting the hepatotoxic effect of niacin. This study aimed at determining niacin and caffeine influences in inducing the disruption of liver physiology and the dose of niacin and caffeine in energy drink model which can induce the disruption of liver physiology by measuring AST, ALT, and GGT. As many as 28 male albino wistar rats were divided into seven groups;1 control group and 6 experimental groups, for 49 days. At day 50th, the rats were terminated to measure the AST, ALT, and GGT. The AST and ALT enzyme levels were not significantly correlated (p> 0,05). Otherwise, the GGT enzyme level was significantly correlated (p<0,05). It was showed that niacin and caffeine elevate GGT enzyme at doses 0,491 mg/ 200gr body weight/ day niacin and 1,2275 mg/ 200gr body weight/ day caffeine. These doses are equal to the consumption of three packs of energy drinks per day in human. Keywords: niacin, caffeine, energy drinks, AST, ALT, GGT

Abstract

The niacin in energy drinks has metabolic product that cause oxidative stress and liver damage, while the liver damage can be prevented by hepatoprotective agents. Scoparone in Artemisia vulgaris L. can act as a hepatoprotector by its antioxidant effect. This study aimed to investigate the effectivity of Artemisia vulgaris L. extract as a hepatoprotector in wistar hepatocytes induced by niacin. This study used 25 male rats which were divided into 5 groups: normal, the negative control, the positive control, the treatment group I, and II. Treatment was conducted for 28 days. The samples were terminated and the hepatocyte were prepared for histological examination. Histological appearance was catagorized as mild, moderate, and severe damage with or without inflamatory cells activity. The data analysis by Kruskal Wallis showed significant difference (p<0,001). Further analysis by Mann Whitney revealed significantly difference (p<0,05) between normal group and all groups, negative control group and positive control group, and positive control group and treatment group I, but not significantly difference between negative control group and treatment group I, negative control group and treatment group II, positive control group and treatment group II, and between treatment groups. The study concluded that the effectivity of Artemisia vulgaris L. extract has not been proven as a hepatoprotector but further study is needed to draw a definite conclusion. Abstract

Malaria is an infectious disease caused by Plasmodium spp. Death in malaria is resulting from the parasite’s resistance to malaria drugs and immune system problem. This study was conducted to determine the immunostimulant activity of bangle extract (Zingiber cassumunar Roxb.) in mice infected with Plasmodium berghei. Mice were divided into 5 groups; negative control group [K(-)], positive control group [K(+)] and 3 treatment groups (P1, P2, P3). All groups except K(-) were infected with P. berghei, P1 was treated with Bangle ethanolic extract 0,904 mg/gBW, P2 was given with Bangle ethanolic extract 0,904 mg/gBW and artemisinin 0,0364 mg/gBW, P3 was treated with artemisinin 0,0364 mg/gBW. After 4 days treatment, blood TNF-α from intracardial was measured. The result showed that TNF-α of P1, P2, P3, K(+) and K(-) were 77,46 ± 33,96 ρg/ml, 55,08 ± 15,19 ρg/ml, 85,56 ± 22,43 ρg/ml, 102,81 ± 61,76 ρg/ml, 41,29 ± 2,28 ρg/ml, respectively. The Mann-Whitney test showed p value ≥ 0,05. In conclusion, Zingiber cassumunar Roxb. extract had no effect on TNF-α of P. berghei-infected mice. Abstract

Fracture is a serious musculoskeletal trauma. The healing process of fracture is disturbed by the oxidative stress, because of imbalance quantity of Reactive Oxygen Species (ROS) and an antioxidant. An antioxidant such as polyphenol, that can be found in cocoa, is needed to suppress oxidative stress. The study aimed to investigate the effect of cocoa ethanolic extract on decreasing oxidative stress by measuring malonaldialdehyde (MDA) concentration in fracture healing process. Trial test were conducted in eleven rats to determine appropriate plaster technique. The study used thirty rats and divided into five groups; the normal group without fracture, the negative control group and three treatment groups with manually fractured on left tibia under anaesthesia and immobilized by leucodur plaster. Each treatment group was treated with cocoa ethanolic extract in a dose of 125 mg/kgBW, 250 mg/kgBW, or 500 mg/kgBW orally, respectively for 21 days. The result showed that there was a significant effect of cocoa ethanolic extract on serum and bone MDA concentration in which MDA concentration decreased after cocoa ethanolic extract treatment. The study concluded that cocoa ethanolic extract had a positive effect on supression of oxidative stress in fracture healing process.   Keywords: Cocoa ethanolic extract, polyphenol, fracture healing process, MDA, ROS

Abstract

The data from the Health Research Association of Indonesian Ministry of Health in 2010 showed approximately 35.6% infants having PEM. The diet therapy for severe PEM is modisco. However, due to protein deficiency such as a protease enzyme resulting in nutrients malabsorption, modisco has not been able to compensate the lack of protein in PEM. It requires an active substance to increase protein absorption by adding a proteolytic enzyme. One of the source of proteolytic enzyme is pineapple. The study aimed to prove that addition of pineapple extract on modisco can increase levels of albumin and hemoglobin in severe PEM rats. This was a true experimental study with pre-post test control group design. The sample were 25 male Wistar rats aged 2 months. The data analysis using the Pairwise T test, one way ANOVA followed by LSD showed a significantly difference between pre and post test for both albumin and haemoglobin. The pineapple extract at a dose of 9 and 11 mg/day increased albumin level. And the dose of 7, 9 and 11 mg/day pineapple extract increase haemoglobin level. In conclusion, a modisco modification using pineapple extract for nutritive therapy increased both albumin and hemoglobin levels on severe MEP rats.   Keywords: Severe PEM, Modisco, Pineapple extract

