Abstract

Dengue Hemorrhagic Fever (DHF) is a significant health problem in Semarang, Indonesia. A Certain type of insecticides has been widely applied beginning in the last decade as an effort to control the mosquito vectors of dengue virus. This practice could lead to increased resistance in mosquito populations toward these types of insecticides. This research aimed to describe the resistance status of Aedes sp. populations in Semarang. A cross-sectional design study was conducted in 2015, with Aedes sp. populations sampled in several villages of Semarang. Field strains of Aedes sp. eggs were collected using ovitraps and larvae from entomological surveys. Mosquito adults were raised from the collected eggs and larvae under standard conditions and used for biochemical assays. ELISA examined Aedes sp. resistance status. The results showed that there was monooxygenase activity in Aedes sp. populations. In quantitative terms, 77.8% of mosquito samples showed an Optical Density (OD) more than the cut-off point (0.165). Resistance to synthetic pyrethroid insecticide in Aedes sp. mosquitoes population in Semarang may be caused by the mechanism of monooxygenase detoxification enzymes in particular. This case indicated that Aedes sp. has a high capacity as a vector to transmit the dengue virus in Semarang.

Abstract

Siomay is a mixed food that has a limit of colony growth 1 x 104 colony/g or mL. A fish or meat-based food such as siomay is easily contaminated by pathogenic bacteria such as Escherichia coli. The contamination on siomay could be caused by the hygiene practices of food that was not applied properly. This research aimed to analyze the association between hygiene practices and microbiological quality of siomay stall in Tembalang Sub-District. The type of this research was analytic observasional study with cross-sectional approach. The population of this research was all of the siomay stall in Tembalang Sub-District. Sampling procedure used accidental sampling technique and it was obtained 37 samples. Interview and observation were conducted on hygiene practices, raw materials quality and raw materials storage practices at siomay stall. The data were analysed using chi square test. The results revealed that samples of 73% had the MPN values exceeding the limit, 70,3% was contaminated Escherichia coli exceeding requirement, and 32,4% was grown yeast exceeding the standards, then 91,9% samples did not supply the microbiological quality of food. The analysis showed that there was an association between hygiene practice (p = 0,047) in siomay stall with the microbiological quality. However, the raw materials quality and the raw materials storage practices of siomay had been quite good.

Abstract

Asthma is characterized by both chronic inflammation and remodeling of the airways. Eosinophils may contribute to inflammation associated with bronchial asthma by causing tissue damage and subsequently organ dysfunction, but also by the generation of cytokines and lipid mediators. It was reported in previous studies that pharmacological activities were identified in various compounds extracted fromEuphorbia hirta L. such as antiinflammatory, histamine inhibitor, lipoxygenase inhibitor, anti-allergic and cyclooxygenase inhibitor. This study was aimed to determine the effect of E. hirtaextract onperipheral blood eosinophils count in mice model of allergic asthma. MaleBalb/C mice were sensitized and challenged intraperitoneally (i.p) with ovalbumin(OVA). Mice were immunized i.p. on days 1 and 14 with 2.5 mgof OVA adsorbed to 7.75ml of Aluminium hydroxide gel. OVA challenges (10 mgOVA in l ml of PBS) were administered aerosolly on days 21, 23, 25 and 27, andperipheral blood eosinophilscount was determinedon day 28.A one-way analysis of variance (ANOVA) with LSDpost hocanalysis was used to determine significant differences. Results were expressed as mean+SEM, and values of p 0.05wereconsidered statistically significant. The results showed thatperipheral blood eosinophilscountin the OVA group was51.0+10.4cells/mm E. hirtaextract dose 500 mg/BW/day was30.2+6.3cells/mm3,E. hirtaextract dose 1,000 mg/BW/day was 30.4+6.0cells/mm and antihistamine group was32.2+8.6cells/mm respectively, whilecontrol mice showed46.6-8.8 cells/mm3. It was concluded that E. hirta extract could minimizeperipheralblood eosinophilscount.
