Abstract

The aims of this study were to determine the prevalence and risk factors of porcine cysticercosis. Thesurvey was carried out in eight districts of Jaya Wijaya, Papua and Jibama market, during October 2009to June 2011. A total of 111 pigs were tested serologically. Serum samples were tested for the presence ofcirculating parasite antigen using monoclonal antibody-based sandwich enzyme-linked immunosorbentassay (MoAb-ELISA). Forty five samples (40.54%) were found positive by MoAb-ELISA and the highestprevalence occurred from the District of Asolokobal (92.86%), followed by Musatfak (75%), Kurulu (65.22%),Bolakme (33.33%), Asologaima (31.82%), Hom-hom (18.18%), Hubikosi (14.29%), Jibama trade (14.29%),and the lowest prevalence from Wamena Kota is 5.88%. Free-range pig husbandry system (OR=4.63;P<0.01) and uncook pork feed (OR=3.65; P<0.05) were important risk factors for porcine cysticercosis. It istherefore necessary to anthropology approach about pig husbandry system and pattern of cook pork feed.

Abstract

Sukabumi District is one of the districts in West Java Province that has not been rabies-free since there are still reported rabies dog bites and rabies-positive cases in humans. The aim of the study was to measure the knowledge, attitude and practice of dog’s owner towards rabies which will then be useful for designing the appropriate rabies control programs. A total of 141 dog owners were selected as samples/ respondents. Respondents were taken from two sub-districts in Sukabumi District, which were 66 people from Jampang Tengah Sub-district and 75 people from Cisolok Sub-district. The data were taken using structured questionnaire. The result showed that most of respondent’s level of knowledge (51.1%) was in moderate category, meanwhile the respondent’s level of attitude related to rabies control program mostly (69.5%) was in positive category. But the positive attitude was not reflected with their practice towards rabies control program which many of them (63.1%) was in bad category of rabies control practices. From analytical result showed that only one characteristic variablewhich was the information access about rabies that had significant correlation (c2=9.959, p=0.006) towards respondent’s level of knowledge. Furthermore, another result showed that there was a significant relationship between knowledge variable with respondent’s attitude level (p= 0.000, r= 0.275).

Abstract

Enzyme-linked immunosorbent assay (ELISA) is a serological test method most widely used for thediagnosis of paratuberculosis, because it has a better sensitivity compared to other serological test.Protoplasmic antigen (PPA) or cellular extract is still the main choice for the diagnosis of paratuberkulosisdevelopment. The aim of research was to use the PPA Mycobacterium avium subspeciesparatuberculosis(MAP) field isolates for the development of paratuberculosis ELISA (ELISA PPA-L). As many as 322cattle sera (300 negative and 22 positive) were tested using this method and compared with IDEXXcommercial kit. The sensitivity and specificity of ELISA PPA-L test results were 68.18% and 97.0%,whereas for the IDEXX kit were 63.64% and 97.33%respectively. ELISA PPA-L had higher sensitivity andlower specificity compared to the IDEXX commercial kit. ELISA test using protoplasmic antigen of MAPfield isolates has good ability for paratuberculosis serological test and can be used for screening test of thedisease in Indonesia.

Abstract

Importation of feeder cattle from farms constantly using antibiotics in feed could result in the occurrenceof antibiotic resistant bacteria. A study was therefore conducted to investigate the possible use antibioticsand the antibiotic resistance of commensal Salmonella sppagainst several antibiotics. Salmonella spp.bacteria (n=50)were isolated from 100 samples of feeder cattlesfaeces. Total of 50 Salmonella spp. isolateswere subjected for Salmonella spp. examination and the isolated Salmonella spp. was tested for theantibiotic resistance using 10 antibiotics (ampicillin, cephalotin, erythromycin, tetracycline, streptomycin,chloramphenicol, trimethoprim, trimethoprim-sulfamethoxazole, nalidixic acid, and enrofloxacin) usingdisk diffusion method on Muller-Hinton agar following Clinical and Laboratory Standards Institute (CLSI)guidelines for interpretation. The isolated Salmonella showed resistance towards erythromycin 98%,ampicillin 34%, streptomycin 22%, nalidixic acid 8%, cephalotin 6%, tetracycline 4%, and chloramphenicol2%. There was no resistance against enrofloxacin, trimethoprim, and trimethoprim-sulfamethoxazole. Salmonella spp. bacteria derived from imported feeder cattle which have been resistant to antibiotics arepotential for spreading the antibiotic resistant bacteria to other susceptible animals. Such bacteria canalsotransfer the antibiotic resistant gene to other bacteria in Indonesia which ould be a potential threatfor public and animal healths.

Abstract

Keamanan bahan pangan merupakan masalah yang amat penting bagi konsumen dan industri pangan. Cemaran bakteri Escherichia coli dan Coliform dianggap sebagai indikator sanitasi dalam proses pengolahan bahan pangan. Pelacakan bakteri patogen dalam pangan juga telah dilakukan secara rutin, termasuk yang bersifat zoonosis seperti Escherichia coli O157:H7. Bakteri ini menghasilkan toksin yang dikenal dengan Shiga toxin. Toksin ini dapat menimbulkan diare berdarah, colitis haemorrhagi dan hemolytic uremic syndrome (HUS) pada manusia. Dalam penelitian ini dipelajari hubungan antara tingkat cemaran dan insidensi Coliform, E.coli, E.coli O157 dan E.coli O157:H7 pada daging sapi. Bakteri pertama ditumbuhkan pada media EMBA, selanjutnya dipupuk pada media SMAC dan diakhiri dengan uji aglutinasi lateks untuk memastikan keberadaan bakteri E.coli O157 dan uji antiserum H7 untuk memastikan isolat yang diisolasi merupakan isolat E.coli O157:H7. Hasil isolasi dan identifikasi terhadap 89 sampel daging sapi diperoleh hasil rata-rata tingkat cemran coliform dan E.coli sebesar 93,01+ 2,64x103 cfu/g dan

