Abstract

Damage of spermatozoa membrane in sexing procces with albumin gradient can decrease the quality of spermatozoa, thus to solve that need diluents for protect spermatozoa membrane to make good quality. The aim of this research is to know the best diluents between Andromed and CEP2 (cauda epididymal plasma) + 0% of yolk to keep the quality of Limousin Cow’s Spermatozoa from sexing procces with albumin’s gradient. Parameters measured for the quality of spermatozoa were : spermatozoa motility, spermatozoa viability, spermatozoa abnormality, spermatozoa concentration, and spermatozoa motility, membrane integrity, spermatozoa capacity, and acrosom reaction. Observation of membrane integrity using HOS (Hypoosmotic Swelling Test), observation of capacity and acrosom reaction using fluoresen CTC (Chortetracycline) dye. Result show that Andomed diluents and CEP2 + 10% yolk in top layer and bottom (X) and (Y) layer can keep the quality of Limousin Cow’s spermatozoa in sexing procces include spermatozoa motility (X 62,5%, Y 58%,) (X 56,5%, Y 55%), viability (X 91,64%, Y 91,51%) (X 89,87%, Y 93,51%, spermatozoa’s concentration (X 643 million /ml, Y518 million /ml) (X648 million/ml, Y517 million/ml), total spermatozoa are motil (X373,60 juta/ml, Y187,80 million/ml) (X 296,03 miliion/ml, Y 187,38 million/ml) , and have low abnormality (X 4,11%, Y 4,38%), (X 6,42%, Y 4,04%). Andromed diluents and CEP2 + 10% yolk can keep the integrity of spermatozoa membrane (X 87,60%, Y 80,26%) (X 79,61%, Y 81,03%), remains well and have the spermatozoa are not yet capacitation(X 87,60%, Y83,71%) (X 79,61%, Y 81,03%) remains high, spermatozoa capacity (X 9,76%, Y 9,92%) (X 13,45%, Y 8,95%), dan acrosom reaction (X 4,11%, Y 4,38%), (X 6,42%, Y 4,04%) remains low. Andromed diluents was not different (P > 0,05) from CEP2 diluents + 10% yolk in keeping the quality of Limousin Cow Spermatozoa from the sexing process with albumin’s gradient (egg white). Abstract

The factors that influence the success of Artificial Insemination is affected by acceptors that will be used for AI which have a good Body Condition Score, because BCS has a relationship with animal reproduction, such as fertility and pregnancy. The purpose of this research was to evaluate and determine the relationship of Body Condition Score (BCS) on Service per Conception (S/C) and Conception Rate (CR). The material used was 80 Brahman Cross (BX) cows, the sample was selected by Purposive Sampling with the criteria 2-3 years old with body weight ranged from 300-500 kg, gave birth, and BCS between 4-7 (scale 1-9). The method used in this study was the survey method. The variables observed were the relationship between Body Condition Score (BCS), Service per conception (S/C), and Conception Rate (CR). The data was obtained by using correlation analysis and also a simple regression with the SPSS 2.0 software program, then analyzed descriptively. The results showed that the equation of relationship between BCS and S/C variables obtained r = 0.961% and the regression value equation y = -25.998 + 1.520x (R2) 23.9%. The relationship between BCS and CR variables obtained a value of r = 0.976% and BCS regression equation value with CR y = -39.727 + 1.109x (R2) 29.5%. The research thus concludes that there is a positive relationship that is being between BCS on the S/C and CR of Brahman Cross cows.

