Abstract

Background: Contact bioautography method has been developed for determination of chloramphenicol concentration. Validation of bioautography method has beed carried out by using parameters including linearity, accuracy, precision, and detection limit.Methods: Thin Layer Chromatography of chloramphenicol has been performed by using Silica gel 60 F254 as a stationary phase, chloroform : methanol (80:20, v/v) and UV lamp as a solvent and for spot visualization respectively. Before spotting, analyte of the chloramphenicol was dissolved in aceton as solvent. Bioautography has been performed by using Escherichia coli ATCC 25922 as a bacterial and Nutrient Agar as medium test.Results: It was found that one spot visualized on the chromatogram has Rf value 0.5. The result showed that respon of activity to be linear at the amount of chloramphenicol between 100 ppm-200 ppm, with regression quotion: Y = 2.8X - 4.3, r value =0.9 and Vxo = 1.8%. Accuracy and precision of the method are 2.8% + 2.3 and 96.2% + 4.7 respectively.Conclusion: Detection Limit (DL) value is 0.06 μg could be expressed as Minimum Inhibition Concentration (MIC). Abstract

Fluorescence materials such as carbon dots are unique and non-toxic materials that can be produced using many methods. In this research, synthesis and optical characterization of carbon dots from the peels of dragon fruit and pear have been done. The synthesis uses bottom-up method using microwave with 20 minutes, 40 minutes, and 80 minutes of heating time. The result of optical characterizations showed that the samples that have been heat treated are carbon dots based of the absorbance in the range of 280-350 nm, and the intensity are decreased in the range of visible and infrared wavelength. There are emission and intensity changes by different heat treatments. From the samples of dragon fruits peels, the emission wavelength tended to shift toward visible light at the wavelength of 551.51 nm, 507.98 nm and 487.28 nm with the excitation at the wavelength of 420 nm from pulsed pico-laser. While, the emission wavelength from the samples of pears peels were fluctuate in wavelength of 529.09 nm, 507.52 nm and 519.46 nm. Similarly, the time-resolved photoluminescence characterization test showed that there were time changes in each sample of carbon dot. The decay lifetime of carbon dots was fluctuated between 4.5 ns - 5.4 ns for dragon fruit’s peels and about 1.9 ns - 2.4 ns for pears peels.

Abstract

The study was designed to investigate the optimation of preparation of transdermal patch sodium diclofenac matrix type. In this study, matrix-type base containing diclofenac sodium were prepared using polymeric combination of EC N-20  and PVP K-30 in different ratios ( 9:1 (Formula I); 8:2 (Formula II); 7:3 (Formula III)) and 6:4 (Formula IV). The moisture content was calculated as a difference between initial and final weight with respect to final weight. The homogeneity of patch surface was determined using Scanning Electron Microscope (SEM) Released test using dissolution tester were carried out in 500 mL of  phosphate buffer salin pH 7.4 ± 0.05 at temperature 37 ± 0.5°C with speed of swirl 50 rpm. The concentration of sodium diclofenac released was determined using spectrophotometric method at l 276 nm. Data was analyzed by statistic programmed of SPSS using one way analysis of variance with degree of believed 95% (α = 0.05).  It is concluded that the polymeric combination EC N-20 and PVP K-30 in composition 7:3 (Formula III) was the best formulation. ABSTRAK Telah dilakukan penelitian tentang optimasi sediaan transdermal patch natrium diklofenak tipe matriks. Pada penelitian ini, basis matriks mengandung natrium  diklofenak dibuat dengan kombinasi polimer  etilselulose (EC) N-20  dan polivinilpirolidon (PVP) K-30 dengan berbagai perbandingan yaitu 9:1 (Formula I); 8:2 (Formula II); 7:3 (Formula III)) dan 6:4 (Formula IV). Moisture content (MC) ditentukan dengan cara berat awal dikurangi berat akhir dan dibagi dengan berat akhir. Homogenitas permukaan patch ditentukan dengan menggunakan Scanning Electron Microscope (SEM). Uji pelepasan natrium diklofenak dilakukan dengan alat disolusi dan menggunakan dapar fosfat salin  pH 7,4 ± 0.05 sebanyak 500,0 mL, kecepatan pengadukan 50 rpm dan percobaan dilakukan pada suhu 37 ± 0,5°C. Kadar natrium diklofenak yang terlepas ditentukan dengan metoda spektrofotometer pada panjang gelombang (l) 276 nm. Data yang diperoleh dianalisa secara statistik menggunakan program SPSS dengan derajat kepercayaan 95% (α = 0,05).  Kesimpulan yang didapatkan adalah komposisi polimer EC N-20 dan PVP K-30 dengan perbandingan  7:3 (Formula III) adalah formula yang terbaik.