Abstract

Paederus dermatitis caused by rove beetle makes the tissue damage called dermonecrosis. The MMP on pederin toxins from the symbiosis of rove beetle (Paederus sp.) and Pseudomonas aeruginosa is supposed can delay the wound healing process. Doxycycline gel as a MMP inhibitor is believed can accelerate the healing process. The purpose of this study was to determine the effect of doxycycline gel on the healing process of Paederus dermatitis by observing the number of fibroblasts as an indicator. This study used 40 mice, were divided into four major groups, those were normal control group, the negative control, treatment groups with doxycycline gel 1% and 5%. Termination was performed on days 1, 3, 5, 7, and 15. Data was obtained by calculating the number of fibroblasts per field of view, then processed in ImageJ and tested statistically with comparation test of One Way Anova. The average number of fibroblasts per field of view in treatment groups was higher. The peak occurred on day 7 and decrease on day 15. It indicated that the topical administration of doxycycline gel on Paederus dermatitis can accelerate healing process, showed by the increasing in average number of fibroblasts per field of view. Abstract

Poor environmental sanitation is suspected to be one of the causes of soil transmitted helminthes (STH) infection. Environmental sanitation consists of water sources, latrine, sewerage, garbage disposal facilities, and floor type. The objective of this study was to investigate the relationship between environmental sanitation and the incidence of STH infection in primary school children. This was an observational analytic study with cross-sectional design. The subjects were 80 students of two elementary schools in Jember. Stool samples were collected and examined by Kato-Katz method. Data of environmental sanitation was collected by questionnaire and direct observation. Kato-Katz examination showed that 9 samples (11,3%) infected by Ascaris lumbricoides. The majority respondents had not fulfilled requirements of good environmental sanitation. The result of Spearman test showed that there was a significant correlation between STH infection and latrine (p=0,02) but there were no significant relationship between STH infection and environmental sanitation (p = 0.165), gender (p = 0,669), age (p = 0,92), water source (p = 0.084), sewerage (p = 0.146), garbage disposal facilities (p = 0,728), and floor type (p = 0,065).

Abstract

Isolasi rimpang Bangle menunjukkan bahwa terdapat kandungan bioaktif yaitu kurkumin dalam rimpang Bangle. Penelitian ini bertujuan untuk mengetahui peran ekstrak Bangle terhadap produksi Nitric Oxide dan Malondialdehyde pada mencit yang diinfeksi Plasmodium berghei. Jenis dari penelitian ini adalah quasy experimental, dengan post test only control group design. Sebanyak 30 ekor mencit dibagi ke dalam 5 kelompok, masing-masing terdiri dari 6 mencit. Pemberian ekstak Bangle diberikan selama 14 hari dengan tujuan sebagai imunostimulan. Kontrol negatif diberikan aquadest, kontrol positif diinfeksi dengan Plasmodium berghei. Kelompok perlakuan1(P1) distimulasi Bangle. Kelompok perlakuan2(P2) distimulasi Bangle diterapi Artemisinin sedangkan kelompok perlakuan3(P3) diterapi Artemisinin. Terapi diberikan selama 4 hari apabila Plasmodium telah ditemukan dalam darah. Data hasil penelitian ini menunjukkan bahwa terdapat perbedaan yang bermakna antara produksi NO serum P1 dan P2 dengan kontrol positif. Sedangkan produksi MDA serum tidak menunjukkan adanya perbedaan yang bermakna antarkelompok, sehingga dapat disimpulkan bahwa pemberian ekstrak Bangle mampu meningkatkan produksi NO serum namun tidak berperan terhadap produksi MDA serum.   Kata kunci: Bangle, imunostimulan, Nitric Oxide, Malondialdehyde, Plasmodium berghei

Abstract

Duffy-binding like (DBL) domains of Plasmodium falciparum are believed to be involved in erythrocytes invasion and infected erythrocytes cytoadhesion during the blood stage of malaria infection. In Plasmodium falciparum, DBL domains found in the two different protein families; Erythrocyte Binding Ligand (EBL) including EBA-175, EBA-140, EBA-181 and EBL-1, and Plasmodium falciparum Erythrocyte Membrane Protein-1. The study aimed at investigating the specific sequence of Plasmodium falciparum involved in severe malaria outcome.Blood samples from severe and uncomplicated falciparum malaria cases from Papua and South Kalimantan province, Indonesia, were collected for DNA extraction. A dried blood on filter paper were used for RNA extraction. PCR was performed using UNIEBP primers and directly sequenced. Internal var D primers were designed according to the sequencing of the ~550 bp band produced by UNIEBP primers. The nucleotide sequences were analyzed by NCBI BLAST. Multiple bands ranging from nearly 250 bp to 1 kb were resulted from gDNA in all samples. Two isolates yielded bands of 450 and 525 bp, three isolates showed three bands additionally 250 bp, one isolate presented four bands additionally 800 bp and one isolate resulted one band additionally 1 kb.Amplification of cDNA from severe malaria cases produced one to four bands ranging from 250 bp to 700 bp, and no band observed from cDNA of uncomplicated malaria. Sequencing of the 418 bp bands matched with the eba-175 gene, the 316 bp determined as DBL1a domain and 486 bp band matched with the DBLg domain isolated from placenta of PAM’s Malawian woman. The expression of a 237 bp sequence corresponding to var D gene, was detected solely in severe malaria patients, implicating an association of gene expression and manifestation of severe malaria. Further characterization of the var D gene with a larger sample size is required to draw a definite conclusion. Keywords: DBL domains, Plasmodium falciparum, severe malaria, var D gene.