Abstract

Shiga toxin produced by Escherichia coli O157:H7 can cause outbreaks and sporadic cases of serioushuman diseases. The diseases are indicated by hemorrhagic colitis and hemolytic uremic syndrome. Meatand meat products have been identified as vehicles of food borne disease caused by E.coli O157:H7. Themain aim of this research was to identify the correlation between the level of E.coli O157:H7 contaminationand the presence of Shiga toxin (Stx1 and Stx2) by applying method of Vero toxin Escherichia coli-ReversePassive Agglutination Test (VTEC-RPLA). The results showed that 3 of 7 isolates and 1 of 4 isolatesisolated from feces of cattle and beef, respectively produced Stx 1 (VT1). In the detection of Stx 2 (VT2), 4of 7 isolates and 1 of 4 isolates, isolated from the same samples were found to produce this toxin.According to all isolates, in this research showed, 1 isolate was found to produce VT2, 4 isolates to produceboth VT1 and VT2, while 6 isolates showed negative results either to VT1 or VT2.

Abstract

Trichinellosisis is a zoonotic disease caused by infection of Trichinella spp. and is found world-wide.The seroprevalence of trichinellosis in pig population has never been reported in East Nusa TenggaraProvince particularly Kupang City. The aim of this study was to determine the seroprevalence oftrichinellosis in pig at Kupang City, East Nusa Tenggara. A cross-sectional study was conducted byexamining 376 blood samples of pigs collected from slaughterhouses in Oeba, Kupang using simplerandom sampling method. Blood samples were tested by indirect enzyme linked immunosorbent assay(ELISA) using Screen®Trichinella indirect multi-species ELISA kit. Test results showed that 3 (0.8%)serum samples were positive of trichinella antibody. In conclusion ,some pigs in the city of Kupang hasbeen infected by Trichinella sp. and could be a threat for human health.

Abstract

The emergence of antibiotic-resistant bacteria is a common problem nowadays. This problem has led to more difficult treatment of infectious diseases and the higher cost of treatment for the diseases. Therefore, it is necessary to develop a new type of antimicrobial that has ability to control the pathogenic bacteria without causative resistence. The aim of this research was to measure the actinomycetes ability which produced antimicrobial againts antibiotic-resistant E. coli taken from poultry meat in Tanjung Perak Port Surabaya. The actinomycetes isolates used in this study were Streptomyces genus, Streptomyces 3 (Sp 3), Streptomyces 44 (Sp 44), Streptomyces 46 (Sp 46), Streptomyces 47 (Sp 47) and Streptomyces 413 (413). Two isolates of actinomycetes, Sp 47 and Sp 413, were found to have an inhibitory effect on the growing of antibiotic-resistant E. coli. Based on the average affect of inhibition ability, actinomycetes InaCC was the strongest while InaCC 47 was weak.

Abstract

Salmonella spp. has been recognized as the major cause of food-borne illness in humans worldwidecausing remain relevant to public health. Poultry vaccination is one promising strategy to mitigateSalmonella infection in poultry and, in turn, in humans as well. The objective of this study was to assessthe potential of cross-reaction of serum in Salmonella enteritidis-vaccinated chicken to some serotype ofSalmonella enterica. Four female, Isa Brown layer chickens (20 weeks old), were vaccinated with S. enteritidisstrain Sm24/Rif12/Ssq (intra vena) to induced the production of specific antibodies in serum. Crossreactionof serum in S. enteritidis-vaccinated chicken were assess with agar gel immunodiffusion test(AGID) with S. enteritidis, S. pullorum, S. typhimurium, S. typhi, and Escherichia coli antigens. Serumcould react with S. enteritidis and all types of S enterica used in this study (S. pullorum, S. typhimurium,S. typhi), but could not react with E. coli. The potential of cross-reaction of serum in S. enteritidis-vaccinatedchicken to some serotypes of S. enterica may play a role in reducing the infection caused by that serotype.

Abstract

Mycobacterium avium subspesies paratuberculosis (MAP) is a thermal tolerant bacteria. The presenceof these bacteria in pasteurized dairy milk is associated with infectious bowel disease in human known asCrohn’s disease. The aim of this study was to detect MAP in pasteurized dairy milk sold in Bogor. Fourtytwo samples of plain flavoured milk (180–250 ml) from 7 producers were bought from supermarkets inBogor. The presence of MAP was detected by isolation and conventional polymerase chain reaction (PCR)using IS 900 and F57. Bacterial isolation were done by Herrold’s egg yolk medium with mycobactine J(HEYMj) and without mycobactin J (HEYM) and incubated at 37°C for 20 weeks. The DNA extraction ofall pasteurized dairy milk samples were conducted by DNeasy® Tissue Kit. Amplification conditionsfor PCR were: 1 cycle at 94°C for 10 minutes, 40 cycles at 94°C for 1 minute, 58°C for 1 minute, and72°C for 3 minutes, and 1 cycle at 72°C for 7 minutes. After 20 weeks of incubation, there were no sign ofMAP which grew in all isolation mediums. The PCR IS 900 and F57 did not detect the DNA band of thetarget. In the conclusion, there was no MAP detected in pasteurized dairy milk sold in Bogor.