Abstract

ABSTRACT : This research studied the potential and action mechanism of probiotic milk-guava leaf extract combination as an immunomodulator using mice as an experimental animal. The mouse were given the combination of probiotic milk-guava leaf extract for 7 days, then the levels of TNF-α in serum and lymphocites proliferation were measured. The results showed that the extract of guava leaf can increase levels of TNF-α in serum and the number of lymphoblast, either in single preparation or in combination with probiotic milk. However, administration of probiotic milk cannot improve the TNF-α in serum or increase the number of lymphoblast. It can be concluded that the immunomodulatory effects of the probiotics- milk-guava leaf extract combination was caused by the substances in the extract of guava leaf rather than by the probiotic milk. Jus buah jambu biji telah digunakan secara tradisional oleh masyarakat untuk membantu meningkatkan jumlah trombosit penderita demam berdarah dengue. Kombinasi susu probiotik-ekstrak daun jambu biji telah terbukti mempunyai efek anti bakteri penyebab diare, namun mekanisme kerjanya belum diketahui. Dalam penelitian ini dilakukan kajian potensi dan mekanisme kerja kombinasi susu probiotik-ekstrak daun jambu biji sebagai imunomodulator. Sejumlah mencit diberi perlakuan kombinasi susu probiotik-ekstrak daun jambu biji selama 7 hari, kemudian diukur kadar TNF-α dalam serum dan kecepatan proliferasi limfositnya. Hasil penelitian menunjukkan bahwa ekstrak daun jambu biji dapat meningkatkan kadar TNF-α dalam serum dan dapat meningkatkan jumlah limfoblast, baik dalam bentuk tunggal maupun kombinasinya dengan susu probiotik. Pemberian susu probiotik saja tidak dapat meningkatkan TNF-α dalam serum ataupun meningkatkan jumlah limfoblast. Dari hasil penelitian ini dapat disimpulkan bahwa efek imunomodulator kombinasi susu probiotik-ekstrak daun jambu biji lebih disebabkan oleh kandungan zat zat berkhasiat dalam ekstrak daun jambu biji dari pada oleh susu probiotik. Abstract

Synthesized carbon dots (CDs) from paper flowers (Bougainvillea spectabilis) was performed by microwave assisted. CDs were characterized by ultraviolet visible light (UV-Vis), photoluminescence (PL) and time resolved luminescence (TRPL). PL data shows wavelength intensity of CDs paper flowers at 493 nm for 3.9 ns and 512 nm for 4.2 ns respectively for purple and white paper flowers that excited at 420 nm. The absorbance for CDs white paper flower at 262 nm and 350 nm, while CDs purple flower paper at 260 nm, 324 nm, and 530 nm. CDs from paper flower shows excellent optical properties. So, it is mean that method applied to this experiment effectively, cheap and eco-friendly for synthesis CDs from paper flower.

Abstract

Lycopene is one of the famous ingredients in tomatoes, because of its important role as a potent anti-oxidant. Besides being consumed as fresh fruit, tomato paste can be combined with probiotic fermented milk to increase its health benefits. Probiotics are lactic acid bacteria (LAB), which when consumed in sufficient quantities will benefit health. Probiotic fermented milk is reported to have various properties including anti-microbial, antioxidant, immunomodulatory and anti-cancer, so the combination with tomato paste is expected to increase its benefits for health. This research aimed to study the effect of probiotic fermented milk on the content of tomato paste lycopene. Lactobacillus acidophillus, Lactobacillus bulgaricus, and Lactobacillus casei were used as mixed probiotic cultures. It was found that at the ratio of 2:8 for the LAB fermented milk and tomato paste, the highest lycopene drive was obtained by spectrophotometric measurement at 663, 645, 505, and 453 nm. Determination of the lycopene content was performed according to the Nagata and Yamashita simple methods. In addition, the lycopene content in the LAB fermented milk-tomato paste was greater than the content in the tomato paste without fermented milk addition and was tested stable after storage for 14 days.

Abstract

Clinical use of cefadroxil, particularly in Bangka Tengah Hospital, is proven beneficial to overcome mild to moderate infections which especially occur in soft tissues such as skin, upper respiratory tract, pharyngitis, tonsillitis and urinary tract. For this reason, it is necessary to procure cefadroxil to be available enough for the treatment of cases of these diseases. The cefadroxil used by the Central Bangka Hospital was obtained from several pharmaceutical industries with different prices and distributions, due to the possibility that the active raw materials and ingredients had different origins, so there was concern that the microbiological quality would be different. Drug procurement is carried out using the e-catalog or non-e catalog method. This study aimed to examine the microbiological quality of six preparations (A, B, C, D, E, and F) in terms of their inhibitory activities against Gram-positive and Gram-negative bacteria. The bioassay was carried out by diffusion agar method using Escherichia coli ATCC 29522 and Staphylococcus aureus ATCC 29523 as the bacterial test, and nutrient agar as the test medium. The inhibitory activities were compared to cefadroxil standard for measuring the ratio potency. The results showed that all samples fulfilled USP 41 requirements with potential ratio of 90% to 120% and minimum inhibitory concentration of ≤ 8 ppm and ≤ 2 ppm against Escherichia coli and Staphylococcus aureus respectively. The potency ratios to cefadroxil standard were 95.9%, 99.1%, 100.0%, 96.7%, 96.2% and 98.2% against Staphylococcus aureus while the potency ratios of 95.6%, 99.3%, 103.8%, 97.1%, 95.7% and 100.4% were achieved against Escherichia coli for A, B, C, D, E, and F samples, respectively.

Abstract

The aim of this study was to optimize the use of Ca-Alginates beads as a drug reservoir in the patch system with and without freeze-drying process. Meloxicam, an anti-inflammatory non-steroidal drug, was used as a drug model. Ca-alginate beads were prepared by ionic gelation method using polymer alginate and cross-linker CaCl2. The beads were evaluated for its melting temperature, FTIR spectra, and entrapment efficiency. The FTIR study showed the loss of guluronic peaks of alginate caused by a cross-linking process with Ca2+, characteristics peaks of meloxicam still appeared indicating the compatibility of the drug with the polymers used. The using Ca-alginate beads without freeze-drying process as a drug reservoir resulted in a very wet patch that was difficult to dry, cracked surface, and pharmaceutically not acceptable. Meanwhile, patch using Ca-alginate beads with the freezedrying process as a drug reservoir demonstrated satisfactory characteristics with minimum weight variation among the patches that lead to giving uniformity in the drug content.