Abstract

This research aims to study influence of the breed chicken toward motility, abnormality, and durability of live spermatozoa after storage at a temperature 0f 40C. Cement of chicken that using in this research come from 3 chickens male with the different there are chicken laughing , chicken arab , and chicken burma. The design of this research is laboratories experimentaly by using random design complete steel and torrie ( 1991 ) , consisting of 3 treatment and 6 remedial ( the number of cement). Abstract

ABSTRAK Edible film dari protein whey memiliki sifat baik sebagai bahan pengemas karena memiliki keunggulan seperti transparan, lunak, tidak berbau serta mampu menahan aroma produk pangan yang dikemas. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan konsentrasi karagenan dan jenis plasticizer sorbitol 35% terhadap mikrostruktur edible film berbahan dasar whey dangke. Metode penelitian yang digunakan adalah eksperimental desain dan variabel yang diamati adalah pengamatan mikrostruktur. Perlakuan menggunakan konsentrasi karagenan yang berbeda, terdiri dari konsentrasi karagenan A1=2%; A2=2,5%; A3=3% dan A4=3,5%. Hasil penelitian yang diperoleh dianalisis menggunakan metode deskriptif dengan memberikan gambaran secermat mungkin. Hasil penelitian menunjukkan bahwa adanya polimer yang semakin meningkat maka akan berperan dalam meningkatkan viskositas suspensi film selama terjadinya proses gelatinisasi, sehingga edible film yang dihasilkan memiliki mikrostruktur yang semakin baik dengan peningkatan karagenan. Disimpulkan bahwa mikrostruktur edible film yang lebih homogen diperoleh dengan menggunakan rasio karagenan 3% dimana distribusi karagenan tercampur ke dalam matriks protein whey secara merata.Kata kunci: edible film, karagenan, sorbitol, whey dangkeMicrostructure of Whey Dangke's Edible Film with Karageenan and 35% Plasticizer Sorbitol AdditionABSTRACTEdible film from whey protein has good properties as a packaging because it has advantages including being transparent, soft, odorless, and able to withstand the aroma of the food product it is coated with. This study aims to determine the effect of the addition of carrageenan concentration and the type of sorbitol plasticizer 35% on the edible film made from whey dangke microstructure.  The research was conducted as a laboratory experiment and the observed variable was microstructure observation. The treatments used different carrageenan concentrations, consisting of carrageenan concentration A1 = 2%; A2 = 2.5%; A3 = 3% and A4 = 3.5%. The results obtained were analyzed using descriptive methods by providing a picture as carefully as possible. The results showed that the presence of an increasing polymer would play a role in increasing the viscosity of the film suspension during the gelatinization process so that the resulting edible film had a better microstructure with the increase in carrageenan. It was concluded that a more homogeneous edible film microstructure was obtained using a carrageenan ratio of 3% where the distribution of carrageenan was evenly mixed into the whey protein matrix.Keywords: edible film, carrageenan, sorbitol, whey dangke

Abstract

The purpose of this research was to determine the effect of chicken species on the quality and fertility of spermatozoa. Semen is collected from Kampung, Arab, and Bangkok chickens through a massage method. The semen obtained is collected in a test tube, then diluted with Ringer's Dextrose. Spermatozoa examination is carried out macroscopically and microscopically. There are eighteen (18) female leghorn chickens used to test the fertility of spermatozoa, through the process of hatching eggs in an incubator. The research design used a Completely Randomized Design (CRD), with three (3) treatments and six (6) replications. Data analysis using analysis of variance, if there are differences, continued with the Least Significant Difference test (LSD). The results showed that the fertility of Bangkok chicken spermatozoa was significant (P 0.01) compared to the sperm fertility of Arab chickens and Kampung chickens. The average fertility value of each cement is Kampung chicken: 68.88%, Arabic chicken: 71.32%, Bangkok chicken: 89.96%. Abstract

