Abstract

The richness of biological resources (biodiversity) in mountainous area of Papua is an asset that has to be preserved.Exploitation of natural resources often cause damage on those biological assets and as genetic resources.Care has to be taken to overcome the situation of biological degradation, and alternate steps had been shaped on ex-situ biological conservation. Wamena Biological Gardens, as an ex-situ biological conservation, has been established to keep the high mountain biological and its genetic resources in Papua.

Abstract

Enzymatic activities in soil as due to microbes action in organic matter degradation, lead to propose as indicators for determining soil degree enrichment.In this work, the enzymatic activities of cellulase, invertase and amylase were determined in tropical soil collected from Biological Research Station in Wamena. Result of measurement on five soil samples showed that cellulase activity occurred between 0.10 - 0.31 mg reducing sugar/g soil/hour in 2% Carboxymethylcellulose (CMC) substrate, and about 0.15 - 0.25 mg reducing sugar/g soil/hour in 0.5% Avicel substrate.Amylase and invertase assayed in 6% of amylum and 6% of sucrose substrates correspondingly; and its activities between 2.55 - 3.54 and 0.59 - 1.30 mg reducing sugar/g soil/hour, respectively. Correlation of enzymatic activities and soil organic-C content (1.35 - 2.70%) of soil layer was significant in selulase and amilase, but it was poor correlation in invertase activity.

Abstract

Pseudomonas bacteria plays essential role in soil ecology such as decomposer and biological control. The bacteria were isolated on selective media and identified from five soil samples taken within area of Wamena Biological Gardens.There are six species Pseudomonas were indentified based on morphological characters and biochemical reaction.P. striata was found to be common in soil of the area.No pathogen Pseudomonas was indentified in all soil samples. Preliminary study on biological control for fungal pathogen Fusarium indicated that P. fluorescens, P. striata and P. cepacia have ability to inhibit Fusarium growth in vitro.

Abstract

Research on the effect of weight and composition of media, temperature and chemically stimulation on fruiting body formation of shiitake (Lentinus edodes) had been done in Wamena, Jayawijaya district, Irian Jay a. The research consisted of three experiments. The first experiment consisted of 3 kinds of media such as media A, B and C(A- sawdust and 100% standard nutrition; B = sawdust and 50% standard nutrition; and C = sawdust without nutrition) and three kinds of temperatures were 20°C (room temperature),4°C (refrigerator temperature) and -10°C(freezer temperature).The second experiment used 4 kinds of media. They were media D, E, F and G which were stimulated by SnCh, CoCh, salicilic acid and the extract of bark of "Seno" (Castanopsis acuminatissima).The third experiment used medium B, weight 500 g and 1000 g (medium H and I).The result showed that medium H& I produced the highest fruiting body. However medium I produced the higher fruiting body compare to medium H).The added of the extract of the bark of C. acuminatissima.SnCh, CoCh and salicilic acid stimulated the fruiting body formation.
