Abstract

Irama sirkadian dipengaruhi oleh cahaya lampu dapat menghambat pembentukan hormon melatonin. Baik dan buruknya tidur dilihat dari kualitas tidur. Kualitas tidur buruk memengaruhi keseimbangan fisiologis dan psikologis. Khususnya pada mahasiswa menyebabkan penurunan konsentrasi dan disfungsi aktivitas di siang hari. Penelitian dilakukan pada bulan Juli−November 2016 di Fakultas Kedokteran Universitas Padjadjaran, Jatinangor dan bertujuan untuk mengetahui perbandingan kualitas tidur mahasiswa Fakultas Kedokteran Universitas Padjadjaran yang menggunakan dan tidak menggunakan cahaya lampu saat tidur.  Penelitian dengan pendekatan potong lintang ini dilakukan pada populasi yaitu mahasiswa Fakultas Kedokteran Universitas Padjadjaran. Besar sampel berdasarkan rumus analitik komparatif kategorik-kategorik tidak berpasangan sejumlah 80 responden berjenis kelamin laki-laki untuk setiap kategorinya. Instrumen penelitian ini adalah kuesioner Pittsburgh Sleep Quality Index (PSQI) dengan analisis statistik yang digunakan adalah uji Chi-Square. Dari 160 subjek penelitian didapatkan sejumlah 80 orang yang menggunakan lampu dan 80 orang tidak menggunakan lampu saat tidur. Perbandingan kualitas tidur antara kelompok subjek yang menggunakan lampu dan tidak menggunakan lampu dengan uji chi-square didapatkan perbedaan yang tidak bermakna dengan mayoritas subjek memiliki kualitas tidur buruk (p=0,191). Hasil menunjukkan bahwa tidak terdapat perbedaan kualitas tidur mahasiswa Fakultas Kedokteran Universitas Padjadjaran yang menggunakan dan tidak menggunakan cahaya lampu saat tidur.Kata Kunci: Cahaya lampu, kualitas tidur, mahasiswa

Abstract

High intensity exercise (HI) induces oxidative stress. Running for 2400 meters is one of the simple exercise form which can be counted as HI. During HI, increase of oxygen consumption happened in muscle cell resulted in an increase of oxidant level. The aim of the study was to measure the effects of Indonesian green tea supplementation, before and after HI to the level of lipid peroxidation (TBARSMalonyldialdehyde (MDA)). The subjects of study were 42 male students from Faculty of Medicine, Universitas Padjadjaran (FMUP) and Faculty of Physical Exercise, Indonesia University of Education (FPOK-UPI). The subjects were divided into trained and untrained subject. Subjects were divided into groups using Astrand Harvard step test. The purpose of this test was measuring the level of VO2 max. Blood samples for MDA level were collected at 3 hours before and after high intensity aerobic  exercise. Oxidative stress was induced by 2400 meters run. The result showed that green tea supplement-ation was effective to decrease MDA plasma level especially after exercise. Green tea could serve as natural antioxidant resources, which potentially can be used as potential sport supplemental drink.Key words: Aerobic exercise, Malonyldialdehyde (MDA), pre and post exerciseStudi Pra dan Pasca Pemberian Suplemen Ekstrak Teh Hijauterhadap Kadar MDAHigh intensity exercise (HI) dapat menginduksi stres oksidatif. Berlari sejauh 2400 meter merupakan salah satu bentuk latihan sederhana yang dapat digolongkan sebagai HI. Selama HI, konsumsi oksigen pada sel otot meningkat yang dapat menyebabkan peningkatan kadar oksidan. Penelitian ini bertujuanuntuk mengukur efek dari suplemen teh hijau Indonesia sebelum dan setelah HI terhadap kadar lipid peroksida (TBARS-Malonyldialdehyde (MDA)). Subjek penelitian ini adalah 42 mahasiswa laki-laki dari Fakultas Kedokteran Universitas Padjadjaran dan Fakultas Pendidikan Olahraga dan Kesehatan,Universitas Pendidikan Indonesia. Subjek penelitian dibagi ke dalam kelompok terlatih dan tidak terlatih dengan menggunakan cara pengujian Astrand Harvard. Tes ini bertujuan untuk mengukur kadar VO2 max. Sampel darah untuk kadar MDA diambil 3 jam sebelum dan setelah high intensity aerobic exercise. Stres oksidatif diinduksi dengan berlari sejauh 2400 meter. Hasil penelitian menunjukkan bahwa suplemen teh hijau efektif dalam menurunkan kadar MDA dalam plasma khususnya setelah berlari. Teh hijau dapat menjadi sumber antioksidan alami yang berpotensi sebagai minuman suplemen olahraga.Kata kunci: Aerobic exercise, Malonyldialdehyde (MDA), sebelum dan setelah berlari

Abstract

Spinocerebellar ataxia (SCA) 3 is a neurodegenerative disease which involves cerebellum and extra cerebellum. Neuropathy in SCA3 manifests in various ways, including axonal and demyelination lesions in sensory and motor nerves. There has not been any study that describes the peripheral neuropathy characteristics of SCA3 patients in Indonesia at the time of this publication. This paper reports a case of a 43-year-old male with known spinocerebellar ataxia 3 presented with hereditary ataxia and mild numbness in both palms since two years before. No abnormalities were found during the sensory examination. The NCS showed severe axonal demyelinating sensorimotor peripheral neuropathy. In magnetic resonance imaging (MRI), an atrophy in the cerebellum with cerebral multiple lacunar infarction was identified. Electrophysiological results revealed profound axonal lesion in peripheral nerves. To conclude, peripheral neuropathy in SCA3 represents the dominance of axonal lesions in motor nerves.

Abstract

Breast cancer (BC) and cervical cancer (CC) have a high prevalence and mortality rate worldwide. Despite the availability of advanced treatment, resistance to conventional chemotherapies has emerged. Myrmecodia pendens, one of the species of Sarang Semut (local name), possess a potential of antitumor effects by inducing cell death different cancer cell entities. This study aimed to assess anti-tumor activities of n-hexane fraction of M. pendens in inhibiting cell survival and cell migration in BC and CC cells. M. pendens was extracted in methanol then fractionated using n-hexane or ethyl acetate. BC cells including MCF-7 (luminal A), HCC-1954 (HER2+) cells and CC Hela cells were treated with M. pendens extracts to evaluate cytotoxic activity using 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay as well as anti-cell migration using scratch assay. We also analyzed inhibitory concentration 50 (IC50) of n-hexane fraction in BC and CC cells. We started with comparing cytotoxicity activities of methanol extract, ethyl acetate and n-hexane fractions of M. pendens. Data showed that the n-hexane fraction was the most potent inducing BC cell death. Therefore, we used the n-hexane fraction for further experiments. Interestingly, IC50 of this fraction in HCC-1954 and Hela cells were lower than in MCF-7 cells, 16; 13 and 60 ppm, respectively. Moreover, the low concentrations of n-hexane fraction inhibited HeLa cells migration, compared to control group (p<0.05). The n-hexane fraction of M. pendens shows promising anti-cancer agent, by inhibiting BC and CC cell survival as well as inhibiting CC cells migration.Keywords: breast cancer, cervical cancer, MTT assay, Sarang Semut, scratch assay