Antimicrobial edible films made from clove essential oil play a role in reducing, inhibiting, or slowing the growth of bacteria that may be existed in food packaging or packaging materials to extend the shelf life of packaged foods. This study aims to produce whey and clove oil films with film thickness, elongation, tensile strength, water vapor transmission rate, optical properties, antimicrobial properties of clove oil, and its inhibition against pathogenic microbes. The physical and antimicrobial characteristics of edible film were investigated using an experimental method in a completely randomized design with four treatments. The treatment used different concentrations of clove oil (C0: 0% clove essential oil, C1: 5% clove essential oil, C2: 10% clove essential oil, and C3: 15% clove essential oil). The percentage value of elongation and microbial inhibitory activity were significantly affected (p<0.01) by the concentration of clove essential oil. Based on all variables, 10% clove essential oil concentration is the best treatment with a thickness of 0.035 mm, elongation of 78%, tensile strength of 8.82 N, water vapor transmission rate of 7.80 g.mm-2.day-1, and inhibitions of E. coli and S. aureus resulted in high rates. The results conclude that the addition of clove essential oil has a promising potential to improve the antimicrobial properties of whey edible film composites. The use of clove essential oil at the level of 10% is the best treatment.

Abstract

Impacts of Implementation Regulation of The Ministry of Marine Affairs and Fisheries No. 1/Permen-Kp/2015 to Lobster Traffic Volume in Gorontalo Province. The implementation of the Permen NO. 1/PERMEN-KP/2015 with the aim to protect, preserve, and utilize crustacean resources. This study aimed to analize impact of implementation Permen NO. 1/PERMEN-KP/2015 to lobster traffic volume in Gorontalo Province. The research result that highest of lobster traffic volume on 2012 and lowest on 2016. There was a significant difference (p <0.05) between lobster traffic volume before and after implementation of Permen KP NO. 1/PERMEN-KP/2015. Implementation of Permen KP NO. 1/PERMEN-KP/2015 turned positive impact to lobster because provides a lobster opportunity to regenerate.

Abstract

Penelitian ini bertujuan untuk mengetahui kualitas semen ayam KUB menggunakan pengencer ringer dextrose dan ringer laktat pada suhu 5?. Pengumpulan data variabel motilitas, viabilitas, dan abnormalitas yang diperoleh dalam penelitian ini dicatat kemudian ditabulasi menggunakan program excel kemudian dianalisis secara deskriptif analitik. Variabel penelitian yang diamati adalah motilitas, viabilitas dan abnormalitas. Hasil penelitian menunjukkan bahwa penggunaan ringer laktat mampu mempertahankan kualitas semen ayam KUB dengan lama simpan 6 hari dengan menghasilkan motilitas, viabilitas yang lebih baik serta tingkat abnormalitas yang lebih rendah dibandingkan dengan pengencer ringer dextrose yang hanya mampu mempertahankan kualitas semen dengan lama simpan 4 hari. Berdasarkan hasil ini dapat disimpulkan bahwa Pengencer ringer laktat memiliki kualitas semen yang lebih tinggi dengan lama simpan mencapai 6 hari dengan motilitas individu mecapai 60%, viabilitas 48,76%, dan abnormalitas 6,36% sedangkan pengencer ringer dextrose dapat mempertahankan kualitas semen dengan lama simpan 4 hari dengan motilitas individu 53%, viabilitas 30,1% dan abnormalitas 7,13%.Kata kunci: Ayam KUB, Ringer Dextrose, Ringer Laktat, Semen.

Abstract

The research aims to observe the effect of konjac glucomannan and different types of plasticizers on the microstructure of whey film. The microstructure of whey film was investigated using an experimental method. The research was conducted in descriptive methods with two treatments.The treatment used konjac glucomannan concentration (P4= 2.5 %; P7= 3 %) and different types of plasticizers (C1= glycerol 30 %; polyethylene glycol 30 %). The results showed that the combination of whey and konjac glucomannan had an effect on the microstructure of the film. The addition of konjac glucomannan concentration, 30 % glycerol, and 30 % PEG plasticizers gave microstructural characteristics with good whey film surface. The microstructure of this film was determined during the heating, drying process and the contribution of the gel from konjac glucomannan to the whey film structure. A more homogeneous edible film microstructure was obtained using a 2.5 % concentration of KGM with the use of 30 % glycerol plasticizer where the distribution of konjac glucomannan was mixed into the whey protein matrix evenly (homogeneous).
